Hypothesized pathogenesis of acardius acephalus, acormus, amorphus, anceps, acardiac edema, single umbilical artery, and pump twin risk prediction.

BACKGROUND: Acardiac twinning complicates monochorionic twin pregnancies in ≈2.6%, in which arterioarterial (AA) and venovenous placental anastomoses cause a reverse circulation between prepump and preacardiac embryos and cessation of cardiac function in the preacardiac. Literature suggested four acardiac body morphologies in which select (groups of) organs fail to develop, deteriorate, or become abnormal: acephalus (≈64%, [almost] no head, part of body, legs), amorphus (≈22%, amorphous tissue lump), anceps (≈10%, cranial bones, well-developed), and acormus (≈4%, head only). We sought to devel

Polygenic Scores for Adult Testosterone and SHBG Levels Are Associated With Reproductive Hormone Levels in Male Infants.

CONTEXT: The hypothalamic-pituitary-gonadal axis's transient activity in infancy, i.e, minipuberty, is considered crucial for male reproductive function. Historically, minipuberty has been considered a passive response triggered by the withdrawal of placental steroids at birth. However, given its potential link to adult reproductive function, we hypothesize that minipuberty is a partially genetically regulated process, suggesting a link between the genetic architecture of reproductive hormone concentrations across lifespan. OBJECTIVE: To investigate the association of UK Biobank Study-based p

Folate metabolizing gene polymorphisms and genetic vulnerability to preterm birth in Korean women.

BACKGROUND: The folate metabolism that converts homocysteine to methionine is closely related to the accumulation of homocysteine. Increased homocysteine levels lead to an impaired antithrombotic function of the vascular endothelium and uterine-placental circulation, resulting in abnormal pregnancy outcomes. Previous studies have reported that gene polymorphisms in folate metabolism are associated with the development of preterm birth (PTB) in various populations. OBJECTIVE: we performed a case-control study to evaluate the association between five polymorphisms in folate metabolic genes (MTH

Adaptation of pancreatic islet B-cells during the last third of pregnancy: regulation of B-cell function and proliferation by lactogenic hormones in rats.

In rodents, placental lactogen (PL)-I is considered to be the first trigger to enhance pancreatic islet B-cell function, and after its secretion is diminished at mid-pregnancy, PL-II takes over this role. However, little information is available on the regulation of islet B-cell function and proliferation by lactogenic hormones during the last third of pregnancy. This was the focus of the present study using rats in which pregnancy was forcibly prolonged. This rat possesses unique characteristics in that PL-I is re-secreted during the prolonged period of pregnancy and the peak concentrations i

A Major Role of VEGFR1 Blockade in Protecting Glomerular Endothelium in Experimental Diabetes.

BACKGROUND: Microvascular injury is a critical factor in the pathogenesis of diabetic nephropathy. Dysregulation in vascular endothelial growth factor-A (VEGF-A) signalling pathways exacerbates endothelial dysfunction, contributing to the progression of kidney disease. In this study, we tested whether selective blockade of VEGFR1, a decoy receptor that limits VEGF-A availability for VEGFR2, could restore glomerular microvascular integrity in experimental diabetic nephropathy. METHODS: Male BTBR ob/ob mice, a model of advanced Type 2 diabetic nephropathy, received chronic treatment with a mono

Methyltransferase-like 14 (METTL14)-mediated N6-methyladenosine (m6A) Modification of Forkhead Box Protein 1 (FOXP1) Regulates Trophoblast Inflammation and Function via Transmembrane BAX Inhibitor Motif-Containing 6 (TMBIM6).

Pre-eclampsia (PE) represents a serious pregnancy complication characterized by impaired trophoblast function. Although methyltransferase-like 14 (METTL14) has been implicated in PE pathogenesis and trophoblast dysfunction, its precise molecular mechanisms remain unclear. mRNA expression was detected by quantitative real-time polymerase chain reaction (qRT-PCR). Immunoblot analysis was used for protein expression evaluation. Total N6-methyladenosine (m6A) RNA methylation was analyzed using an assay kit. Cell tube formation, invasion and colony formation were assessed by tube formation, transwe

Selenium supplementation and pregnancy outcomes.

