A dual-response near-infrared-emitting fluorescent probe for the evaluation of liver disease-related drugs and multi-level monitoring of lipid droplet abnormalities.

The viscosity and polarity of lipid droplets (LDs) are two crucial physical parameters in their microenvironment. They act as "regulators" of the LDs functions, jointly determining the metabolic activity, stability of LDs, and their roles in cellular stress. Any abnormal change in any one of these indicators can directly affect the metabolic health and fate of cells. Therefore, developing analytical tools that can simultaneously and in real-time monitor the dynamic changes of both of these factors is of crucial significance for revealing the mechanisms of LDs-related metabolic diseases. Based …

Natural Schistosoma mansoni Infection in the Wild Reservoir Nectomys squamipes Leads to Excessive Lipid Droplet Accumulation in Hepatocytes in the Absence of Liver Functional Impairment.

Schistosomiasis is a neglected tropical disease of a significant public health impact. The water rat Nectomys squamipes is one of the most important non-human hosts in the schistosomiasis mansoni transmission in Brazil, being considered a wild reservoir. Cellular mechanisms that contribute to the physiological adaptation of this rodent to the Schistosoma mansoni parasite are poorly understood. Here we identified, for the first time, that a hepatic steatosis, a condition characterized by excessive lipid accumulation with formation of lipid droplets (LDs) within hepatocytes, occurs in response t…

Intermittent Fasting Ameliorates β-Amyloid Deposition and Cognitive Impairment Accompanied by Decreased Lipid Droplet Aggregation Within Microglia in an Alzheimer's Disease Model.

SCOPE: Alzheimer's disease (AD) is the most prevalent form of dementia, lack of effective therapeutic interventions. In this study, we inves … reduced complexity of their processes. Importantly, IF significantly decreased the accumulation of lipid droplets (LDs) within microglia. These microglia with less LDs may contribute to enhanced β-amyloid (Aβ) phagocytosis, thereby ameliorating Aβ deposition in the brains of APP/PS1-IF mice. CONCLUSION: Our findings demonstrate that IF ameliorates amyloid depo …

Prepared Radix Polygoni Multiflori and emodin alleviate lipid droplet accumulation in nonalcoholic fatty liver disease through MAPK signaling pathway inhibition.

As one of the most common liver diseases, nonalcoholic fatty liver disease (NAFLD) affects almost one-quarter of the world's population. Although the prevalence of NAFLD is continuously rising, effective medical treatments are still inadequate. Radix Polygoni Multiflori (RPM) is a traditional Chinese herbal medicine. As a processed product of RPM, prepared Radix Polygoni Multiflori (PRPM) has been reported to have antioxidant and anti-inflammatory effects. This study investigated whether PRPM treatment could significantly improve NAFLD. We used recent literature, the Herb database and the Swis

Exercise ameliorates lipid droplet metabolism disorder by the PLIN2-LIPA axis-mediated lipophagy in mouse model of non-alcoholic fatty liver disease.

Excessive hepatic lipid droplets (LDs) accumulation-induced lipid metabolism disorder contributes to the development of non-alcoholic fatty liver disease (NAFLD). Exercise is a promising therapeutic strategy for NAFLD. However, the mechanism by which exercise ameliorates NAFLD through regulating the catabolism of hepatic LDs remains unclear. In the present study, we investigated the effect of perilipin2 (PLIN2)-lysosomal acid lipase (LIPA) axis mediating exercise-triggered lipophagy in a high-fat diet (HFD)-induced NAFLD mouse model. Our results showed that exercise could reduce HFD-induced he…

The Combined Inhibition of Autophagy and Diacylglycerol Acyltransferase-Mediated Lipid Droplet Biogenesis Induces Cancer Cell Death during Acute Amino Acid Starvation.

Lipid droplets (LDs) are dynamic organelles involved in the management of fatty acid trafficking and metabolism. Recent studies suggest that autophagy and LDs serve complementary roles in the protection against nutrient stress, but the autophagy-LD interplay in cancer cells is not well understood. Here, we examined the relationship between autophagy and LDs in starving HeLa cervical cancer- and MDA-MB-231 breast cancer cells. We found that acute amino acid depletion induces autophagy and promotes diacylglycerol acyltransferase 1 (DGAT1)-mediated LD accumulation in HeLa cells. Inhibition of aut…

Adipose Triglyceride Lipase Is a Key Lipase for the Mobilization of Lipid Droplets in Human β-Cells and Critical for the Maintenance of Syntaxin 1a Levels in β-Cells.

