Numerical Comparison of Nasal Aerosol Administration Systems for Efficient Nose-to-Brain Drug Delivery.

PURPOSE: Nose-to-brain drug administration along the olfactory and trigeminal nerve pathways offers an alternative route for the treatment of central nervous system (CNS) disorders. The characterization of particle deposition remains difficult to achieve in experiments. Alternative numerical approach is applied to identify suitable aerosol particle size with maximized inhaled doses. METHODS: This study numerically compared the drug delivery efficiency in a realistic human nasal cavity between two aerosol drug administration systems targeting the olfactory region: the aerosol mask system and t

The alteration of odor-induced c-Fos immunoreactivity in the rat olfactory bulb after olfactory nerve transection.

We used the rats in which one olfactory nerve had been transected and observed the odor (Propionic acid) -induced c-Fos immunoreactivity in the bulb at different times (2, 4, 8 weeks) after nerve transection. The exposure to odor produced a strong cluster of c-Fos positive cells in the mediodorsal region of the intact bulb. On the other hand, the transected bulb showed much less reactivity 2 weeks after neurectomy; however, a large number of positive cells were observed in the whole of the bulb from 4 weeks after neurectomy. Furthermore, we measured the levels of mRNA for tyrosine hydroxylase

The olfactory route for cerebrospinal fluid drainage into the peripheral lymphatic system.

Drainage of the cerebrospinal fluid through the olfactory nerves into the nasal lymphatics has been suggested repeatedly. To investigate precisely the morphology of this pathway, India ink was injected into the subarachnoidal space of the rat brain, and samples including the olfactory bulbs, olfactory tracts and the nasal mucosa were observed by light and electron microscopy. Under the dissecting microscope, ink particles were found within the subarachnoid space and along the olfactory nerves. At the nasal mucosa, a lymphatic network stained in black was identified near the olfactory nerves, w

Directed expression of the growth-associated protein B-50/GAP-43 to olfactory neurons in transgenic mice results in changes in axon morphology and extraglomerular fiber growth.

B-50/GAP-43, a neural growth-associated phosphoprotein, is thought to play a role in neuronal plasticity and nerve fiber formation since it is expressed at high levels in developing and regenerating neurons and in growth cones. Using a construct containing the coding sequence of B-50/GAP-43 under the control of regulatory elements of the olfactory marker protein (OMP) gene, transgenic mice were generated to study the effect of directed expression of B-50/GAP-43 in a class of neurons that does not normally express B-50/GAP-43, namely, mature OMP-positive olfactory neurons. Olfactory neurons hav

Central projections of the nervus terminalis in four species of amphibians.

The central projections of the nervus terminalis were investigated in two anuran and two urodele species by means of horseradish peroxidase injections into one nasal cavity. In anurans, the nervus terminalis projects to the medial septum, to the preoptic nucleus, to the nucleus of the anterior commissure and to the hypothalamus. In addition to these structures, the dorsal thalamus, the infundibulum and the mesencephalic tegmentum are innervated in urodeles. The structure containing the highest density of terminals in the amphibians investigated is the hypothalamus. In one anuran and one urodel

Immunocytochemical localization of luteinizing hormone-releasing hormone within the olfactory bulb of pigs.

LHRH was immunocytochemically localized within the olfactory bulb of prepubertal (n = 3), ovariectomized (n = 3), and hypophyseal-stalk-transected (HST) female pigs (n = 3). Perikarya of LHRH-immunoreactive neurons of all pigs were sparsely distributed mostly in the rostral half of the olfactory bulb, along the ventromedial and ventrolateral edge of the olfactory nerve layer, or at its interace with the glomerular layer. Processes from these cells and other LHRH containing axons either entered individual glomeruli forming a network within its interior or coursed around glomeruli penetrating in

Connections of the olfactory bulb in the gymnotiform fish, Apteronotus leptorhynchus.

