Prostate cancer is the second most common cancer in men, accounting for 14.1% of new cancer cases in 2020. The aggressiveness of prostate cancer is highly variable, depending on its grade and stage at the time of diagnosis. Despite recent advances in prostate cancer treatment, some patients still experience recurrence or even progression after undergoing radical treatment. Accurate initial staging and monitoring for recurrence determine patient management, which in turn affect patient prognosis and survival. Classical imaging has limitations in the diagnosis and treatment of prostate cancer, b

One of the major applications of the CRISPR-Cas9 system is the visualization of DNA by using nuclease-deactivated Cas9 (dCas9), which, following complexation with a single-guide RNA (sgRNA), specifically binds to target genomic sequences without inducing DNA breaks. In this approach, either dCas9 or the sgRNA is labeled with fluorescent proteins or dyes, or they are engineered to recruit such molecules. A key advantage of CRISPR-based imaging is that genomic elements in living cells can be tracked, because of the ability to express all system components in vivo. Although CRISPR-based imaging h

Globally, deep vein thrombosis (DVT) is one of the major preventable causes of mortality and morbidity. Due to the paucity of objective clinical examination findings and non-specific subjective complaints, optimal modalities for the diagnosis of DVT remain unclear. Two adhesion molecules that are engaged in thrombus formation and inflammation, P- and E-selectins, have shown a potential to serve as excellent biomarkers for the early detection of DVT. The review investigates the contribution of these selectins to thrombosis and their use as diagnostic biomarkers and molecular imaging agents. The

Innate immune signalling is uniquely driven through the formation of supramolecular organising centres (SMOCs), which couple receptor activation with downstream effector recruitment. The adaptor protein TRIF coordinates interferon, inflammatory, and cell death responses downstream of TLR3 and TLR4 activation, yet exactly how this protein drives such diverse outputs remains unresolved. Recent structural studies of recombinant TRIF, alongside single-molecule fluorescence imaging of endogenously expressed TRIF in live cells, reveal that TRIF assembles into complexes with heterogeneous size and mo

The incorporation of sp 3 -hybridized carbon centers into planar sp 2 carbon nanostructures is a powerful strategy in solution chemistry for tuning their electronic and structural properties. While on-surface synthesis has demonstrated efficient coupling strategies for sp 2 systems, C s p 3 ─ C s p 2 bond formation on surfaces remains largely unexplored. Here, we report the on-surface analogue of Friedel-Crafts alkylation on a metal surface: an intramolecular C s p 3 ─ C s p 2 coupling reaction based on a specifically designed fluorinated aromatic reactant, with the metal surfac

As typical compounds with aggregation induced emission (AIE) property, tetraphenylethylene-based photosensitizers with unique molecular structures and excellent photophysical properties have been widely applied in the bio-specific imaging, diagnosis, photodynamic therapy (PDT) and other related fields. In this work, we have developed a concise and efficient synthetic route to modify the structure of the AIE photosensitizer PS2, incorporating an aminomethyl side chain at the thiophene terminus. This modification offers a versatile platform for subsequent structural diversification. Following th

Microribonucleic acids (microRNAs, miRNAs) are important emerging biomarkers, yet their detection in clinical samples remains challenging due to their low concentrations and matrix effects, which typically require complex workflows including miRNA extraction, reverse transcription, and amplification. Here, we introduce a new approach to miRNA detection that combines a digital large-field-of-view surface plasmon resonance imaging platform capable of detecting individual gold nanoparticle labels, each associated with a single target miRNA molecule, with a molecularly triggered nanoparticle relea

Glioblastoma, the most aggressive primary brain tumor, carries a dismal prognosis, with median survival remaining under 15 months despite standard therapies. This is largely because of the tumor's infiltrative nature, the restrictive blood-brain barrier, and intratumoral heterogeneity. Chlorotoxin (CTX), a 36-amino acid peptide derived from scorpion venom, has emerged as a promising multifunctional agent with high specificity for neuroectodermal tumors. In this comprehensive review, we highlight CTX's potential to address critical limitations of current glioma treatments by bridging diagnostic

Bacterial biofilms are resilient multicellular communities that underlie persistent infections and environmental survival. Dispersal from biofilms is a pivotal event for transmission and pathogenesis, yet the host signals and bacterial mechanisms orchestrating this transition remain poorly understood. Here, we show that nitric oxide (NO), a ubiquitous host-derived signaling molecule, acts as a rapid trigger for biofilm dispersal in Vibrio cholerae, a highly motile gram-negative bacterium and the etiologic agent of cholera, by promoting the development of motility. NO exposure induces broad upr