Lipid droplets (LDs) are frequently increased when excessive lipid accumulation leads to cellular dysfunction. Distinct from mouse β-cells, LDs are prominent in human β-cells. However, the regulation of LD mobilization (lipolysis) in human β-cells remains unclear. We found that glucose increases lipolysis in nondiabetic human islets but not in islets in patients with type 2 diabetes (T2D), indicating dysregulation of lipolysis in T2D islets. Silencing adipose triglyceride lipase (ATGL) in human pseudoislets with shRNA targeting ATGL (shATGL) increased triglycerides (TGs) and the number and siz…

Critical roles for α/β hydrolase domain 5 (ABHD5)/comparative gene identification-58 (CGI-58) at the lipid droplet interface and beyond.

Mutations in the gene encoding comparative gene identification 58 (CGI-58), also known as α β hydrolase domain-containing 5 (ABHD5), cause neutral lipid storage disorder with ichthyosis (NLSDI). This inborn error in metabolism is characterized by ectopic accumulation of triacylglycerols (TAG) within cytoplasmic lipid droplets in multiple cell types. Studies over the past decade have clearly demonstrated that CGI-58 is a potent regulator of TAG hydrolysis in the disease-relevant cell types. However, despite the reproducible genetic link between CGI-58 mutations and TAG storage, the molecular me…

Toward anti-bluing shift: A rationally designed photo-induced electron transfer-based polarity responsive lipid droplets-targeting fluorescent probe for real-time monitoring of drug-induced liver injury.

Drug-induced liver injury (DILI) is a significant public health issue due to its unpredictability and potential severity. DILI primarily manifests as hepatic steatosis or phospholipidosis, accompanied with an elevated lipid droplets (LDs) number. Therefore, developing powerful polarity-responsive probes that target LDs is critical for elucidating the precise function of LDs in DILI. Common solvatochromic polarity-sensitive probes often show a significant blue shift in their emission wavelength as the polarity decreases, with some probes exhibiting a shift of over 100 nm, leading to low accurac…

Shikonin Attenuates Neuroinflammation and Depression-Like Behaviors by Regulating Microglial Polarization Through Inhibiting cGAS-STING/NF-κB Signaling and Lipid Droplet Accumulation.

BACKGROUND: Major depressive disorder is a disabling psychiatric illness with a growing global burden. Neuroinflammation, particularly microglial polarization, is increasingly recognized as a key pathogenic factor and is closely linked to lipid metabolic regulation. Shikonin, a bioactive naphthoquinone from Lithospermum erythrorhizon, exhibits anti-inflammatory and antioxidant properties, yet its antidepressant effects remain unclear. This study examined whether shikonin alleviates depressive-like behaviors by suppressing neuroinflammation. METHODS: Mice were orally treated with shikonin for

Bempedoic acid attenuates glial lipid droplet accumulation and inflammatory responses and alleviates chronic social defeat stress-induced depressive-like behaviors in mice.

Depression is a common and disabling psychiatric disorder, and chronic psychosocial stress is an important risk factor for stress-related depressive phenotypes. Neuroinflammation and glial changes have been implicated in chronic stress, but whether lipid droplets accumulate in specific brain cell types after stress remains unclear. Bempedoic acid (BPA) is an orally available ATP-citrate lyase inhibitor used for lipid lowering and has anti-inflammatory effects in preclinical studies. Here, male C57BL/6J mice were exposed to 10 days of chronic social defeat stress (CSDS) and treated orally with …

PLIN2-PGAM5-regulated lipid droplet-mitochondria contacts drive microglial neuroinflammation after spinal cord injury via fatty acid metabolic reprogramming.

Persistent neuroinflammation is a hallmark of central nervous system (CNS) injury, driving neuronal loss and functional deficits. While micr … s interaction inverts canonical metabolic flux by redirecting fatty acids from mitochondria back to lipid droplets, thereby suppressing β-oxidation and inducing mitochondrial dysfunction, which subsequently triggers STING-dependent inflammation. Notably, the PLIN2 220-392 domain is essential for PGAM5 binding and contact formation; disrupting this interaction at …

A Phosphoproteomic Platform Identifies Erythrocyte Membrane Protein Band 4.1-Like 3-Mediated Lipid Droplet Remodeling Linked to Liver Cancer Invasion and Migration.

Aberrant lipid metabolism is a hallmark of hepatocellular carcinoma (HCC), yet the regulatory mechanisms governing lipid droplet (LD) dynamics and their contribution to tumor progression remain poorly understood. Here, we developed an ultrasensitive phosphoproteomic platform using high-affinity HPDA@Ti4+ nanospheres to map LD-associated phosphorylation events across six HCC cell lines. By correlating phosphoproteomic signatures with LD morphology, we identified distinct regulatory signatures associated with LD size and abundance. Functional perturbation screens identified two distinct phosphop

Astrocytic SCAP deletion ameliorates diabetes-associated cognitive impairment by suppressing microglial neuroinflammation and lipid droplet accumulation via the LCN2-24p3R-mTOR axis.

