Comparative Biophysical Analysis of Healthy and Inflamed Intestinal Membrane Models Using Langmuir Monolayers.

Intestinal inflammation, including Crohn's disease, represents a chronic condition that increases the risk of colon cancer and involves complications from long-term pharmacotherapy. Since drugs must pass through lipid bilayers to reach their targets, understanding the surface properties of biological membranes is crucial. In this study, the Langmuir technique and Brewster angle microscopy (BAM) were employed to characterize simplified models of healthy and inflamed intestinal cell membranes. Both the outer and inner leaflets of the lipid bilayer were modeled to investigate how inflammation-ind…

A closer look at calcium-induced interactions between phosphatidylserine-(PS) doped liposomes and the structural effects caused by inclusion of gangliosides or polyethylene glycol- (PEG) modified lipids.

The effects of polyethylene glycol- (PEG) modified lipids and gangliosides on the Ca2+ induced interaction between liposomes composed of palmitoyl-oleoyl phosphatidylethanolamine (POPE) and palmitoyl-oleoyl phosphatidylserine (POPS) was investigated at physiological ionic strength. Förster resonance energy transfer (FRET) studies complemented with dynamic light scattering (DLS) and cryo-transmission electron microscopy (Cryo-EM) show that naked liposomes tend to adhere, rupture, and collapse on each other's surfaces upon addition of Ca2+, eventually resulting in the formation of large multilam

Cell-Free Expression of Proton-Coupled Folate Transporter in the Presence of Nanodiscs.

Proton coupled folate transporter (PCFT) is an integral membrane protein with 12 transmembrane segments localized to the plasma membrane. PC … environments in which PCFT is most soluble. Traditional in vitro expression and reconstitution into lipid bilayers of integral membrane proteins requires separate steps, which are costly and time-consuming. In this chapter, we describe a protocol for in vitro translation of PCFT into preformed lipid nanodiscs using a cell-free expression system, which helps to a …

Therapeutic potential for amyloid surface inhibitor: only amyloid-β oligomers formed by secondary nucleation disrupt lipid membrane integrity.

Inhibition of amyloid-β peptide (Aβ) aggregation is a promising therapeutic strategy for Alzheimer's disease (AD), as Aβ aggregation is generally believed to trigger AD pathology. Pre-fibril Aβ-oligomers induce membrane disruption and are crucial to neurotoxicity. We have previously designed a short peptide called cyclic helical amyloid surface inhibitor (cHASI) that can selectively bind to the Aβ fibril surface. Here, we use cHASI to efficiently inhibit the surface-catalysed secondary nucleation process of Aβ in a lipid membrane environment. By incubating Aβ monomers with lipid vesicles, we s

Slow motions in oriented phospholipid bilayers and effects of cholesterol or gramicidin. A 19F-NMR T1 rho study.

In an extension of our earlier work (Peng, Z.-y., V. Simplaceanu, I. J. Lowe, and C. Ho. 1988. Biophys. J. 54:81-95), the rotating-frame nuclear spin-lattice relaxation (T1 rho) technique has been used to investigate the slow molecular motions (10(-4) - 10(-6) s) in lipid bilayers prepared from protonated or perdeuterated 19F-labeled phospholipids in the absence and presence of cholesterol or gramicidin as membrane-interacting molecules. Complications caused by the 19F-1H cross-polarization observed previously can be removed by the substitution of 2H for 1H in the acyl chains. Only a weak depe…

Incorporation of Oligo(Styrene), Oligo(Butadiene), Oligo(Methyl Methacrylate), and Oligo(tert-Butyl Acrylate) Droplets in Giant Unilamellar Vesicles.

In the environment, microplastics fragment and degrade into plastic oligomers, posing potential risks to ecosystems and human health. In the present study, the interactions between giant unilamellar vesicles (GUVs) and liquid droplets of oligo(styrene), oligo(butadiene), oligo(methyl methacrylate), and oligo(tert-butyl acrylate) were investigated using fluorescence microscopy. GUVs were used in a homogeneous fluid state or in a phase-separated state consisting of either a liquid-disordered (Ld) phase and a solid-ordered (So) phase, or a Ld phase and a liquid-ordered (Lo) phase. A scanning calorimetric study of the interaction of anthracyclines with neutral and acidic phospholipids alone and in binary mixtures.

High sensitivity differential scanning calorimetry was employed to study the thermotropic behavior of multilamellar vesicles of neutral and acidic phospholipids and binary mixtures thereof in the presence of anthracycline antibiotics. Adriamycin and its lipophilic analogue, N-trifluoroacetyladriamycin-14-valerate (AD32) were investigated and compared to chlorpromazine and quinidine with respect to their ability to affect the pretransition and the main transition of the phospholipids suspended in physiological buffer. With liposomes of neutral dipalmitoylphosphatidylcholine the observed effects

Nanodiscs for drug delivery.

Lipid nanodiscs have emerged as a versatile and promising tool for drug delivery due to their biocompatibility, structural flexibility, and ability to mimic native cell membrane environments. These nanoscale assemblies, composed of lipid bilayers stabilized by scaffold amphipathic proteins, peptides, synthetic polymers, or saponins, offer a stable membrane mimetic system for encapsulating hydrophobic drugs and membrane proteins. Ongoing research continues to expand the diversity of nanodisc formulations, each with distinct advantages and limitations. Their tunable size, surface functionality, …

Molecular Insights into Osmolyte-Mediated Adaptation of Lipid Membrane of Extremophiles.

