BACKGROUND & AIMS: We recently showed that the functional capacity for ureagenesis is deficient in non-alcoholic fatty liver disease (NAFLD) patients. The aim of this study was to assess expression of urea cycle-related genes to elucidate a possible gene regulatory basis to the functional problem.
METHODS: Liver mRNA expression analyses within the gene pathway governing hepatic nitrogen conversion were performed in 20 non-diabetic, biopsy-proven NAFLD patients (8 simple steatosis; 12 non-alcoholic steatohepatitis [NASH]) and 12 obese and 14 lean healthy individuals. Sixteen NAFLD patients wer

UNLABELLED: Flex fuel vehicles (FFVs) typically operate on gasoline or E85, an 85%/15% volume blend of ethanol and gasoline. Differences in FFV fuel use and tailpipe emission rates are quantified for E85 versus gasoline based on real-world measurements of five FFVs with a portable emissions measurement system (PEMS), supplemented chassis dynamometer data, and estimates from the Motor Vehicle Emission Simulator (MOVES) model. Because of inter-vehicle variability, an individual FFV may have higher nitrogen oxide (NOx) or carbon monoxide (CO) emission rates on E85 versus gasoline, even though ave

The dual challenges of excessive carbon dioxide emissions and the accumulation of agricultural biomass waste pose significant environmental concerns. A sustainable solution lies in repurposing biomass-derived materials for CO2 mitigation. Biomass wastes are one of the primary sources of carbonaceous materials, and with a rational synthetic strategy, these substrates can be turned into a cost-effective solution for CO2 capture. In this work, we develop a nitrogen-doped activated carbon-based sorbent from peanut shells, optimizing its porosity and basicity for selective CO2 adsorption. Peanut sh

Sustainable conversion of NO offers a promising technology for artificial nitrogen cycle, but faces challenges in integrating electrocatalysis with NO capture. Herein, a biomimetic NO capture-activation-conversion strategy was firstly formulated by encapsulating N-acetylcysteine (NAC) within a nicotinamide adenine dinucleotide (NADH) mimic-containing coordination capsule, enabling efficiently NH3 electrosynthesis. The installed NADH mimics onto the capsule interacted with the highly reactive NO-adduct generated from NAC within the microenvironment akin to the pocket of enzyme, facilitating int…

The availability of ambient N nutrient is often correlated with the occurrences of harmful algal bloom formed by certain dinoflagellates, making it important to understand how these species might be responding to such conditions. Here, transcriptome sequencing of Karenia mikimotoi was conducted to understand the underlying molecular mechanisms by which this dinoflagellate copes with nitrogen (N) deficiency. Transcriptomic analysis revealed 8802 unigenes (3.56%) that were differentially expressed with ≥ 2-fold change. Under N-depleted conditions, genes involved in glycolysis, fatty acid metabol

The indoline scaffold, a privileged class of nitrogen-containing heterocycles, is ubiquitous in bioactive natural products and pharmaceuticals; consequently, the development of efficient synthetic strategies remains a forefront of organic synthesis. Herein, we report a visible-light-induced Markovnikov-selective sulfonylation/cyclization cascade, enabling the mild and efficient construction of 3-sulfonyl indolines from o-aminoarylalkynes and sodium sulfinates. Mechanistic studies reveal that the transformation proceeds via a radical-mediated pathway, initiated through either an electron donor-

Migratory functionalization of C-H bonds through metal migration from carbon to carbon under transition metal catalysis is a process of significant academic and industrial interest. Herein, a palladium-catalyzed migratory cyclization of α-bromoalkene derivatives ArXCBr=CH2, in which X denotes a phosphorus (P(O)R), silicon (SiR2), sulfur (SO2), carbon (C(O)), nitrogen (NTs), or oxygen-based moiety, affording various benzoheterocyclic compounds has been developed. Mechanistic investigations have demonstrated that the cyclization reaction proceeds through an unexpected cascade, with trans-1,2-pal

We investigate the dynamics of particulate matter, nitrogen oxides, and ozone concentrations in Hong Kong. Using fluctuation functions as a measure for their variability, we develop several simple data models and test their predictive power. We discuss two relevant dynamical properties, namely, the scaling of fluctuations, which is associated with long memory, and the deviations from the Gaussian distribution. While the scaling of fluctuations can be shown to be an artifact of a relatively regular seasonal cycle, the process does not follow a normal distribution even when corrected for correla

Isoprene is the dominant non-methane organic compound emitted to the atmosphere1-3. It drives ozone and aerosol production, modulates atmospheric oxidation and interacts with the global nitrogen cycle4-8. Isoprene emissions are highly uncertain1,9, as is the nonlinear chemistry coupling isoprene and the hydroxyl radical, OH-its primary sink10-13. Here we present global isoprene measurements taken from space using the Cross-track Infrared Sounder. Together with observations of formaldehyde, an isoprene oxidation product, these measurements provide constraints on isoprene emissions and atmospher…

UNLABELLED: Mosquito larvae rely on a living microbiota for normal development because the microbiota supplies essential nutrients, particularly vitamins. Beyond vitamin provision, transcriptomic data suggest that the microbiota also supports other key nutritional processes. Here, we explored these roles by conducting a metabolomics analysis on Aedes aegypti third instar larvae following microbiota depletion. We sampled larvae and dissected guts 12 and 20 h post-decolonization, and analysed methanol-soluble metabolites using untargeted gas chromatography-mass spectrometry. Our findings reveal

