An indirect enzyme-linked immunosorbent assay (ELISA) was developed to measure humoral antibody responses of chickens against Pasteurella multocida. A standard indirect hemagglutination (IHA) test was used to compare serologic results with those of ELISA. The ELISA was also used following challenge with P. multocida to compare the efficacy of three commercial fowl cholera vaccination regimens. Although antibody titers measured by ELISA and IHA were highly correlated, ELISA was at least twice as sensitive as IHA. Antibody measured by ELISA and IHA also correlated significantly with protection a…

Multidrug-resistant (MDR) bovine respiratory pathogens can significantly decrease treatment success and have an effect on dairy farm productivity. Although antibiograms have emerged as a tool to support empirical decision making for disease treatment, the prevalence of MDR isolates on a farm can have a negative effect on the selection of the best antibiotic treatment using antibiogram data. This study aimed to characterize the MDR profile of Pasteurella multocida and Mannheimia haemolytica isolates from the deep nasopharyngeal swabs of calves, heifers, and cows diagnosed with bovine respirator…

Background/Objectives: Pasteurella multocida is responsible for a wide variety of animal diseases worldwide, causing major economic losses. These infections are usually treated with antibiotics; however, the emergence of multidrug-resistant (MDR) strains is increasingly hindering. Understanding antibiotic resistance in P. multocida is important for effective treatment strategies and public health, as it impacts both animal and human welfare. Methods: In this study, the antibiotic susceptibility of 80 P. multocida isolates was evaluated by phenotypic (disk diffusion and broth microdilution) and…

Historically, the use of antibiotics was not well regulated in veterinary medicine. The emergence of antibiotic resistance (ABR) in pathogen … the sensitive range, while 11 isolates were in the resistant range. Seventeen isolates (58.62%) of Pasteurella multocida had MIC values in the susceptible range and 41.38% (12 isolates) had an intermediate MIC value. Mutant prevention concentration determinations as done in this study is effective for the antibiotic treatment of bacterial infections and minimising the …

Bovine respiratory disease (BRD) causes high morbidity and mortality in beef cattle worldwide. Antimicrobial resistance (AMR) monitoring of BRD pathogens is critical to promote appropriate antimicrobial stewardship in veterinary medicine for optimal treatment and control. Here, the susceptibility of Mannheimia haemolytica and Pasteurella multicoda isolates obtained from BRD clinical cases (deep lung swabs at post-mortem) among feedlots in four Australian states (2014-2019) was determined for 19 antimicrobial agents. The M. haemolytica isolates were pan-susceptible to all tested agents apart fr

Pasteurella multocida is a Gram-negative organism characterized morphologically as coccobacillus. It is the commonest organism infecting pet bites. However, severe infections may occur in the absence of animal bites or scratches. Pasteurella multocida serves as an opportunistic pathogen in humans, especially in patients with depressed immune system. Few cases in the literature identify Pasteurella multocida as the causative agent of septic shock, especially in cirrhotic patients. We report a patient who presented with septic shock as a result of Pasteurella multocida bacteremia, without prior …

Pasteurella multocida toxin (PMT) is a virulence factor responsible for the pathogenesis of some Pasteurellosis. PMT exerts its toxic effects through the activation of heterotrimeric GTPase (G(q), G(12/13) and G(i))-dependent pathways, by deamidating a glutamine residue in the α subunit of these GTPases. However, the enzymatic characteristics of PMT are yet to be analyzed in detail because the deamidation has only been observed in cell-based assays. In the present study, we developed rat monoclonal antibodies, specifically recognizing the deamidated Gα(q), to detect the actions of PMT by immun…

Pasteurella (P) multocida exists in a variety of animals and causes diverse infections in humans due to animal bites and scratches, usually by cats or dogs, and oral and respiratory infection. We report a case of P multocida sepsis due to a scratch from a pet cat, complicated with disseminated intravascular coagulation in a post-chemotherapy neutropenic patient with non-Hodgkin lymphoma. The patient was a febrile 79-year-old woman with disturbed consciousness and subcutaneous abscess in her right hand due to a scratch from a pet cat. She was successfully treated with empirical antibiotic thera

