The interplay between sleep and cognition in adults with Epilepsy: evidence from a systematic review.

Sleep disturbances and cognitive impairment are common among people with epilepsy (PWE). The relationship between sleep and epilepsy has bee … een at least one sleep parameter and cognitive function. Four studies investigating sleep-dependent memory consolidation demonstrated a retained benefit of sleep on memory in PWE. Four studies found that sleep microstructure played an independent role. Another finding shared by six studies was the negative effect of nocturnal interictal epileptic discharges (IEDs) on …

Conditions under which lorazepam can facilitate retrieval.

Memory is composed of three stages: acquisition, consolidation, and retrieval. By impairing acquisition processes, benzodiazepines cause anterograde amnesia while leaving intact information learned before the drug was taken. In some circumstances, retrieval of this information is even improved by benzodiazepines. It has been hypothesized that this phenomenon is not a true facilitation of retrieval processes, but is the result of reduced interference from items presented after drug administration and is thus a secondary consequence of drug-induced amnesia. Experiment 1 investigated the effect o

Scaphoid waist fractures fixation with staple. Retrospective study of a not widespread procedure.

INTRODUCTION: In carpal scaphoid fractures, the surgical treatment with screw is considered the gold standard; shape memory staple however presents substantial advantages. The authors report a study on unstable fractures of the scaphoid waist (type B1, B2, B5, according to Herbert classification) treated with shape memory staple on a large sample of patients, with the aim to confirm the usefulness of this method, the quality of reduction and fixation, the functional results, the time of union and the possible complications. MATERIALS AND METHODS: A retrospective analysis of 131 patients with

Hippocampal lesions produce both nongraded and temporally graded retrograde amnesia in the same rat.

Rats were administered contextual fear conditioning and trained on a water-maze, spatial memory task 28 days or 24 h before undergoing hippocampal lesion or control surgery. When tested postoperatively on both tasks, rats with hippocampal lesions exhibited retrograde amnesia for spatial memory at both delays but temporally graded retrograde amnesia for the contextual fear response. In demonstrating both types of retrograde amnesia in the same animals, the results parallel similar observations in human amnesics with hippocampal damage and provide compelling evidence that the nature of the task

Dentate gyrus network regulation by somatostatin- and parvalbumin-expressing interneurons differentially impacts spatial memory processing.

GABAergic interneurons regulate circuit dynamics of memory-processing brain structures. However, the role of the dentate gyrus (DG) and its predominant interneuron subtypes - somatostatin-expressing (SST+) and parvalbumin-expressing (PV+) - in different stages of memory processing is unknown. We tested how the chemogenetic manipulation of DG SST+ and PV+ interneurons in mice influences encoding, consolidation, and retrieval of hippocampus-dependent object-location memory (OLM). The activation of DG SST+ interneurons impairs OLM encoding and retrieval, dramatically suppresses DG granule cell ac

Roles of NMDA NR2B subtype receptor in prefrontal long-term potentiation and contextual fear memory.

Cortical plasticity is thought to be important for the establishment, consolidation, and retrieval of permanent memory. Hippocampal long-term potentiation (LTP), a cellular mechanism of learning and memory, requires the activation of glutamate N-methyl-D-aspartate (NMDA) receptors. In particular, it has been suggested that NR2A-containing NMDA receptors are involved in LTP induction, whereas NR2B-containing receptors are involved in LTD induction in the hippocampus. However, LTP in the prefrontal cortex is less well characterized than in the hippocampus. Here we report that the activation of t

Ventral tegmental area glutamatergic neurons suppress hippocampal ripples and induce head movements in mice.

Ripples are brief, high-frequency hippocampal oscillations crucial for memory consolidation, occurring predominantly during non-rapid-eye-movement (NREM) sleep. While ripples are generated intrinsically within the hippocampus, growing evidence suggests that their occurrence is modulated by subcortical regions. Whether the ventral tegmental area (VTA)-a subcortical region that directly projects to the hippocampus-regulates ripple activity remains unknown. To investigate this, we optogenetically activated VTA neurons during NREM sleep and recorded ripples in the dorsal hippocampus via field reco…

Hippocampal interictal discharges induce frontal spindles and enhance spindle-slow oscillation coupling in temporal lobe epilepsy.

