Molecular identification of Ancylostoma caninum isolated from cats in southern China based on complete ITS sequence.

Ancylostoma caninum is a blood-feeding parasitic intestinal nematode which infects dogs, cats, and other mammals throughout the world. A highly sensitive and species-specific PCR-RFLP technique was utilised to detect the prevalence of A. caninum in cats in Guangzhou, southern China. Of the 102 fecal samples examined, the prevalence of A. caninum in cats was 95.1% and 83.3% using PCR-RFLP and microscopy, respectively. Among them, the prevalence of single hookworm infection with A. caninum was 54.90%, while mixed infections with both A. caninum and A. ceylanicum were 40.20%. Comparative analysis

Revealing cryptic parasite diversity in a definitive host: echinostomes in muskrats.

Many trematode groups have a long history of systematic revision, which can make parasite identification a difficult task. The trematode parasites of muskrats are no exception. Here, we highlight the systematic issues associated with trematodes of muskrats (Ondatra zibethicus). Then, we demonstrate the utility of using both morphological and molecular tools to identify these parasites. Morphological examinations of specimens from muskrats (n = 63) first suggested that at least 4 genera were present including Echinostoma, Wardius, Quinqueserialis, and Notocotylus. For the latter 3 groups, the 2

Human U6 promoter drives stronger shRNA activity than its schistosome orthologue in Schistosoma mansoni and human fibrosarcoma cells.

Blood flukes or schistosomes are the causative agents of human schistosomiasis, one of the major neglected tropical diseases. Draft genome sequences have been reported for schistosomes, but functional genomics tools are needed to investigate the role and essentiality of the newly reported genes. Vector based RNA interference can contribute to functional genomics analysis for schistosomes. Using mRNA encoding reporter firefly luciferase as a model target, we compared the performance of a schistosome and a human promoter from the U6 gene in driving shRNA in human fibrosarcoma cells and in cultur

A preliminary survey of feline filarial parasites in Kerala State, India.

Feline filarial parasites remain among the least investigated in India, despite the well-established endemicity of filariosis in canine and human populations. This study aimed to assess filarial infections in domestic cats in Kerala using both microscopy and molecular techniques, with a focus on evaluating their potential role as reservoir hosts. From September 2023 to February 2024, 300 owned cats presented to the University Veterinary Hospital, Mannuthy, were screened for circulating microfilariae using wet blood film examination. Four cats (1.33%) tested positive, of which three harbored sh

Import of exotic and zoonotic trematodes (Heterophyidae: Centrocestus sp.) in Xiphophorus maculatus: implications for ornamental fish import control in Europe.

Ornamental fish, Xiphophorus maculatus, were imported from Singapore to Denmark for distribution to local aquarists. Importers observed lethargic and erratic swimming patterns among fish and forwarded a total of 30 fish for pathological examination to a university diagnostic service. All fish were diagnosed infected with encysted Centrocestus sp. metacercariae in gills (prevalence of 100% and mean intensity of 454.5 ± 161.9 parasites per fish). Metacercariae were identified by morphological and molecular methods. Cysts (mean length 163.3 ± 13.7 μm and mean width 113.3 ± 10.6 μm) contained a be

Differences in microthrix form between stomach and spiral intestine stages of the cestode, Trilocularia: interspecific variation or development?

Tetraphyllidean cestodes of the genus Trilocularia have been reported from Squalus acanthias throughout this shark's global distribution, occupying both the stomach and spiral intestine. This study investigated whether variation seen in microthrix form among specimens within this host species is indicative of interspecific variation or is attributable to developmental transformation of a single cestode species. Sequence data for the 2 nuclear genes 28S rDNA (D1-D3) and ITS1, and the mitochondrial gene 16S rDNA, were generated for 22 specimens of Trilocularia; 3 from the stomach and 19 from the

Structure of three high voltage-activated calcium channel alpha1 subunits from Schistosoma mansoni.

