Role of arachidonic acid metabolism in intervertebral disc degeneration: identification of potential biomarkers and therapeutic targets via multi-omics analysis and artificial intelligence strategies.

BACKGROUND: Intervertebral disc degeneration (IVDD) is widely recognized as the primary etiological factor underlying low back pain, often necessitating surgical intervention as the sole recourse in severe cases. The metabolic pathway of arachidonic acid (AA), a pivotal regulator of inflammatory responses, influences the development and progression of IVDD. METHODS: Initially, a comparative analysis was conducted to investigate the relationship between AA expression patterns and different stages of IVDD using single-cell sequencing (scRNA-seq) data. Additionally, three machine learning method

The modified Polsby-Popper score, a novel quantitative histomorphological biomarker and its potential to predict lymph node positivity and cancer-specific survival in oral tongue squamous cell carcinoma.

BACKGROUND: The significance of different histological spreading patterns of tumor tissue in oral tongue squamous cell carcinoma (TSCC) is well known. Our aim was to construct a numeric parameter on a continuous scale, that is, the modified Polsby-Popper (MPP) score, to describe the aggressiveness of tumor growth and infiltration, with the potential to analyze hematoxylin and eosin-stained whole slide images (WSIs) in an automated manner. We investigated the application of the MPP score in predicting survival and cervical lymph node metastases as well as in determining patients at risk in the

Analyzing multidrug resistance patterns across the food supply chain using association rule mining.

We used the machine learning method association rule mining to analyze multidrug resistance (MDR) among cattle-associated Escherichia coli along the food supply chain in the USA. All datasets were stratified by year, source, and resistance indicator (genotypic/phenotypic). Pruned rulesets were compared by calculating the proportion of rules from a comparison ruleset that are captured in a reference ruleset. Rulesets were compared across years within each source and indicator type to quantify how MDR patterns change over time. At the class level, on average nearly 50 % or more of the MDR patter

Population-specific association and risk discrimination utility of the cardiometabolic index for prevalent hypertension-diabetes comorbidity: insights from CHARLS and NHANES.

The comorbidity of cardiometabolic diseases, such as hypertension and diabetes, is increasingly prevalent worldwide, particularly among middle-aged and elderly populations. The cardiometabolic index (CMI), a novel composite biomarker integrating features of dyslipidemia and abdominal obesity, has emerged as a potential indicator. However, its association with prevalent hypertension-diabetes comorbidity and its risk discrimination utility across Chinese and American populations remain insufficiently investigated. This investigation utilized data from CHARLS and NHANES, encompassing 12,261 Chine

Revealing global stoichiometry conservation architecture in cells from Raman spectral patterns.

Cells can adapt to various environments by changing their biomolecular profiles while maintaining physiological homeostasis. What organizational principles in cells enable the simultaneous realization of adaptability and homeostasis? To address this question, we measure Raman scattering light from Escherichia coli cells under diverse conditions, whose spectral patterns convey their comprehensive molecular composition. We reveal that dimension-reduced Raman spectra can predict condition-dependent proteome profiles. Quantitative analysis of the Raman-proteome correspondence characterizes a low-d

Artificial Intelligence-Enabled Evaluation of Pain Sketches to Predict Outcomes in Headache Surgery.

BACKGROUND: Recent evidence has shown that patient drawings of pain can predict poor outcomes in headache surgery. Given that interpretation of pain drawings requires some clinical experience, the authors developed a machine learning framework capable of automatically interpreting pain drawings to predict surgical outcomes. This platform will allow surgeons with less clinical experience, neurologists, primary care practitioners, and even patients to better understand candidacy for headache surgery. METHODS: A random forest machine learning algorithm was trained on 131 pain drawings provided p

Predictive modelling of hypoxic ischaemic encephalopathy risk following perinatal asphyxia.

Hypoxic Ischemic Encephalopathy (HIE) remains a major cause of neurological disability. Early intervention with therapeutic hypothermia improves outcome, but prediction of HIE is difficult and no single clinical marker is reliable. Machine learning algorithms may allow identification of patterns in clinical data to improve prognostic power. Here we examine the use of a Random Forest machine learning algorithm and five-fold cross-validation to predict the occurrence of HIE in a prospective cohort of infants with perinatal asphyxia. Infants with perinatal asphyxia were recruited at birth and neo
