Using the pharmacology of latent inhibition to model domains of pathology in schizophrenia and their treatment.

Latent inhibition (LI) is a cross species selective attention phenomenon reflecting a modulating effect of past experience with inconsequential stimuli on current task performance involving these stimuli. Specifically, it refers to the observation that organisms that receive nonreinforced preexposure to the to-be conditioned stimulus are less efficient in generating a conditioned response to that stimulus when it is subsequently paired with reinforcement. Because dysfunction of attentional selectivity is a fundamental deficit associated with schizophrenia, disrupted LI is long considered to mo

Response inhibition and impulsivity in schizophrenia.

Response inhibition in schizophrenia remains controversial, with behavioral correlates largely unknown. Inpatients with schizophrenia and controls completed a stop task and an impulsiveness questionnaire. Slower inhibitory processes were evident in schizophrenia, but there was no association with impulsivity. The nature of inhibition and impulsivity in schizophrenia is complex, and could reflect schizophrenia subgroups or disease states.

Strabismic suppression is mediated by inhibitory interactions in the primary visual cortex.

Most strabismic observers do not suffer from double vision because of suppression from conscious perception of 1 of the 2 eyes' conflicting views. Direct evidence for the site and neural substrate of strabismic suppression has not been available so far, although psychophysical data suggest a cortical origin. On the other hand, cross-orientation suppression among conflicting stimuli presented monocularly has recently been shown to have a strong thalamic component. Here we present evidence, using both visual stimulation and pharmacological techniques, that strabismic suppression occurs in the pr

Neuroimmune interaction in the gut: from bench to bedside.

The enteric nervous system plays a crucial role in the control of gastrointestinal motility and coordinates relaxation and contraction of the smooth muscle cells. Relaxation of the gut is mainly mediated by inhibitory non-adrenergic non-cholinergic (NANC) neurons, a subpopulation of enteric neurons which release a neurotransmitter different from acetylcholine and noradrenaline. Via in vitro work performed in Antwerp, we demonstrated in 1990 that these neurons release nitric oxide. Thereafter, my research has focused on the role of the inhibitory NANC neurons in the pathophysiology of postopera

Transcranial Magnetic Stimulation in the investigation and treatment of schizophrenia: a review.

Transcranial Magnetic Stimulation (TMS) is a non-invasive method of stimulating the brain that is increasingly being used in neuropsychiatric research and clinical psychiatry. This review examines the role of TMS in schizophrenia research as a diagnostic and a therapeutic resource. After a brief overview of TMS, we describe the application of TMS to schizophrenia in studies of cortical excitability and inhibition, and we discuss the potential confounding role of neuroleptic medications. Based on these studies, it appears that some impairment of cortical inhibition may be present in schizophren

Directionality derived from pinna-cue spectral notches in cat dorsal cochlear nucleus.

We tested two hypotheses to determine whether dorsal cochlear nucleus (DCN) neurons are specialized to derive directionality from spectral notches: DCN neurons exhibit greater spectral-dependent directionality than ventral cochlear nucleus (VCN) neurons, and spectral-dependent directionality depends on response minima (nulls) produced by coincidence of best frequency (BF) and spectral-notch center frequency. Single-unit responses to 50-ms noise and tone bursts were recorded in barbiturate-anesthetized cats (BFs: 4-37 kHz). Units were classified using BF tone poststimulus time histograms. Pause

Neuronal activity in rat red nucleus during forelimb reach-to-grasp movements.

The activity of 259 task-related red nucleus neurons was recorded using chronic electrophysiological methods in free moving rats. Modulations in activity were analysed in relation to onset (first detected wrist movement) and end (arrival of the paw over the food) of a reach-to-grasp movement. Excitatory peaks were found to begin before, during and after the reach, but there were clear peaks in the distribution of onset times after reach-onset and before reach-end, reflecting the fact that one third of all peaks began specifically during the reach, although this occupied only a small fraction o

Plasticity in an electrosensory system. II. Postsynaptic events associated with a dynamic sensory filter.

1. This report summarizes studies of the changes in postsynaptic potentials that occur as pyramidal cells within the primary electrosensory processing nucleus learn to reject repetitive patterns of afferent input. The rejection mechanism employs "negative image inputs" that oppose or cancel electroreceptor afferent inputs or patterns of pyramidal hyperpolarization or depolarization caused by intracellular current injection. Feedback pathways carrying descending electrosensory as well as other types of information provide the negative image inputs. This study focuses on the role of a directly d

Prevalence of bowel dysfunction in patients with multiple sclerosis and bladder dysfunction.

Urinary dysfunction is common in cases of multiple sclerosis (MS). The close proximity of those neural pathways which control the bladder to those which control anorectal function might be expected to lead to a high coexistence of bladder and bowel symptoms. Seventy-seven consecutive patients with clinically definite MS attending a uroneurology clinic were interviewed about their bowel function. All patients had clinical evidence of spinal cord disease with varying degrees of impaired mobility and sufficiently severe disturbance of bladder control to seek medical advice. Thirty-six per cent of

Interocular control of neuronal responsiveness in cat visual cortex.