Voltage-gated calcium (Ca2+) channels contribute to impulse propagation in excitable cells and also regulate intracellular levels of Ca2+. High voltage-activated (HVA) Ca2+ channels are heteromultimeric membrane proteins. The pore-forming, voltage-sensing subunit is the alpha1 subunit. We have cloned 3 HVA Ca2+ channel alpha1 subunit cDNAs from Schistosoma mansoni. One of these sequences most closely resembles the L-type class of HVA alpha1 subunits. The other two sequences are most closely related to non L-type alpha1 subunits. These schistosome alpha1 subunits have many of the features commo

Immunodiagnosis of fasciolosis using recombinant procathepsin L cystein proteinase.

Cathepsin L1, a cysteine protease secreted by the gastrodermis of juvenile and adult Fasciola hepatica, was expressed in Escherichia coli as a fusion protein containing the proregion, supplied with six histidyl residues at the N-terminal end (rproCL1). In this study we tested its potential as antigen for the serologic diagnosis of F. hepatica infections by enzyme-linked immunosorbent assay (ELISA). The analyzed human sera included 16 positive samples, 99 negative controls and 111 from individuals affected by other parasitic and non parasitic diseases. The sensitivity and specificity of the rpr

The humoral response to heat shock protein 70 in human and murine Schistosomiasis mansoni.

We have expressed two cDNA sequences encoding 121 and 230 amino acids of the C-terminus of the Schistosoma mansoni Hsp 70 in Escherichia coli. The products were synthesized as polypeptides fused to the RNA polymerase of bacteriophage MS2, and their reactivities were tested in ELISAs, using sera from human and murine infections. Anti-Hsp70 antibodies were detected in a significant number of individuals suffering from chronic schistosomiasis mansoni, but not in patients with known recent infections. This, together with the finding that antibodies directed at S. mansoni-specific Hsp70 determinant

Intermittent Administration of Helminth-Derived Fh15 Modulates Gut Microbiota and Partially Mitigates Dysbiosis in Early Stages of Severe Experimental Colitis.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by dysbiosis of the gut microbiota. Helminth infections are known to modulate host immunity and intestinal microbial composition; however, the therapeutic use of live parasites poses safety challenges. The recombinant Fasciola hepatica fatty acid-binding protein Fh15 is a helminth-derived molecule with anti-inflammatory effects in models of septic shock and dextran sulfate sodium (DSS)-induced colitis. Whether Fh15 also influences gut microbial composition during colitis remains unknown. Male C57BL/6 mice received 4%

The history continues: From Steringotrema microacetabularis (Fellodistomidae) to Bacciger microacetabularis (Baccigeridae) genetic evidence reveals a new genus of Cryptogonimidae.

Steringotrema microacetabularis Suriano & Martorelli, 1983 (Fellodistomidae) was described from the flounder Paralichthys orbignyanus. Later, it was redescribed, based on new material from the same host and type locality, and reconsidered as Bacciger microacetabularis (Baccigeridae). The main difference noted in the redescription was the presence of spines on the body. However, the lack of DNA data made confirming the true affiliation of this digenean challenging. New specimens sampled from P. orbignyanus allowed us to sequence the 28S, ITS, and COI genes. Fresh specimens were stained to compa

NEW SPECIES OF AXINE ABILDGAARD, 1794 (MONOGENOIDEA: AXINIDAE) INFECTING GILL LAMELLAE OF ATLANTIC FLYINGFISH, CHEILOPOGON MELANURUS (VALENCIENNES) (EXOCOETIDAE) IN THE WESTERN ATLANTIC OCEAN WITH PHYLOGENETIC ANALYSIS AND COMMENTS ON THE FEMALE GENITALIA OF AXINE SPP.