This work examines the connectivity of the olfactory bulb in the gynmotiform fish Apteronotus leptorhynchus. Wheat germ agglutinin conjugated horseradish peroxidase was iontophoresed in different areas and depths of the bulb in order to define its efferent and afferent connections. The olfactory bulb projects bilaterally via the medial (medial and centromedial fascicles) and lateral olfactory (lateral and centrolateral fascicles) tracts. The nervus terminalis courses through the ventromedial aspect of the bulb to terminate in parts of the medial subpallium and hypothalamus. Its telencephalic c

Sphingosine 1-phosphate promotes the proliferation of olfactory ensheathing cells through YAP signaling and participates in the formation of olfactory nerve layer.

Olfactory ensheathing cells (OECs) are unique glial cells with axonal growth-promoting properties in the olfactory epithelium and olfactory bulb, covering the entire length of the olfactory nerve. The proliferation of OECs is necessary for the formation of the presumptive olfactory nerve layer (ONL) during development and OECs transplantation. However, the molecular mechanism underlying the regulation of OEC proliferation in the ONL still remains unknown. In the present study, we examined the role of sphingosine 1-phosphate (S1P) and S1P receptors (S1PRs) on OEC proliferation. Initially, rever

Loss of NEIL1 causes defects in olfactory function in mice.

Oxidative DNA damage accumulation has been implicated in neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. The base excision repair pathway is a primary responder to oxidative DNA damage. Effects of loss of base excision repair on normal brain function is a relatively nascent area of research that needs further exploration for better understanding of related brain diseases. Recently, we found that loss of a versatile DNA glycosylase endonuclease 8-like 1 (NEIL1) causes deficits in spatial memory retention using the Morris water maze test. Furthermore, we found tha

Evidence of SARS-CoV2 Entry Protein ACE2 in the Human Nose and Olfactory Bulb.

Usually, pandemic COVID-19 disease, caused by SARS-CoV2, presents with mild respiratory symptoms such as fever, cough, but frequently also with anosmia and neurological symptoms. Virus-cell fusion is mediated by angiotensin-converting enzyme 2 (ACE2) and transmembrane serine protease 2 (TMPRSS2) with their organ expression pattern determining viral tropism. Clinical presentation suggests rapid viral dissemination to the central nervous system leading frequently to severe symptoms including viral meningitis. Here, we provide a comprehensive expression landscape of ACE2 and TMPRSS2 proteins acro

Glial fibrillary acidic protein (GFAP)-like immunoreactivity in normal and transected rat olfactory nerve.

Normal and transected rat olfactory nerves were stained immunohistochemically using a monoclonal antibody previously shown to selectively detect GFAP-like immunoreactivity in central astrocytes but not in peripheral Schwann cells. Low levels of "central" type GFAP were found in the olfactory nerves, presumably in ensheathing cells. The levels of GFAP increased dramatically after nerve transection. A population of strongly GFAP-positive cells was detected at the junction between the olfactory epithelium and initial part of the nerves, of possible relevance to the regenerative abilities of this

Incidentally Found Olfactory Schwannoma: A Rare Case Report.

BACKGROUND: Olfactory nerve schwannomas are sporadic, benign neurogenic tumors arising from Schwann cells within the olfactory pathway. Their clinical presentation is typically nonspecific, and diagnosis often requires careful radiologic and histopathologic evaluation. CASE PRESENTATION: We describe a 19-year-old male patient with an olfactory schwannoma that was incidentally discovered during evaluation for head trauma. The patient was asymptomatic, with preserved olfactory function. MANAGEMENT AND RESULTS: The patient underwent complete surgical excision via bifrontal craniotomy. Immunohis

Studies of development and plasticity in the olfactory sensory neuron.

The olfactory system is favorable for studying mechanisms of development, plasticity and regeneration. Monoclonal antibodies have been generated which differentially stain olfactory axons and can identify their earliest trajectories in the fetal rat. The developing olfactory pathway also shows differential metabolic activity, as revealed by the 2-deoxyglucose method, and these patterns show plasticity as judged by both physiological and behavioral measures. The sensory neurons undergo dieback and neurogenesis following axonal transection; electrophysiological methods are being used to reveal t

Cadmium-induced olfactory dysfunction in rainbow trout: Effects of binary and quaternary metal mixtures.