Neurons in the cat primary visual cortex are selective for particular contour orientations but their responsiveness can vary under certain conditions. After prolonged stimulation (adaptation), the contrast sensitivity of cortical cells is reduced and the 'gain' (the strength of response as a function of contrast) falls. The response to an optimal contour is also reduced when a different stimulus is superimposed on the receptive field in the same eye. Here we report that the sudden appearance of an inappropriate stimulus in one eye can interocularly suppress the activity of cortical neurons if

[Effect of the beta-carboline derivative FG 7142 on inhibition in hippocampal sections].

The action of anxiogenetic derivative of beta-carboline FG7142 on evoked activity of neurons in hippocampal sections was investigated using extra- and intracellular recordings. The activity in CAI area was registered upon stimulation of Schaffer's collaterals (SC). Excitatory effect of FG7142 (5 microM) on population spike (PS) was blocked by simultaneous diazepam (5 microM) or RO15-1788 (5 microM) application. This suggests that FG7142 action is mediated by benzodiazepine receptors. To evaluate the action of FG7142 on hippocampal inhibitory processes paired-pulse stimulation of SC was used. I

Comparative pharmacological effects on visual cortical neurons in monocularly deprived cats.

Monocularly deprived (MD) cats show a loss of responsiveness to visual stimulation of the deprived eye among visual cortical neurons. Several lines of evidence suggest that this effect involves, at least in part, a suppression of deprived eye input, possibly mediated by GABA inhibition. In order to better understand the nature of this suppression we have evaluated the effectiveness of different types of disinhibitory and excitatory agents to reverse the effects of MD. We investigated bicuculline (a GABA antagonist); picrotoxin (a GABA antagonist with a different mechanism of action from bicucu

An intracellular analysis of dendrodendritic inhibition in the turtle in vitro olfactory bulb.

1. Intracellular recordings from an in vitro preparation of turtle olfactory bulb were used to determine the pathway responsible for producing synaptic inhibition of mitral cells.2. Inhibitory post-synaptic potentials (i.p.s.p.s) could be elicited in mitral cells by both olfactory nerve (orthodromic) and mitral cell axon (antidromic) stimulation and by suprathreshold depolarizing current pulses injected intracellularly through the recording micro-electrode. Reversing the chloride gradient by either intracellular injection of chloride or lowering the external chloride concentration reversed the

Inhibition of cutaneous nociception by deep musculoskeletal pain. A clinical observation.

A patient is reported in whom deep musculoskeletal pain apparently blocked transmission from nociceptive cutaneous fibers in an adjacent region. When the deep musculoskeletal pain was abolished with local anesthesia, the cutaneous hypalgesia disappeared. Naloxone did not influence the hypalgesia. Possible mechanisms are discussed.

The effect of activation of carotid body chemoreceptors on baroreceptor inhibition of sympathetic activity.

The effect of combined stimulation of peripheral chemo- and baroreceptors upon sympathetic discharge was studied in cats anesthetized with chloralose and urethan, immobilized with tricuran, and artificially ventilated. The right carotid sinus was arterially isolated, the left sinus nerve and both vago-sympathetic trunks were cut. A rise of pressure in isolated carotid sinus from 100 to 200 mm Hg produced an inhibition of sympathetic activity recorded simultaneously from the inferior cardiac and the renal nerves. For a combined chemo- and baroreceptor stimulation, the carotid sinus was perfused

Carotid sinus nerve efferents: properties and physiological significance.

The significance of a sympathetic efferent nerve supply, vasoconstrictor to the carotid body and hence facilitatory to carotid chemoreceptors, is well understood. The significance of a second efferent pathway, whose impulses, passing down the sinus nerve, are inhibitory to the chemoreceptors, is less certain. Activity in these sinus nerve efferents is increased by injection of pressor agents, by hypoxia, by hypercapnia, by application of alkaline solutions to the ventral medulla, and by severe hypocapnia. All these stimuli appear to act centrally. Sinus nerve efferents can also be activated re

The action of botulinum A neurotoxin on the inhibition by antidromic stimulation of the lumbar monosynaptic reflex.

1. Botulinum toxin type A was injected into gastrocnemius muscles of cats and 2 days later its action on the monosynaptic gastrocnemius reflex and on the recurrent inhibition was investigated. 2. In early local botulism, no significant change in the monosynaptic gastrocnemius reflex was observed. 3. The inhibition by antidromic stimulation of the monosynaptic gastrocnemius reflex was significantly reduced. 4. During the early phases of local botulism an action of botulinum toxin on parts of the soma membrane of the alpha-motoneuron is suggested.

The inhibitory influence of the pyloric antrum on gastric secretion in dogs.

The effect of the pyloric antrum and of subsequent vagotomy on the secretory responses of an innervated or a denervated gastric pouch in the dog to histamine and pentagastrin was determined. The increased sensitivity of the gastric pouch to stimulation after these procedures is in keeping with the concept that an inhibitory reflex is concerned with the control of acid gastric secretion.
