Mx proteins are induced by type I interferons (IFN alpha and beta) in mice and humans and inhibit the replication of orthomyxoviruses and some other single-stranded RNA viruses. Recently, Mx genes have been cloned from Atlantic salmon. Mx transcripts were shown to be induced in head-kidney, liver and gills of the fish by the synthetic double-stranded RNA polyinosinic polycytidylic acid (poly I:C). In the present work we have studied expression of Mx protein in organs of Atlantic salmon treated with poly I:C. A quantitative immunoblot method was established to monitor expression of Mx protein a

BACKGROUND: Recent findings implicate the histone deacetylase (HDAC) inhibitor largazole as an inhibitor of Musashi RNA-binding protein function. Here, we assess this interplay and evaluate the relevance of largazole for triple-negative breast cancer (TNBC) progression and resistance to radiotherapy.
METHODS: Primary patient-derived TNBC cells and cell lines were treated with largazole and cell vitality, proliferation, motility, cell cycle, DNA synthesis as well as repair, and stemness were analyzed via MTT assay, digital holographic microscopy, and flow cytometry. To unravel the connection b

Major-groove RNA triple helices are conserved tertiary structures formed when a third strand inserts into the major groove of a classic double helix either via Hoogsteen base pairing or reverse Hoogsteen base pairing. These structures are widely distributed in eukaryotic, prokaryotic, and viral RNAs. This paper systematically summarizes the structural classification, stability-influencing factors, and identification methods of RNA triple helices. A growing body of evidence indicates that RNA triple helices are extensively involved in diverse biological processes, including RNA stability regula

BACKGROUND: Rehmannia glutinosa, a herb of significant medicinal and food value, is highly susceptible to pathogen infections. Fusarium oxysporum f.sp. R. glutinosa (FORg), a pathogenic strain isolated from R. glutinosa, induces root rot disease, leading to severe yield losses. Currently, resistant cultivars and effective control strategies for R. glutinosa root rot remain very limited. This study aims to investigate the feasibility and potential of SIGS technology in preventing and controlling R. glutinosa root rot caused by FORg.
RESULTS: This study demonstrates that FORg hyphae can capably

The biology of the viral life cycle essentially includes two structural and functional entities-the viral genome and protein machinery constituting the viral arsenal and an array of host cellular components which the virus closely associates with-to ensure successful perpetuation. The obligatory requirements of the virus to selectively evade specific host cellular factors while exploiting certain others have been immensely important to provide the platform for designing host-directed antiviral therapeutics. Although the spectrum of host-virus interaction is multifaceted, host factors that part

Retinoic acid-inducible gene I (RIG-I), a key pattern recognition receptor (PRR) detecting cytosolic 5'-triphosphorylated double-stranded RNA (5'-ppp dsRNA), mediated antitumor immunity. Here, we evaluated RIG-I agonists as potential antitumor agents. To analyze the anti-tumor efficacy, we engineered a panel of 5'-PPP-modified stem-loop-structured RNAs leveraging the non-coding sequence of SARS-CoV-2. Through systematic screening, nCoV-L emerged as a potent RIG-I agonist that induced death in hepatocellular carcinomas (HCC), pulmonary carcinomas, and colorectal cancer (CRC) in vitro and suppre

Hepatitis B virus (HBV) specifically infects hepatocytes and causes severe liver diseases. The HBV life cycle is unique in that the genomic DNA (relaxed-circular partially double-stranded DNA: rcDNA) is converted to a molecular template DNA (covalently closed circular DNA: cccDNA) to amplify a viral RNA intermediate, which is then reverse-transcribed back to viral DNA. The highly stable characteristics of cccDNA result in chronic infection and a poor rate of cure. This complex life cycle of HBV offers a variety of targets to develop antiviral agents. We provide here an update on the current kn

Plant-parasitic nematodes cause major yield and economic losses and remain difficult to manage because of their soil-borne nature, persistent biology, and broad host range. Conventional nematicides provide rapid suppression but face regulatory, safety, and environmental concerns, while cultural practices, biological control, and host resistance are constrained by variable field performance, durability, or breeding limitations. Molecular biopesticides, including double-stranded RNA (dsRNA), recombinant proteins/peptides, and characterized secondary metabolites, offer mechanism-based targeting o

The pine wood nematode, Bursaphelenchus xylophilus, causes severe damage to pine trees. Current chemical control methods have raised environmental concerns and the resistance issue of the nematode. Due to its high specificity, RNA interference (RNAi) has emerged as a promising approach for controlling pests, including plant-parasitic nematodes, through the application of double-stranded RNA (dsRNA). This study has developed an effective dsRNA-based nematicide for controlling B. xylophilus under field conditions via trunk injection. First, the core RNAi machinery of B. xylophilus was identified

In response to DNA double-strand breaks (DSBs), DNA damage repair factors are recruited to DNA lesions and form nuclear foci. However, the underlying molecular mechanism remains largely elusive. Here, by analyzing the localization of DSB repair factors in the XY body and DSB foci, we demonstrate that pre-ribosomal RNA (pre-rRNA) mediates the recruitment of DSB repair factors around DNA lesions. Pre-rRNA exists in the XY body, a DSB repair hub, during meiotic prophase, and colocalizes with DSB repair factors, such as MDC1, BRCA1 and TopBP1. Moreover, pre-rRNA-associated proteins and RNAs, such