We collected specimens of Axine trickyvagina Brule and Bullard n. sp. (Monogenoidea: Axinidae) from the gill lamellae of Atlantic flyingfish, Cheilopogon melanurus (Valenciennes) (Exocoetidae) in the north-central Gulf of America. Specimens of the new species were heat-killed and formalin-fixed for morphology, and others were preserved in 95% EtOH for DNA extraction and sequencing of the 28S gene and ITS1 region. The new species differs from all congeners by the combination of having a long haptor (∼40-50% of the total body length), a male copulatory organ with 10-15 spines, and a genital atri

Molecular identification of heterophyids in human stool samples from the Philippines.

The family Heterophyidae includes endoparasitic minute intestinal flukes that are primarily transmitted through the consumption of raw or undercooked fish, causing heterophyidiasis. Several heterophyid species have been reported to infect humans in the Philippines. Heterophyidiasis is diagnosed by detection of parasite ova in stool samples through Kato-Katz stool microscopy, but species identification is challenging due to the morphologically similar eggs of different heterophyid species and other minute intestinal flukes such as Clonorchis and Opisthorchis. This study addresses the need for a

A Novel Filarial-Multiplexed Probe-Quantitative PCR for the Advance in the Diagnosis of Multiple Infections With Human Filariasis.

The routine diagnostic method used for clinical samples suspected of filarial infection in our laboratory is the Filaria-real time-PCR (F-RT-PCR). The drawback of this method is the need for melting temperature analysis and PCR products' electrophoresis to identify the filarial species. Therefore, the aim of this study was to design a quantitative real-time PCR (qPCR) assay using filarial-specific hydrolysis probes, targeting 18S rRNA and ITS1 genes, allowing the simultaneous diagnosis of loiasis, mansonellosis and other human filariasis without the need of electrophoresis or melting temperatu

Phylogenetic position of Ancyrocephalus (sensu lato) curtus Achmerov, 1952 (Monopisthocotylea, Dactylogyridae), a parasite of fish Perccottus glenii Dybowski, 1877(Gobiiformes: Odontobutidae).

The genus Ancyrocephalus sensu lato is a large assemblage of species of dactylogyrid monopisthocotyleans without clear taxonomic boundaries. Despite an urgent need for revision, only three representatives of this taxon have been molecularly characterised so far. We found specimens of Ancyrocephalus curtus, a previously non-genotyped species, in gills of Perccottus glenii caught in the River Syumnyur, Amur Basin, Russia. The aim of this study was to assess the phylogenetic position of this parasite using partial sequences of 28S rRNA gene. In the phylogenetic tree, A. curtus appeared as a siste

Redescription and Molecular Characterisation of Derogenes ruber Lühe, 1900 (Hemiuroidea: Derogenidae) from Chelidonichthys lastoviza (Scorpaeniformes: Triglidae) in the Western Mediterranean.

PURPOSE: Derogenes ruber Lühe, 1900, the type-species of the genus Derogenes Lühe, 1900, is a poorly known derogenid digenean. The original description of this species was not illustrated and aspects of the morphology of the parasite from the type-host remain scarce. Available records of this species were brief and/or lacked illustrations and were based on morphology alone. Additionally, molecular data for Derogenes spp. are warranted to untangle species complexes as they provide a better assessment of interspecific genetic divergence. METHODS: Derogenes ruber is redescribed based on newly co

High genetic diversity in the Dirofilaria repens species complex revealed by mitochondrial genomes of feline microfilaria samples from Narathiwat, Thailand.

Dirofilaria repens is a zoonotic, mosquito-borne filaria infecting carnivores, particularly dogs. It is expanding its range in Europe but epidemiological information is sparse for other Eurasian regions. In Hong Kong and India, the closely related species Candidatus Dirofilaria hongkongensis was proposed. Previous analysis of 2.5 kb partial mitochondrial genome sequences containing the particularly variable non-coding control region revealed low diversity in European D. repens while Asian nematodes showed high diversity. Sequences derived from feline blood samples from Narathiwat (Thailand) le