A functioning olfactory response is essential for fish to be able to undertake essential behaviors. The majority of work investigating the effects of metals on the olfactory response of fish has focused on single-metal exposures. In this study we exposed rainbow trout to cadmium, copper, nickel, zinc, or a mixture of these four metals at or below the current Canadian Council of Ministers of the Environment guidelines for the protection of aquatic life. Measurement of olfactory acuity using an electro-olfactogram demonstrated that cadmium causes significant impairment of the entire olfactory sy

Tyrosine hydroxylase expression in rat olfactory bulb transplants: an electron microscope study.

Juxtaglomerular (JG) neurons of rat olfactory bulb (OB) have been shown to express tyrosine hydroxylase (TH), the rate-limiting enzyme in the catecholamine synthesis pathway. These JG neurons act as inhibitory dopaminergic interneurons, modulating the incoming signal from the primary olfactory afferents. The JG neurons, comprised of periglomerular, external tufted, and short axon cells, stop expressing TH after lesions of the olfactory nerve or closure of the nares, both of which cause a loss of functional input. Upon reinnervation by a continuously regenerating olfactory nerve, these cells re

Glucose-6-phosphate dehydrogenase activity in the olfactory system of the young rat: an enzyme histochemical study using computerized image analysis.

An understanding of olfactory system glucose metabolism is necessary for the interpretation of radiolabeled 2-deoxyglucose studies of odor processing since the relationship between glucose uptake and neural activity is based on assumptions regarding cellular glucose utilization. As part of an ongoing study examining divergent pathways of glucose metabolism in the olfactory system, the relative activity of glucose-6-phosphate dehydrogenase, the rate-limiting enzyme of the hexose monophosphate shunt, was examined among cells of the rat olfactory bulb and anterior olfactory nucleus, by using enzy

Intranasal VACV VR-1354 infection impairs chemosensory function and induces olfactory bulb neuroinflammation in mice.

Vaccinia virus (VACV) and monkeypox virus (MPXV) are closely related members of the family Poxviridae, genus Orthopoxvirus, both capable of causing systemic infections with potential neurological complications. Although live, replication-competent VACV strains were historically used in smallpox vaccination, their clinical use was associated with rare but severe central nervous system (CNS)-related adverse events. Despite this, the mechanisms underlying VACV-induced CNS pathology, particularly olfactory dysfunction, remain poorly characterized. In this study, we found VACV-VR1354, a tissue cult

Mapping of uterine-related neurons in central nervous system of mice by trans-synaptic tracing with pseudorabies virus.

The uterus, a key organ for maintaining reproductive health and fertility, is precisely regulated by central nervous system (CNS). This study employed multimodal neural tracing and c-Fos staining to investigate the multisynaptic pathways connecting uterus and CNS in mice, as well as the potential neural mechanisms through which olfactory stimuli influence uterine activity. First, the PRV-CAG-EGFP was injected into uterine myometrium. PRV-infected neurons were observed to ascend over time from spinal cord and medulla oblongata to forebrain, ultimately appearing in regulatory nuclei such as para

Locusts adopt IP3 as a second messenger for olfactory signal transduction.

Insects, unlike vertebrates, use heteromeric complexes of odorant receptors and co-receptors for olfactory signal transduction. However, the secondary messengers involved in this process are largely unknown. Here, we use the olfactory signal transduction of the aggregation pheromone 4-vinylanisole (4VA) as a model to address this question. When locusts detect 4VA, the pheromone is transported by OBP10 and OBP13 to the OR35-Orco receptor complex, thereby activating downstream pathways in the antenna. A pivotal downstream molecule, the lipid-binding protein Clvs2, facilitates phosphatidylinosito