Herpes simplex virus 1 (HSV-1), a double-stranded DNA virus, infects epithelial surfaces and establishes latency in the central nervous system, where astrocytes are a major immune cell type. Here, we report changes that occur in the expression of pathogen recognition receptors, such as Toll-like receptors, DNA and RNA sensors, interferons, and interferon-stimulated genes, when astrocytes are infected with HSV-1 strain F. We observed upregulation of Toll-like receptors 2, 6 and 9, MDA5, and DAI along with an increase in the expression of type I interferons and interferon-stimulated genes such a

The T7 RNA polymerase-dependent transcription was studied as a function of nucleotide sequence structures positioned upstream of the T7 promoter. Model double-stranded DNA templates were constructed for this purpose. They contained a target sequence of 485 base pairs (cDNA fragment of Venesuelian encephalomyelitis equine virus genome), T7 promoter consensus and different extra base sequences upstream of the T7 promoter. The level of the target sequence transcription was clearly determined by the extra base sequence. The presence of one extra base pair G.C ensured the most pronounced effect, tr

The helicase domain of nonstructural protein 3 (NS3H) unwinds the double-stranded RNA replication intermediate in an ATP-dependent manner during the flavivirus life cycle. While the ATP hydrolysis mechanism of Dengue and Zika viruses NS3H has been extensively studied, little is known in the case of the tick-borne encephalitis virus NS3H. We demonstrate that ssRNA binds with nanomolar affinity to NS3H and strongly stimulates the ATP hydrolysis cycle, whereas ssDNA binds only weakly and inhibits ATPase activity in a noncompetitive manner. Thus, NS3H is an RNA-specific helicase, whereas DNA might

Positive-stranded viruses synthesize their RNA in membrane-bound organelles, but it is not clear how this benefits the virus or the host. For coronaviruses, these organelles take the form of double-membrane vesicles (DMVs) interconnected by a convoluted membrane network. We used electron microscopy to identify murine coronaviruses with mutations in nsp3 and nsp14 that replicated normally while producing only half the normal amount of DMVs under low-temperature growth conditions. Viruses with mutations in nsp5 and nsp16 produced small DMVs but also replicated normally. Quantitative reverse tran

Avian reovirus (ARV) is a double-stranded RNA virus that can cause immunosuppression, irregular bleeding, spleen necrosis, and other symptoms in ducks, posing a serious threat to the poultry industry. Previous studies have confirmed that the duck-derived linc000889 can inhibit ARV replication. In this study, qRT-PCR revealed that linc000889 could promote the expression of interferon-β (IFN-β) and its downstream genes Mx and OASL. Subsequently, using RNA pull-down and protein mass spectrometry, we identified that the host protein NLRX1 associates with the sense strand of linc000889. This bindin

Rabies virus is a highly neurophilic negative-strand RNA virus with high lethality and remains a huge public health problem in developing countries to date. The double-stranded RNA-binding protein Staufen1 (STAU1) has multiple functions in RNA virus replication, transcription, and translation. However, its function in RABV infection and its mechanism of action are not clear. In this study, we investigated the role of host factor STAU1 in RABV infection of SH-SY-5Y cells. Immunofluorescence, TCID50 titers, confocal microscopy, quantitative real-time PCR and Western blotting were carried out to

UNLABELLED: Persistent viral infections are common in nature and can play a significant role in the ecology and evolution of the host organism. However, our understanding of these prolonged virus-host interactions remains limited. This study characterizes the persistent infection strategy of Pseudomonas phage phi6, an enveloped virus with a tri-segmented, double-stranded (ds)RNA genome that infects Pseudomonas syringae. Phi6 is predominantly lytic but can alternatively establish a non-productive chronic infection (carrier cell state) in which viral particles assemble and replicate viral RNA wi

Virus recognition and response by the innate immune system are critical components of host defense against infection. Activation of cell-intrinsic immunity and optimal priming of adaptive immunity against West Nile virus (WNV), an emerging vector-borne virus, depend on recognition by RIG-I and MDA5, two cytosolic pattern recognition receptors (PRRs) of the RIG-I-like receptor (RLR) protein family that recognize viral RNA and activate defense programs that suppress infection. We evaluated the individual functions of RIG-I and MDA5 both in vitro and in vivo in pathogen recognition and control of

Hepatitis B virus infection continues to be a major global health problem with an estimated 350 million carriers. The response to available treatment modalities is not impressive. The advent of RNA Interference--a phenomenon of sequence-specific degradation of RNAs mediated by double-stranded RNA--holds promise as a potential therapy for chronic hepatitis B virus infection. Synthetic preparations of short RNA (21-23 bp long) can be used to mediate this process of gene silencing with a lower immune response. The duration of suppression can be further increased by using a vector delivery system.

Adenosine deaminases acting on RNA (ADAR) catalyze the deamination of adenosine to inosine in double-stranded RNA. Because inosine is read as guanosine during translation, this process enables programmable A-to-G recoding at the transcript level. ADARs can be harnessed for therapeutic correction of pathogenic mutations through site-directed RNA editing with guide RNAs. To expand the design space of editing-enabling guides, we applied EMERGe, a high-throughput screening platform, to identify motifs targeting a premature termination codon in the MeCP2 transcript associated with Rett syndrome.
