Development of Liquid-Liquid Extraction-Assisted, Derivatization-Free Gas Chromatography Integrated With the Flame Ionization Detection (GC-FID) Method for the Direct Determination of Fatty Acids in Cream Formulations.

Marketed cream formulations contain fatty acids as core constituents, required for its texture, uniformity, shelf life, and skin penetration. Given their importance in the cosmetic and pharmaceutical industries, real-time quantification and analysis of these compounds are crucial for assessing authenticity and ensuring compliance with regulatory specifications. In the present analysis, we employed gas chromatography with flame ionization detection (GC-FID) due to its sensitivity, robustness, and quantitative reliability. Overcoming the traditional method for fatty acid determination, we analyz

Terpenoid and Fatty Acid Profiling along with Anti-Tyrosinase and Anti-Urease Potentials of Scorzonera pygmaea Sibth. & Sm. an Endemic Plant of Turkey.

Petroleum ether (PE), chloroform (CH), ethyl acetate (EA) and butanol (BU) fractions were obtained from ethanol extracts of the roots (SPR) and the above ground (SPH) parts of S. pygmaea. PE fractions were evaluated by GC/MS for profiling the terpenoids and the fatty acids of the plant. 29 compounds in total including saturated (major palmitic acid), unsaturated (major linoleic acid) fatty acids and triterpenes were tentatively determined. Short-term extraction methods might be preferred to avoid esterification of fatty acids. Phytochemical profiles of SPR-PE and SPH-PE were found to be simila

Lipid binding to gastric mucin: protective effect against oxygen radicals.

Gastric mucus forms a viscous gel overlying the gastric mucosa and is thought to protect the underlying mucosa from noxious agents such as acid, proteases, and bile salts. A common property of mucin, the principal glycoprotein in mucous secretions, is its ability to bind lipids. The purpose of this study was to determine if lipids bound to gastric mucin protect the mucin from oxygen radical attack. Pig gastric mucin, partially purified by Sepharose 4B gel chromatography, was found to contain large amounts of free fatty acids and cholesterol as well as lesser amounts of sphingomyelin and phosph

High milk lipase activity associated with breast milk jaundice.

Human milk samples that inhibit bilirubin-UDP-glucuronyl transferase (UDPGT) activity in vitro have been associated with prolonged unconjugated hyperbilirubinemia in newborn infants. We measured the concentration of nonesterified fatty acids (total and individual fatty acids), total fat and protein, and lipase activities (with and without bile salt stimulation) in milk samples from two groups of women. Women whose infants had prolonged unconjugated hyperbilirubinemia and whose milk inhibited the activity of UDPGT were in the first group (N = 9). Volunteers with healthy infants acted as control

Diet-related toxemia in pregnancy. I. Fat, fatty acids, and cholesterol.

Toxemia in pregnancy (preeclampsia)is characteristerized by a combination of at least two of the following clinical symptoms: hypertension, edema, and proteinuria. In three successive trials over three consecutive years, the dietary intake of a selected number of young pregnant women attending a Maternal and Infant Care Program at Tuskegee Institute were evaluated for total lipids, individual fatty acids, and cholesterol. Women with toxemia or with any of the individual symptoms were identified and women without toxemia or these symptoms served as controls. Results were variable from repetitio

Cerulenin-induced changes in the lipopolysaccharide content and phospholipid composition of Proteus mirabilis.

Inhibition of Proteus mirabilis growth by cerulenin, a specific inhibitor of fatty acid biosynthesis, was reversed by exogenously supplied fatty acid mixtures containing oleic acid and palmitic or pentadecanoic acids. The growth rate of the cells treated with cerulenin in the presence of the fatty acid mixtures was slower, however, than that of untreated cells, and their lipopolysaccharide content was decreased by 30-50%, resulting in an increased sensitivity of the organisms to rifamycin and vancomycin. Polyacrylamide gel electrophoresis of the lipopolysaccharide fraction from cerulenin-treat

A component-resolved in vitro skin barrier model for assessing nanoplastic retention and barrier susceptibility.