A Pasteurella multocida species-specific oligonucleotide probe, pmhyb449, targeting 16S rRNA was designed and evaluated by whole-cell hybridization against 22 selected reference strains in animal tissues. It differentiated P. multocida from other bacterial species of the families Pasteurellaceae and Enterobacteriaceae and also from divergent species of the order Cytophagales (except biovar 2 strains of Pasteurella avium and Pasteurella canis, which have high 16S rRNA similarity to P. multocida). The potential of the probe for specific identification and differentiation of P. multocida was furt…

Immunization of mice with a potassium thiocyanate extract of Pasteurella multocida type A (bovine strain) imparted complete protection against an experimental challenge infection with 1.6 x 10(3) colony-forming units of the homologous strain. Formalin-treated vaccine (one or two doses) was much less effective. Significantly higher bactericidal antibody titres were observed in the sera of mice immunized with the extract compared with those given the vaccine. The bactericidal activity was localized in the IgM and IgG fractions. The extract consisted of protein(s) and carbohydrate(s) and on immun…

Peripheral blood mononuclear cells (PBMCs) represent a critical component of the immune system, orchestrating both cellular and humoral responses while maintaining immunological homeostasis. This study investigates the PBMC immune response to Pasteurella multocida (P. multocida) infection in goats using single-cell RNA sequencing (scRNA-seq). PBMCs from goats before and after P. multocida infection were used for scRNA-seq), and pathological examinations were performed on different tissues of the infected hosts. Results demonstrated that recombinase polymerase amplification-lateral flow dipstic…

Pasteurella multocida (Pm) is a significant cause of respiratory disease in beef cattle, with serious consequences for the cattle industry. The bacterial quorum-sensing (QS) system is used for communication and regulation of various processes, including bacterial growth, virulence, biofilm formation and antimicrobial resistance. This study investigated the effects of the C6 signaling molecule on Pm resistance to the antimicrobial enrofloxacin (ENR). Bacteria were divided into groups, with one group (Pm-E1) treated with sub-inhibitory concentrations of ENR, a second treated with sub-inhibitory …

The zoonotic bacterium Pasteurella multocida infects a wide range of animals worldwide. While the genetic diversity of this pathogen is well described in production animals, it remains underexplored in companion animals. In Australia, most P. multocida genomes come from commercial poultry. Here, 59 P. multocida clinical isolates obtained from Australian pets, (cats, dogs, rabbits), farm animals (birds, ruminants, porcine) and captive wildlife (Quolls, Serval) between 2006 and 2023 were sequenced and compared to 523 representative RefSeq genomes. Clustering and phylogenomic analyses placed 24/2…

Pasteurella multocida is notorious for its role as an opportunistic pathogen in infectious bronchopneumonia, the economically most important disease facing cattle industry and leading indication for antimicrobial therapy. To rationalize antimicrobial use, avoiding imprudent use of highly and critically important antimicrobials for human medicine, availability of a rapid antimicrobial susceptibility test is crucial. The objective of the present study was to design a MALDI Biotyper antibiotic susceptibility test rapid assay (MBT-ASTRA) procedure for tetracycline resistance detection in P. multoc…

Male Holtzman rats were given subcutaneous doses of a purified Pasteurella multocida group D heat-labile toxin on alternate days for up to 22 days. Rats were necropsied at 18 days or 36 days (14 days after last dose of toxin) or when moribund, and testicles were taken for histologic and ultrastructural examination. Other selected tissues, including liver and spleen, were taken for histologic examination. Histologically, testicular and splenic lesions occurred more consistently and at much smaller doses when compared with lesions in other target organs such as liver. Testicular and splenic lesi…