The agriculture sector contributes 22 % of global greenhouse gas emissions, with fertilizers accounting for 10.6 % of that portion. To reduce this, biofertilizers can be employed due to their lower emissions throughout production and application. Past studies have quantified either the upstream or downstream emissions of biofertilizers, yet the direct and indirect emissions from a life cycle perspective remain unclear. Additionally, most studies did not consider local conditions such as soil organic carbon and soil nitrogen content, leading to inaccuracies in the calculated greenhouse gas (GHG

UNLABELLED: In contrast to their important structural and regulatory functions, such as in the metabolism of cyanobacteria, genes encoding small proteins are often not well characterized. Cyanobacteria use redox equivalents and energy from oxygenic photosynthesis to produce organic carbon compounds from inorganic carbon (C i ) and organic nitrogen compounds from inorganic nitrogen sources. Therefore, the assimilation and metabolism of carbon and nitrogen are coordinated at multiple levels in cyanobacteria. Here, we analyzed the Synechocystis sp. PCC 6803 gene ssr3189 encoding a 55 amino acids

BACKGROUND: Viral warts represent a large workload for dermatology departments. Cryotherapy with liquid nitrogen is the most widely used method of treatment by dermatologists and is increasingly used by general practitioners in the U.K. Existing data relating to the response to cryotherapy are virtually all derived from the use of a cotton wool bud as the applicator. There is an increasing trend to use the cryo-spray to freeze warts and it has been assumed that this is equally effective. In view of the workload involved it is important to test this assumption.

The key challenges in the application of biochar in slow-release fertilizers due to the defects of poor pores and few functional groups. In this study, a chitosan-modified biochar-based infiltration nitrogen fertilizer (ZBC@CS-BUF) was synthesized using zinc chloride-chitosan co-modified biochar (ZBC@CS) as a fertilizer carrier with molten urea. The results showed that ZnCl2 promoted the formation of micropores in biochar (SSA is 306.04 m2/g), and the introduction of chitosan increased the abundance of functional groups containing nitrogen (-NH2) and oxygen (-CO, -OH). The fitting results of t

The processes of denitrification and dissimilatory NO3- reduction to ammonium (DNRA) occupy a competing pivotal point in the nitrogen cycle. Denitrification leads to a loss of fixed nitrogen, while DNRA recycles NO3- as bioavailable ammonium. Iron (II) is known to enhance DNRA, by acting as an electron donor, however, the organisms responsible for iron driven DNRA remain poorly understood. Here we undertook incubations of sediment with NO3- and Fe2+ additions followed by process measurements and metagenomic analysis to measure DNRA and microbial community structure. Addition of Fe2+ stimulated…

The fate of excess nitrogen in estuaries is determined by the microbial-driven nitrogen cycle, being denitrification a key process since it definitely removes fixed nitrogen as N2. However, estuaries receive and retain metals, which may negatively affect this process efficiency. In this study, we evaluated the role of salt marsh plants in mediating cadmium (Cd) impact on microbial denitrification process. Juncus maritimus and Phragmites australis from an estuary were collected together with the sediment involving their roots, each placed in vessels and maintained in a greenhouse, exposed to na…

Tumor lysis syndrome (TLS) is a fatal complication associated with chemotherapy. We herein report a case of TLS in a 73-year-old woman with metastatic BRAFV600E mutated colon cancer after she received combined treatment with cetuximab and encorafenib. The serum uric acid, urea nitrogen, and creatinine levels were elevated on day four of the first cycle. The fibrin degradation product (FDP) and D-dimer levels were also high. Diuresis and rasburicase were initiated for TLS, and the laboratory data all normalized on day 8. Thus, the possibility of TLS being induced by targeted drugs in patients w

Posttranslational acetylation of proteins by acetyl-CoA is a crucial regulator of proteostasis and substrate metabolism. Ethanol metabolism in the liver induces protein accumulation, acetylation, and metabolic disruption. Although acetylation impacts enzyme activity and stability, its role in ethanol-related protein accumulation and metabolic dysfunction remains unclear. Using stable isotope-based proteomics, acetylomics, and metabolic profiling in a mouse model of chronic ethanol-induced liver injury, we demonstrate that ethanol induces hepatic steatosis, inflammation, oxidative stress, and p

Urea cycle disorder (UCD) is a group of inherited metabolic diseases with high disability or fatality rate, which need long-term drug treatment and diet management. Except those with Citrin deficiency or liver transplantation, all pediatric patients require lifelong low protein diet with safe levels of protein intake and adequate energy and lipids supply for their corresponding age; supplementing essential amino acids and protein-free milk are also needed if necessary. The drugs for long-term use include nitrogen scavengers (sodium benzoate, sodium phenylbutyrate, glycerol phenylbutyrate), ure

Urea cycle disorder (UCD) is an inborn error of the metabolic pathway producing urea from ammonia, which occurs primarily in the liver. Decreased excretion of nitrogen in the urea cycle due to deficiency of carbamoyl phosphate synthase I (CPSI), ornithine transcarbamylase (OTC), argininosuccinate synthase (ASS), argininosuccinate lyase (ASL), and N-acetyl glutamate synthase (NAGS) causes hyperammonemia. We examined the clinical manifestations, treatment, and prognosis of 177 patients with UCD from January 1999 to March 2009 in Japan. Compared with a previous study conducted in Japan, a larger