Diabetes-associated cognitive impairment (DACI) is a prevalent and debilitating complication of type 2 diabetes, yet the mechanisms linking metabolic dysregulation to neuroinflammation and cognitive decline remain incompletely understood. In this study, we identify astrocytic SCAP (SREBP cleavage-activating protein) as a crucial regulator of glial metabolic-inflammatory crosstalk in DACI. Using a high-fat diet-induced mouse model of diabetes, we demonstrate that astrocytic SCAP expression is significantly upregulated, whereas astrocyte-specific SCAP deletion alleviates cognitive deficits, redu

Ceramide synthase 6 (CerS6) is upregulated in alcohol-associated liver disease and exhibits sex-based differences in the regulation of energy homeostasis and lipid droplet accumulation.

OBJECTIVE: Alcohol-associated liver disease (ALD) is the leading cause of liver-related mortality worldwide. Current strategies to manage ALD focus largely on advanced stage disease, however, metabolic changes such as glucose intolerance are apparent at the earliest stage of alcoholic steatosis and increase the risk of disease progression. Ceramides impair insulin signaling and accumulate in ALD, and metabolic pathways involving ceramide synthase 6 (CerS6) are perturbed in ALD during hepatic steatosis. In this study, we aimed to investigate the role of CerS6 in ALD development and the relevanc

Photoactivable AIEgen-based Lipid-Droplet-Specific Drug Delivery Model for Live Cell Imaging and Two-Photon Light-Triggered Anticancer Drug Delivery.

Lipid droplets (LDs) are dynamic complex organelles involved in various physiological processes, and their number and activity are linked to multiple diseases, including cancer. In this study, we have developed LD-specific near-infrared (NIR) light-responsive nano-drug delivery systems (DDSs) based on chalcone derivatives for cancer treatment. The reported nano-DDSs localized inside the cancer microenvironment of LDs, and upon exposure to light, they delivered the anticancer drug valproic acid in a spatiotemporally controlled manner. The developed systems, namely, 2'-hydroxyacetophenone-dimeth…

Mystery of bisphenol F-induced nonalcoholic fatty liver disease-like changes: Roles of Drp1-mediated abnormal mitochondrial fission in lipid droplet deposition.

As one of the major substitutes for bisphenol A (BPA), bisphenol F (BPF) has been widely used. Our previous study demonstrated that BPF exposure facilitates lipid droplet deposition in hepatic cells, contributing to nonalcoholic fatty liver disease (NAFLD)-like changes. However, the underlying mechanisms remain poorly understood. Here, with a metabolic cage system, we observed the perturbation of energy metabolism in mice treated with BPF. BPF obviously suppressed metabolic capacity, which manifested as decreased energy expenditure, low O2 consumption and CO2 levels in mice. Consistent with th

Acidosis-Induced TGF-β2 Production Promotes Lipid Droplet Formation in Dendritic Cells and Alters Their Potential to Support Anti-Mesothelioma T Cell Response.

For poorly immunogenic tumors such as mesothelioma there is an imperious need to understand why antigen-presenting cells such as dendritic cells (DCs) are not prone to supporting the anticancer T cell response. The tumor microenvironment (TME) is thought to be a major contributor to this DC dysfunction. We have reported that the acidic TME component promotes lipid droplet (LD) formation together with epithelial-to-mesenchymal transition in cancer cells through autocrine transforming growth factor-β2 (TGF-β2) signaling. Since TGF-β is also a master regulator of immune tolerance, we have here ex

Metabolic stress remodels the Golgi-sphingolipid-lipid droplet axis in INS-1E β-cells: an integrated molecular signature of early β-cell vulnerability.

BACKGROUND: Glucotoxicity, lipotoxicity, and glucolipotoxicity contribute to pancreatic β-cell dysfunction in type 2 diabetes. Although endoplasmic reticulum stress has been widely studied, the response of the Golgi apparatus to metabolic stress and its relationship with sphingolipid metabolism, lipid droplets, and insulin granule-associated pathways remain poorly understood. We examined these processes together in INS-1E β-cells exposed to different forms of metabolic stress. METHODS AND RESULTS: INS-1E cells were exposed to high glucose (33 mM), palmitic acid (0.5 mM), or both for 24 h. Cel…