OBJECTIVE: To investigate how hippocampal interictal epileptiform discharges (IEDs) influence cortical spindles, slow oscillations (SOs), and spindle-SO coupling, which are key components of sleep-dependent memory consolidation. METHODS: Simultaneous intracranial and scalp EEG was recorded during NREM sleep in 10 patients with temporal lobe epilepsy (TLE). Hippocampal IEDs were identified using depth electrodes, while spindles and SOs were detected from ipsilateral scalp EEG in frontal, central, parietal, and temporal regions. Peri-IED time histograms and spectral analyses were performed to e…

HDAC3-selective inhibitor enhances extinction of cocaine-seeking behavior in a persistent manner.

Nonspecific histone deacetylase (HDAC) inhibition has been shown to facilitate the extinction of drug-seeking behavior in a manner resistant to reinstatement. A key open question is which specific HDAC is involved in the extinction of drug-seeking behavior. Using the selective HDAC3 inhibitor RGFP966, we investigated the role of HDAC3 in extinction and found that systemic treatment with RGFP966 facilitates extinction in mice in a manner resistant to reinstatement. We also investigated whether the facilitated extinction is related to the enhancement of extinction consolidation during extinction

Investigating the transition from recent to remote memory using advanced tools.

Remote memories, weeks to decades long, are usually the ones most important to the organism, as the longevity of a memory is tightly connected to its significance. Retrograde amnesia studies in human patients as well as lesions and immediate early gene expression investigation in animal models, suggested that the hippocampus has a time dependent role in memory consolidation. Namely, that as a memory matures it becomes independent of the hippocampus and instead depends on extra-hippocampal areas. However, accumulating evidence implies that this temporal segregation is not as rigid as originally…

Approach memory turns to avoidance memory with age.

Ontogenetic modification of an early memory is relatively poorly understood. And an important question is whether the memory output is more determined by the age at acquisition or at retention? Here we explore the expression of odor-shock conditioning in the rat pup. Acquisition at post-natal day 6 (P6) leads to an approach response and at post-natal day 12 (P12) to an avoidance response when the retention test is 24h later. In both cases, anisomycin injected immediately post-acquisition induced a retrograde amnesia. Controls show that, in either case, short-term memory measured 4h after acqui

The forgotten insular cortex: its role on recognition memory formation.

For a long time, the insular cortex (IC) has been related with taste physiology and taste memory processes in animal studies. Recently, the role of the IC has been highlighted by findings involving the IC in non-taste memory formation in both human and animal studies. Recognition memory is based on the ability to assess the familiarity of a previously encountered stimulus, and it is considered a form of declarative memory. In this work, I am proposing that the IC and its related circuitry are highly involved in the conversion of novel to familiar stimulus for both object and taste recognition

Transcranial direct current stimulation enhances offline consolidation of implicit sequential learning in individuals with insomnia.

BACKGROUND: Insomnia not only compromises sleep quality but may also impair learning and memory processes within cognitive function. OBJECTIVE: This study aims to investigate the effects of transcranial direct current stimulation (tDCS) on offline consolidation of implicit sequence learning and sleep quality in individuals with insomnia. METHODS: We employed a randomized, double-blind, controlled trial, assigning individuals with insomnia to either an active stimulation group or a sham stimulation group for consecutive 7-day interventions. Sleep quality (PSQI) and implicit sequence learning

Learned Motor Patterns Are Replayed in Human Motor Cortex during Sleep.

Consolidation of memory is believed to involve offline replay of neural activity. While amply demonstrated in rodents, evidence for replay in humans, particularly regarding motor memory, is less compelling. To determine whether replay occurs after motor learning, we sought to record from motor cortex during a novel motor task and subsequent overnight sleep. A 36-year-old man with tetraplegia secondary to cervical spinal cord injury enrolled in the ongoing BrainGate brain-computer interface pilot clinical trial had two 96-channel intracortical microelectrode arrays placed chronically into left

Amygdala's involvement in facilitating associative learning-induced plasticity: a promiscuous role for the amygdala in memory acquisition.

It is widely accepted that the amygdala plays a critical role in acquisition and consolidation of fear-related memories. Some of the more widely employed behavioral paradigms that have assisted in solidifying the amygdala's role in fear-related memories are associative learning paradigms. With most associative learning tasks, a neutral conditioned stimulus (CS) is paired with a salient unconditioned stimulus (US) that elicits an unconditioned response (UR). After multiple CS-US pairings, the subject learns that the CS predicts the onset or delivery of the US, and thus elicits a learned conditi
