A review of the effects of nicotine on schizophrenia and antipsychotic medications.

OBJECTIVE: Research on the impact of nicotine on schizophrenia and antipsychotic medications was reviewed to determine ways to improve treatment planning for patients with schizophrenia who smoke and to evaluate smoking cessation programs for this population. METHODS: All major research databases were searched. The review focuses on reports published since 1990. RESULTS: Smoking improves processing of auditory stimuli (sensory gating) by patients with schizophrenia and may lessen negative symptoms by increasing dopamine in the nucleus accumbens and the prefrontal and frontal cortex. Use of t…

Norfloxacin nicotinate pharmacokinetics in unweaned and weaned calves.

The pharmacokinetics of norfloxacin nicotinate were investigated in unweaned and weaned calves. Following intravenous administration of 7.5 and 15 mg/kg (calculated as norfloxacin base) the clearance values were 8.5 +/- 2.0 or 7.7 +/- 1.2 mL/min.kg (unweaned calves) and 11.7 +/- 3.2 or 16.1 +/- 3.3 mL/min.kg (weaned calves). Norfloxacin mean residence time and volume of distribution values were 211 +/- 33 or 227 +/- 41 min (unweaned calves) and 185 +/- 79 or 128 +/- 18 minutes (weaned calves), and 1.8 +/- 0.3 or 1.7 +/- 0.1 L/kg (unweaned calves) and 2.0 +/- 0.7 or 2.1 +/- 0.7 L/kg (weaned cal

Agmatine acts as an antagonist of neuronal nicotinic receptors.

1. Tritiated agmatine has been used by others in ion flux methods to measure nicotinic receptor function in neurones. However, as shown here, agmatine blocks nicotinic receptor function in both the chick retina and the rat superior cervical ganglion at high concentrations. 2. In intact chick retina, agmatine 1 mM decreases dimethylphenylpiperazinium (DMPP)-induced depolarizations measured in the optic nerve by approximately 70%, while having little effect on responses induced by glutamate analogues. DMPP dose-response curves are reduced in a manner consistent with a non-competitive effect of a

The nicotinic acid test in the evaluation of unconjugated hyperbilirubinemia.

In 42 young adults with Gilbert's syndrome and in 9 patients with inactive, well-compensated liver cirrhosis the peroral nicotinic acid test was performed. Of the 13 parameters tested, the groups differed significantly in the sum of total and unconjugated serum bilirubin concentrations, respectively, during the test; in the difference between the maximal and initial concentrations of total and unconjugated bilirubin; in the retention of total bilirubin at the third, fourth and fifth hour of the test; and in the size of the area under the curve of unconjugated bilirubin increase. However, due t

Pharmacokinetics of nicotinic acid-salicylic acid interaction.

Both nicotinic acid and salicylic acid undergo glycine conjugation in human beings. Competitive inhibition may therefore be possible when these substances are used concomitantly in patients with hyperlipidemic disorders. The aim of this study was to determine, in six healthy subjects, whether nicotinic acid steady-state levels and total clearance are affected by concomitant aspirin administration. Steady-state nicotinic acid concentrations were obtained in all six volunteers by infusion of nicotinic acid solutions at constant rates (0.075 to 0.100 mg/kg/min) for 6 hours; aspirin (1 gm) was adm

The interaction of nicotinic receptor antagonists at muscarinic receptors.

1. The interaction of some neuromuscular blocking drugs such as gallamine, pancuronium and stercuronium with muscarinic receptors has several features which distinguish these compounds from competitive antagonists in functional and binding studies. 2. They also differentiate between muscarinic receptors and may produce effects on ion channels.

Noninvasive assessment of local nicotinate pharmacodynamics by photoplethysmography.

The local pharmacodynamics of a topical vasodilator (methyl nicotinate) has been followed noninvasively using photopulse plethysmography. This technique is sensitive to changes in blood flow through the cutaneous microcirculation and responds to the pharmacologic stimulus of the vasoactive agent employed. Five different application sites for the drug were studied and the time course of the local effect (i.e., onset, duration, and decay) was recorded. The applied amount of drug elicited, within a short period, a response which was saturable such that the observed increase in blood flow reached

Nicotinic acid action on gastrointestinal smooth muscle.

The mechanism of nicotinic acid action in smooth muscle was studied by testing the drug in rat stomach fundus and descending colon using either the superfusion or perfusion technique. In the rat stomach strips nicotinic acid (or Na-nicotinate) induces concentration-dependent contractions that are drastically antagonized by indomethacin but not by the other inhibitors tested (scopolamine, phenoxybenzamine, propranolol, methysergide, pyrilamine, imidazole, anacardic acid). Reduced glutathione, on the contrary, clearly potentiates the contractile effect of nicotinic acid in the tissue. The respon

Nicotinic nociceptors on perivascular sensory nerve endings.