Extremophiles survive and thrive in extreme conditions on Earth, which pose challenges to their survivability. Osmotic stress, caused by different extreme conditions, such as water loss due to evaporation in hot and dry conditions, osmotic imbalance due to hypersalinity or excessive pressure, and freeze-drying, causes water loss, which severely affects cell membranes by altering the physicochemical properties of the lipid bilayer. While the organisms employ various adaptive strategies, osmolyte-mediated adaptation (OMA) plays a crucial role in maintaining membrane stability under desiccation.

Structure and morphology of vesicular dispersions based on novel glycophospholipids with various monosaccharide head groups.

Glycophospholipids combine the structural versatility of phospholipids and carbohydrates, but their potential as excipients and performance in other related applications remains largely unexplored due to their low natural abundance. We have synthesized four novel phosphatidyl saccharide conjugates with different carbohydrate head groups; glucose, galactose, fructose and xylose by using a Phospholipase D catalysed transphosphatidylation reaction. The combination of Small Angle X-ray Scattering (SAXS) and Cryogenic Transmission Electron Microscopy (cryo-TEM) data allowed us to characterize the d

Role of isopropyl myristate, isopropyl alcohol and a combination of both in hydrocortisone permeation across the human stratum corneum.

The influence of isopropyl myristate (IPM), isopropyl alcohol (IPA) and a combination of both was studied in view of hydrocortisone (HC) permeation across the human stratum corneum (SC). IPM, IPA and their combination were incorporated into water-containing hydrophilic ointment (WHS), and the resulting effects on HC permeation and on HC accumulation in human SC were investigated as well as the influence of these substances on the microstructure of the SC. Differential scanning calorimetry as well as wide- and small-angle X-ray diffraction show that IPM incorporation into SC results in densely

Effects of Serum Incubation on Lipid Nanoparticle PEG Shedding, mRNA Retention, and Membrane Interactions.

Lipid nanoparticles (LNPs) are widely used for RNA delivery, but their efficiency remains limited, largely due to poor endosomal escape. Upon administration, proteins bind to the surface of the LNPs, influencing cellular uptake and potentially altering their interfacial properties. Such alterations may also affect their interaction with endosomal membranes, thus influencing the critical endosomal escape step. Using fluorescence microscopy imaging with single-LNP resolution, this study investigates how incubation in 10% fetal bovine serum alters the PEG modification and mRNA content of LNPs, as

The Molecular Mechanism of Fluorescence Lifetime of Fluorescent Probes in Cell Membranes.

Fluorescence probes play crucial roles in unraveling the structure and dynamics of cell membranes including membrane fluidity, polarity, and lipid molecule ordering. The fluorescence lifetime of probes describes the average duration of time that a fluorescent molecule remains in an excited state before returning to the ground state, which is sensitive to environmental changes. However, the molecular mechanism and inherent properties to determine the fluorescence lifetimes remain unexplored and inadequately studied. Furthermore, the effects of the probe on the membrane are also unclear. In this

Substitution of bridge carbons for sulphur in calix[4]arene-bis-α-hydroxymethylphosphonic acid transformed mobile carrier into ionic channel accompanied with evoked muscle contraction and impaired neurotransmission powered by membrane action of resulting thiocalix[4]arene-bis-α-hydroxymethylphosphonic acid.

The action of calix[4]arenes C-424, C-425 and C-1193 has been investigated on suspended cholesterol/egg phosphatidylcholine lipid bilayer in a voltage-clamp mode. Comparative analysis with the membrane action by calix[4]arene-bis-α-hydroxymethylphosphonic acid (C-99) has shown that the substitution of bridge carbons for sulphur and addition of another methyl group to two alkyl tales in the lower rim of former dipropoxycalix[4]arene C-99 transformed mobile carrier that C-99 created in lipid bilayer (Shatursky et al., 2014) into a transmembrane pore as exposure of the bilayer membrane to sulphur

Ethephon interactions with biomimetic sperm plasma membranes and their effects on lipid packing and membrane stability.

Ethephon (ETP) is an organophosphorus plant growth regulator extensively used in agriculture, raising concerns about environmental exposure and potential reproductive toxicity. Although animal studies report reduced sperm count, morphological abnormalities, and testicular damage following ETP exposure, the molecular mechanisms underlying these effects remain poorly understood. Because membrane organization and stability are critical determinants of sperm function, we investigated simplified biomimetic sperm plasma membrane systems in the presence of ETP to elucidate membrane-level interaction

Penetration of lipid membranes by gold nanoparticles: insights into cellular uptake, cytotoxicity, and their relationship.

Nanoparticle penetration into cell membranes is an interesting phenomenon that may have crucial implications on the nanoparticles' biomedical applications. In this paper, a coarse-grained model for gold nanoparticles (AuNPs) is developed (verified against experimental data available) to simulate their interactions with model lipid membranes. Simulations reveal that AuNPs with different signs and densities of surface charges spontaneously adhere to the bilayer surface or penetrate into the bilayer interior. The potential of mean force calculations show that the energy gains upon adhesion or pen
