Mammalian eyes are exposed to visible light but cannot perform photosynthesis. Here, we show that introducing a nanoscale, structurally and functionally preserved thylakoid system, LEAF (light-reaction enriched thylakoid NADPH-foundry), into corneal cells enables light-driven bona fide photosynthetic production of NADPH and ATP, similar to plant leaves, which alleviates oxidative stress and inflammation. LEAF acts in two domains. Intracellularly, it integrates with host cells to supply NADPH and ATP via intact photosynthetic electron transport, restoring redox balance. Extracellularly, photosy

The cometabolic degradation of trichloroethene (TCE) by Rhodococcus sp. L4 was limited by the loss of enzyme activity during TCE transformation. This problem was overcome by repeated addition of inducing substrates, such as cumene, limonene, or cumin aldehyde, to the cells. Alternatively, Rhodococcus sp. L4 was immobilized on plant materials which contain those inducers in their essential oils. Cumin seeds were the most suitable immobilizing material, and the immobilized cells tolerated up to 68 muM TCE and degraded TCE continuously. The activity of immobilized cells, which had been inactivate

Protein-based therapeutics have transformed modern disease treatment and now constitute a rapidly expanding segment of the pharmaceutical market. In this context, plant-based expression systems have emerged as a versatile alternative to conventional mammalian cell platforms for the production of recombinant proteins. Beyond their well-established advantages in cost, scalability, and biosafety, recent advances in transient expression technologies and plant cell culture systems have enabled rapid, high-yield production of complex biologics with increasing control over product quality. Protein synthesis rate is distinct from steady-state protein abundance and must be measured independently to understand how cells regulate protein levels and respond to environments. Technologies used to predict or directly quantify protein synthesis include ribosome profiling, radioactive and stable isotope labelling, and bioorthogonal noncanonical amino acid tagging (BONCAT). Here, we emphasise BONCAT's potential in plants, which, when combined with click-chemistry, enables the detection, enrichment, and characterisation of newly synthesised proteins themselves. We summarise lessons from BON

Plant activators are compounds, such as analogs of the defense hormone salicylic acid (SA), that protect plants from pathogens by activating the plant immune system. Although some plant activators have been widely used in agriculture, the molecular mechanisms of immune induction are largely unknown. Using a newly established high-throughput screening procedure that screens for compounds that specifically potentiate pathogen-activated cell death in Arabidopsis thaliana cultured suspension cells, we identified five compounds that prime the immune response. These compounds enhanced disease resist

The brown planthopper (BPH), Nilaparvata lugens (Stål) (Hemiptera: Delphacidae), is a serious insect pest on rice. It uses its stylet to collect sap by penetrating the phloem and at the same time it delivers saliva into the host plant, which can trigger a reaction. The molecular mechanisms by which BPH salivary proteins result in plant responses are poorly understood. In this study, we screened transcriptomic data from different BPH tissues and found a protein specific to the salivary gland, NlG14, that could induce cell death in plants. We determined that NlG14 is uniquely found in the insect

Plant cell wall-associated polygalacturonase-inhibiting proteins (PGIPs) are widely distributed in the plant kingdom. They play a crucial role in plant defense against phytopathogens by inhibiting microbial polygalacturonases (PGs). PGs hydrolyze the cell wall polysaccharide pectin and are among the first enzymes to be secreted during plant infection. Recent studies demonstrated that herbivorous insects express their own PG multi-gene families, raising the question whether PGIPs also inhibit insect PGs and protect plants from herbivores. Preliminary evidence suggested that PGIPs may negatively

Herein, we have reported on the efficiency of inorganic Zn3N2 nanoparticles for labeling plant cells and animal cells toward imaging applications with negligible toxicity. We have synthesized zinc nitride (Zn3N2) colloidal nanoparticles with an average size of 25 nm at room temperature. The optical band gap of the prepared Zn3N2 nanoparticles is 2.8 eV and gives a visible range emission at 415 nm. With the addition of Zn3N2 colloids to organic dyes such as protoporphyrin, flavin adenine dinucleotide, fluorescein, and neutral red, the emission intensity of the organic dyes enhanced from 3 to 20…

C. Robin Buell has been a leading figure in plant genomics since the advent of DNA sequencing technology. She helped lead multiple consortia to sequence some of the first crop genomes at the turn of the millennium. She has since used the genomes to tackle questions in fundamental biology, plant evolution, and agriculture. Recently, she applied single-cell technologies to uncover how complex biosynthetic pathways are compartmentalized across different rare plant cell types. Now at the University of Georgia, Buell explores in her Inaugural Article how tubers arose repeatedly in the plant family

Polyamines, γ-aminobutyric acid (GABA), proline, and glycine betaine (GB) are important plant stress metabolites that are interconnected in common metabolic cycles. These compounds have a multifunctional protective effect on plant cells under stress conditions, acting as osmoregulators, antioxidants, chaperones, and intracellular pH stabilizers, among other roles. The key role of these compounds, however, lies in their involvement in the signaling network of plant cells. Stress metabolites, in particular, engage in a complex functional interaction with reactive oxygen species (ROS) and nitric …

Polyethylene glycol calcium (PEG-Ca2+)-mediated transfection allows rapid and efficient examination to analyze diverse cellular functions of genes of interest. In plant cells, macromolecules, such as DNA, RNA and protein, are delivered into protoplasts derived from somatic tissues or calli via PEG-Ca2+ transfection. To broaden and develop the scope of investigations using plant gametes and zygotes, a procedure for direct delivery of macromolecules into these cells has recently been established using PEG-Ca2+ transfection. This PEG-Ca2+-mediated delivery into rice egg cells/zygotes consists of …

Hydrogen sulphide (H2 S) is an endogenously produced gasotransmitter that has rapidly emerged as an active signalling component of several plant processes, stomatal movement regulation among them. The guard cells (GCs), pairs of cells that neighbour the stomatal pores, transduce endogenous and environmental signals, through signalling network, to control stomatal pore size. In this complex network, which has become a model system for plant signalling, few highly connected components form a core that links most of the pathways. The evidence summarized in this insight, on the interplay between H

Plant height is a key determinant of wheat plant architecture that affect lodging resistance and grain yield. The GA-responsive (GAR) dwarfing gene Rht5 was previously shown to decrease plant height without compromise of wheat seedling vigor and considered as a promising candidate gene for breeding wheat varieties in water-limited conditions. However, the mechanisms underlying Rht5-mediated dwarfism are unclear. In this study, we investigated the genetic effects of Rht5 on wheat growth and development using recombinant inbred lines (RILs) and found that Rht5 reduces plant height through inhibi

In this study, Hedychium coccineum tetraploid plants and octaploid plants induced by colchicine were used as materials. The ploidy levels were precisely identified by combining root tip squash and flow cytometry analyses, and the differences between plants of different ploidy levels were systematically investigated at cytological, morphological, and molecular levels. The results showed that the highest polyploid induction efficiency was achieved when callus tissues were treated with 0.1 g/L colchicine for 4 days. The fluorescence peak value of the induced plants was twice that of the tetraploi

Biotrophic fungal pathogens such as Ustilago maydis rely on a host to complete their lifecycle. U. maydis has the advantage, in contrast to obligate biotrophs such as rust fungi, that the saprophytic yeast-like sporidia can be cultivated in artificial media, while the dikaryotic filamentous and infectious stage depends on the host to complete the sexual phase of the lifecycle. Key phenotypes during plant infection include filamentous growth after mating, an increase of fungal biomass in developing tumors, production of an extracellular matrix (ECM) around fungal cells, and melanin formation du