In vertebrates and invertebrates, selenium (Se) is an essential micronutrient, and Se deficiency or excess is associated with gonadal insufficiency and gamete dysfunction in both males and females, leading to implantation failure, altered embryonic development and, ultimately, infertility. During pregnancy, Se excess or deficiency is associated with miscarriage, pre-eclampsia (hypertension of pregnancy), gestational diabetes, fetal growth restriction and preterm birth. None of this is surprising, as Se is present in high concentrations in the ovary and testes, and work in animal models has sho

Exposure to airborne fine particulate matter is associated with impaired endothelial function and biomarkers of oxidative stress and inflammation.

Epidemiological evidence suggests that exposure to air pollution is a leading risk factor for cardiovascular disease (CVD). However, there is little direct evidence linking exposure to vascular dysfunction. We conducted a cross-sectional study of 100 participants, recruited from the University of Louisville Clinics. Endothelial function was assessed by calculating the reactive hyperemia index (RHI). Oxidative stress was indexed by measuring urinary levels of isoprostanes (n = 91). Inflammatory biomarkers were measured in the plasma (n = 80). Daily average PM2.5 levels were obtained from region

Hyperandrogenism and insulin resistance induce gravid uterine defects in association with mitochondrial dysfunction and aberrant reactive oxygen species production.

Women with polycystic ovary syndrome (PCOS) are at increased risk of miscarriage, which often accompanies the hyperandrogenism and insulin resistance seen in these patients. However, neither the combinatorial interaction between these two PCOS-related etiological factors nor the mechanisms of their actions in the uterus during pregnancy are well understood. We hypothesized that hyperandrogensim and insulin resistance exert a causative role in miscarriage by inducing defects in uterine function that are accompanied by mitochondrial-mediated oxidative stress, inflammation, and perturbed gene exp

Intestine-specific expression of human chimeric intestinal alkaline phosphatase attenuates Western diet-induced barrier dysfunction and glucose intolerance.

Intestinal epithelial cell derived alkaline phosphatase (IAP) dephosphorylates/detoxifies bacterial endotoxin lipopolysaccharide (LPS) in the gut lumen. We have earlier demonstrated that consumption of high-fat high-cholesterol containing western type-diet (WD) significantly reduces IAP activity, increases intestinal permeability leading to increased plasma levels of LPS and glucose intolerance. Furthermore, oral supplementation with curcumin that increased IAP activity improved intestinal barrier function as well as glucose tolerance. To directly test the hypothesis that targeted increase in

Out on a limb: bandicoot limb co-variation suggests complex impacts of development and adaptation on marsupial forelimb evolution.

Marsupials display far less forelimb diversity than placentals, possibly because of the laborious forelimb-powered climb to the pouch performed by most marsupial neonates. This is thought to result in stronger morphological integration (i.e., higher co-variance) within the marsupial forelimb skeleton, and lower integration between marsupial fore- and hind limbs, compared to other mammals. Possible mechanisms for this constraint are a fundamental developmental change in marsupial limb patterning, or alternatively more immediate perinatal biomechanical and metabolic requirements. In the latter c

Systematic Methods to Resolve Lineage-Specific Stress States in Early Mammalian Embryos and That May Enable Miscarriage Prediction.

Early mammalian embryos are highly sensitive to environmental, metabolic, hormonal, and genomic stress, yet embryo assessment during In Vitro Fertilization (IVF) relies largely on morphology and ploidy for embryo assessment, but these tests incompletely predict miscarriage. We present a transcriptomics based framework to classify and quantify lineage-specific stress in early embryos by benchmarking human preimplantation embryos against dose-, time-, and quality-dependent stress programs defined in Embryonic and placental Trophoblast Stem Cells (ESCs, TSCs) from the implanting blastocyst. Adverse Fetal Outcomes and Histopathology of Placentas Affected by COVID-19: A Report of Four Cases.

Coronavirus disease 2019 (COVID-19) has had significant impacts on mothers and neonates. In this report, we present four unique cases of COVID-19 infections in pregnancy and its effects on the mother, fetus, and placenta. Four mothers presented to the hospital during their pregnancy. Each had tested COVID-19-positive 1-29 days prior to admission. Gestational age ranged from 16 weeks six days to 36 weeks six days. Three of the four cases resulted in fetal demise or infant expiration. The common finding among all four cases was pathologic changes in the placenta. Most of the placentas were small

miR‑181b‑5p inhibits trophoblast cell migration and invasion through targeting S1PR1 in multiple abnormal trophoblast invasion‑related events.