Human-relevant in vitro models are needed to assess dermal nanoplastic hazards, yet current skin penetration approaches often treat the stratum corneum as a compositionally uniform barrier. This limits mechanistic understanding of how barrier biochemistry relates to nanoplastic retention, particularly in skin states with altered lipid or protein organization. Here, we developed a component-resolved in vitro stratum corneum model to quantify interactions between 50 nm polystyrene nanoplastics and six major skin barrier constituents: ceramide, cholesterol, keratin, palmitic acid, proline, and ph

Luteolin ameliorates palmitate-induced lipotoxicity in hepatocytes by mediating endoplasmic reticulum stress and autophagy.

Abnormal accumulation of lipids in liver leads to uncontrolled endoplasmic reticulum (ER) stress and autophagy. Luteolin is known to have antioxidant, anti-inflammatory, and anti-cancer properties, but whether it protects against lipotoxicity in liver remains unclear. In this study, we challenged AML12 liver cells and mouse primary hepatocytes with palmitic acid (PA) with or without luteolin pretreatment. In the presence of PA, reactive oxygen species (ROS) production was increased at 3 h, followed by enhancement of expression of p-PERK, ATF4, p-eIF2α, CHOP, and TXNIP (ER stress markers) and p

Activity and mRNA levels of enzymes involved in hepatic fatty acid synthesis and oxidation in mice fed conjugated linoleic acid.

The effects of dietary conjugated linoleic acid (CLA) on the activity and mRNA levels of hepatic enzymes involved in fatty acid synthesis and oxidation were examined in mice. In the first experiment, male ICR and C57BL/6J mice were fed diets containing either a 1.5% fatty acid preparation rich in CLA or a preparation rich in linoleic acid. In the second experiment, male ICR mice were fed diets containing either 1.5% linoleic acid, palmitic acid or the CLA preparation. After 21 days, CLA relative to linoleic acid greatly decreased white adipose tissue mass but caused hepatomegaly accompanying a

Effects of substrate length and active-site residue on catalytic function of fatty acid photodecarboxylase.

Fatty acid photodecarboxylases (FAPs) utilize blue light to convert fatty acids into Cn-1 hydrocarbons and CO2, providing a sustainable route to photobiocatalytic fuel production. Despite increasing interest, substrate-dependent phenomena-such as binding affinity, initial electron-transfer (ET) dynamics, and the influence of residues near the FAD cofactor-remain insufficiently characterized. Here, we systematically analyzed substrate binding, substrate-to-product conversion, and initial ET dynamics for three fatty acids of varying chain lengths using two cosolvent buffer systems. Dietary intake of palmitate and oleate has broad impact on systemic and tissue lipid profiles in humans.

BACKGROUND: Epidemiologic evidence has suggested that diets with a high ratio of palmitic acid (PA) to oleic acid (OA) increase risk of cardiovascular disease (CVD). OBJECTIVE: To gain additional insights into the relative effect of dietary fatty acids and their metabolism on CVD risk, we sought to identify a metabolomic signature that tracks with diet-induced changes in blood lipid concentrations and whole-body fat oxidation. DESIGN: We applied comprehensive metabolomic profiling tools to biological specimens collected from 18 healthy adults enrolled in a crossover trial that compared a 3-w

Non-alcoholic fatty liver disease promotes liver metastasis of colorectal cancer via fatty acid synthase dependent EGFR palmitoylation.

Liver metastasis is the major reason for most of colorectal cancer (CRC) related deaths. Accumulating evidence indicates that CRC patients with non-alcoholic fatty liver disease (NAFLD) are at a greater risk of developing liver metastasis. With the growing prevalence of NAFLD, a better understanding of the molecular mechanism in NAFLD-driven CRC liver metastasis is needed. In this study, we demonstrated that NAFLD facilitated CRC liver metastasis as a metabolic disorder and promoted the stemness of metastatic CRC cells for their colonization and outgrowth in hepatic niches. Metabolically, the

Determination of microbial diversities and aroma characteristics of Beitang shrimp paste.

Beitang shrimp paste (BSP) is fermented by different parts of shrimp, such as the head (H), meat (M), or the whole shrimp (S and W). Microbial communities of BSP were dominated by Firmicutes and Proteobacteria at the phyla level and Tetragenococcus at the genus level. However, the microbial diversity of M was the lowest than the others. Non-dominant bacterial communities were presented by a mutual symbiotic model in BSP fermentation. Tetragenococcus, Halanaerobium, Streptococcus, and Brevundimonas were positively correlated with the biosynthesis of amino acids, fatty acids, and metabolic cofac