Intra-arterial injection of increasing doses of acetylcholine, nicotine, carbachol and pilocarpine into the pain reflex ear of the rabbit dose-dependently caused a reflex fall in mean arterial blood pressure. The order of potency of the algesic substances was: nicotine greater than acetylcholine much greater than carbachol much greater than pilocarpine, whereas the purely muscarinic agonist bethanechol was inactive up to 3 mg. Atropine at 1 microgram/ml did not influence the effect of acetylcholine. At 30 micrograms/ml it reduced the effect of acetylcholine but also that of bradykinin suggesti…

Studies of muscarinic and nicotinic binding sites in brain.

Binding properties for the muscarinic antagonist quinuclidinyl benzilate (QNB), the nicotinic antagonist tubocurarine and nicotine to crude synaptosomal fractions from mouse and rat brain have been studied. Marked regional differences in number of binding sites for labelled QNB (3H-QNB) were found with a high concentration in the striatum, cortex and a low concentration in the cerebellum. The specific binding sites for labelled tubocurarine (3H-tubocurarine) showed less regional variation. An increased number of muscarinic binding sites (with no change in receptor affinity) was found in rats f…

Studies on nicotinic acid interaction with bilirubin metabolism.

The mechanism by which intravenous administration of nicotinic acid (NA) increases serum unconjugated bilirubin in patients with the Gilbert's syndrome has been investigated. Studies using the technique of percutaneous transhepatic catheterization of the splenic vein and coil planet centrifuge suggested that following intravenous injection of NA some of the circulating erythrocytes were rendered osmotically fragile and trapped by the spleen and that unconjugated bilirubin increased in the splenic vein blood. In patients with liver cirrhosis, the increments of unconjugated bilirubin were closel

Primitive earth synthesis of nicotinic acid derivatives.

Nicotinonitrile, 2-cyanopyridine, and 4-cyanopyridine can be synthesized under primitive earth conditions by the action of electric discharges on ethylene and ammonia. The electric discharge first synthesizes pyridine and hydrogen cyanide, which react in the discharge to form the cyanopyridines. Nicotinonitrile would have hydrolyzed in the primitive ocean to nicotinamide and nicotinic acid.

[The effects of nicotine pouch usage on the mucous membrane].

This case report explores the oral effects of nicotine pouch use, focusing on two patients with mucosal changes. Findings include gingival inflammation, white lesions, and gingival retraction. Evidence suggests a link between nicotine pouches and periodontal damage. Though considered less harmful than smoking, their discreet use may lead to intense usage. Clinicians should remain vigilant for early signs of harm and inform patients about potential risks. Further research is needed to clarify long-term health effects.

The Effects of THC and Nicotine on Attention: A Narrative Review.

Since cannabis and nicotine are two of most commonly used substances and are often used together, this paper will review the effects of cannabis (specifically THC, or tetrahydrocannabinol) and nicotine on selective attention, sustained attention, visuospatial attention, attentional bias, and attentional disorders. This review includes preclinical and clinical findings throughout all periods of development and adulthood. Selective attention is directly impacted by cannabis use, while reaction time is dependent on the timing of the last cannabis exposure. Among individuals who use cannabis, ther…

Signaling of nicotinic acetylcholine receptors in mononuclear phagocytes.

Nicotinic acetylcholine receptors are not only expressed by the nervous system and at the neuro-muscular junction but also by mononuclear phagocytes, which belong to the innate immune system. Mononuclear phagocyte is an umbrella term for monocytes, macrophages, and dendritic cells. These cells play pivotal roles in host defense against infection but also in numerous often debilitating diseases that are characterized by exuberant inflammation. Nicotinic acetylcholine receptors of the neuronal type dominate in these cells, and their stimulation is mainly associated with anti-inflammatory effects

Nicotinic acetylcholine receptors in neurological and psychiatric diseases.

Neuronal nicotinic acetylcholine receptors (nAChRs) are ligand-gated ion channels that are widely distributed both pre- and post-synaptically in the mammalian brain. By modulating cation flux across cell membranes, neuronal nAChRs regulate neuronal excitability and the release of a variety of neurotransmitters to influence multiple physiologic and behavioral processes including synaptic plasticity, motor function, attention, learning and memory. Abnormalities of neuronal nAChRs have been implicated in the pathophysiology of neurologic disorders including Alzheimer's disease, Parkinson's diseas