Normal placentation and successful maintenance of pregnancy depend on the successful migration and invasion of trophoblasts into maternal tissues. Previous studies reported that microRNAs (miRs) are expressed in trophoblasts, and can regulate their migration and invasion. The present study aimed to investigate miR‑181b‑5p function in HTR‑8/SVneo trophoblasts and explore its underlying mechanism in the pathogenesis of multiple abnormal trophoblast invasion‑related events. Reverse‑transcription quantitative PCR and western blotting were used to test the expression of miR‑181b‑5p and sphingosine‑

Blood urea nitrogen may serve as a predictive indicator of retained placenta in dairy cows.

Retained placentae (RP) results in significant economic losses to dairy farmers. In Experiment 1, to screen biochemical indicators of RP, 21 cows with RP and 21 cows with no retained placenta (NRP) were selected as a control group, and blood was collected at -7 d, 0 h (parturition) and 12 h. Serum biochemical indicators were ascertained. Results indicate serum concentrations of phosphorus (P) and blood urea nitrogen (BUN) in cows of the RP group were markedly greater than in cows of the NRP group at -7 d (P < 0.01). In Experiment 2, to evaluate predictive indicators for RP, 34 cows with RP and

Valproic acid stimulates in vitro migration of the placenta-derived mesenchymal stem/stromal cell line CMSC29.

BACKGROUND: The placenta is an abundant source of mesenchymal stem/stromal cells (MSC), but our understanding of their functional properties remains limited. We previously created a placental-derived chorionic MSC (CMSC) cell line to overcome the difficulties associated with conducting extensive ex vivo optimization and experimental work on primary cells. The aim of this study was to characterize the migratory behavior of the CMSC29 cell line in vitro. METHODS: Stimulators of MSC migration, including two cytokines, stromal cell-derived factor-1α (SDF-1α) and hepatocyte growth factor (HGF), an

Fine-scale quantification of HCG beta gene transcription in human trophoblastic and non-malignant non-trophoblastic tissues.

Human chorionic gonadotropin (HCG) is produced by syncytiotrophoblast of placenta. It delays the apoptosis of corpus luteum and functions in implantation. Its possible role in male reproduction has been raised. HCG beta subunit is encoded by CGB, CGB5, CGB7 and CGB8 genes located at 19q13.3 in a common genome cluster with beta subunit non-coding CGB1 and CGB2. We conducted a sensitive quantification and comparison of CGB gene expression in human trophoblastic (blastocysts, n = 6; normal/failed pregnancy, n = 51) and non-malignant non-trophoblastic tissues (15 different tissue types, samples n

Cancer risk related to mammary gland structure and development.

The breast undergoes dramatic changes in size, shape, and function in association with growth, reproduction, and post-menopausal regression. Those changes impact women's lifetime breast cancer risk. An early first full-term pregnancy exerts a protective effect, emphasizing the need for understanding the role of reproductive influences on breast development and on cancer initiation and progression, and providing a paradigm for developing preventive strategies based on physiological principles. Even though the cause of breast cancer and the ultimate mechanisms through which an early pregnancy pr

The Impacts of Pregnancy on Cognition and Cell Proliferation in a Live-Bearing Fish (Poeciliopsis gracilis).

Pregnancy is a high-energy process which temporarily decreases cognitive function and affects the neurogenic capacity of the maternal brain. Pregnant females exhibit decreased performance in spatial memory tasks which has been linked to altered neurogenesis in the dentate gyrus of the hippocampus. While these processes are well-resolved for mammals, whether they are conserved across other, nonmammalian, live-bearing animal lineages without placentation remains enigmatic. Here, we test the relationship between pregnancy and cognition in the live-bearing fish Poecilipsis gracilis. Female P. grac

Disruption of LDL receptor trafficking in placentas from maternal supraphysiological hypercholesterolemia: A study of key endocytosis and recycling proteins.

INTRODUCTION: During pregnancy, some women experience elevated cholesterol levels, known as maternal supraphysiological hypercholesterolemia (MSPH), which can cause oxidative stress and endothelial dysfunction in the fetus. Although cholesterol levels in newborns of MSPH mothers do not differ from those of mothers with physiological hypercholesterolemia (MPH), cholesterol transport through the placenta is affected. This study examines proteins involved in the endocytosis and recycling of the low-density lipoprotein receptor (LDLR) in MSPH placentas, highlighting alterations in LDLR localizatio