Invasive lobular cancer (ILC) is the most common of the breast cancer special types, accounting for up to 15% of all breast malignancies. The distinctive biological features of ILC include the loss of the cell adhesion molecule E-cadherin, which drives the tumor's peculiar discohesive growth pattern, with cells arranged in single file and dispersed throughout the stroma. Typically, such tumors originate in the lobules, are more commonly bilateral compared to invasive ductal cancer (IDC) and require a more accurate diagnostic examination through imaging. They are luminal in molecular subtype, a

INTRODUCTION: Over 6.8 million fractures occur annually in the US, with 10% experiencing delayed- or non-union. Anabolic therapeutics like PTH analogs stimulate fracture repair, and small molecule salt inducible kinase (SIK) inhibitors mimic PTH action. This study tests whether the SIK inhibitor YKL-05-099 accelerates fracture callus osteogenesis.
METHODS: 126 female mice underwent femoral shaft pinning and midshaft fracture, receiving daily injections of PBS, YKL-05-099, or PTH. Callus tissues were analyzed via RT-qPCR, histology, single-cell RNA-seq, and μCT imaging. Biomechanical testing e

Artificial metallo-nucleases (AMNs) are promising DNA damaging drug candidates. Here, we demonstrate how the 1,2,3-triazole linker produced … ove with subsequent oxidative damage promoted through a superoxide- and peroxide-dependent pathway. Single molecule imaging of DNA isolated from peripheral blood mononuclear cells shows that the complex has comparable activity to the clinical drug temozolomide, causing DNA damage that is recognised by a combination of base excision repair (BER) enzymes.

Point spread function (PSF) engineering is an important technique to encode the properties (e.g., 3D positions, color, and orientation) of a single molecule in the shape of the PSF, often with the help of a programmable phase modulator. A deformable mirror (DM) is currently the most widely used phase modulator for fluorescence detection as it shows negligible photon loss. However, it relies on careful calibration for precise wavefront control. Therefore, design of an optimal PSF not only relies on the theoretical calculation of the maximum information content, but also the physical behavior of

Few genetically dominant mutations involved in human disease have been fully explained at the molecular level. In cases where the mutant gene encodes a transcription factor, the dominant-negative mode of action of the mutant protein is particularly poorly understood. Here, we studied the genome-wide mechanism underlying a dominant-negative form of the SOX18 transcription factor (SOX18RaOp) responsible for both the classical mouse mutant Ragged Opossum and the human genetic disorder Hypotrichosis-lymphedema-telangiectasia-renal defect syndrome. Combining three single-molecule imaging assays in

Abnormal interactions of copper (Cu) ions with amyloid-β (Aβ) peptides are believed to play an important role in the pathogenesis of Alzheimer's disease (AD). However, there is still debate as to the exact role of Cu ions in Aβ amyloidosis despite extensive studies on Aβ-Cu interactions. Unlike previously reported works, we herein study the effect of surface-chelated Cu2+, rather than the more usual solution-phase dissolved Cu2+, on Aβ aggregation. Through the combination of single molecule fluorescent tracking, atomic force microscopy imaging experiments, and all-atom molecular dynamic simula

Nanoparticles are now utilized in many diverse biological and medical applications. Despite this, it remains challenging to tailor their surface for specific molecular targeting while maintaining high biocompatibility. To address this problem, we evaluate a phytochelatin-related peptide surface coating to produce functional and biocompatible nanoparticles (NPs) based on fluorescent InP/ZnS and CdSe/ZnS or superparamagnetic FePt and Fe(3)O(4). Using a combination of transmission electron microscopy, size-exclusion chromatography and gel electrophoresis (GE), we demonstrate the excellent colloid

Toxoplasma gondii is a single-celled eukaryotic parasite with prolific invasion capability. The parasite uses an apical complex comprising proteinaceous structures and secretory organelles to efficiently enter host cells. As a result, the apical complex remains a vital structure of interest, with many studies dedicated to understanding its protein organization. One such protein is the motor myosin H (MyoH), which is indispensable for parasite motility and host-cell invasion. Given the small size of the complex, roughly a diffraction-limited volume in the visible, high-resolution techniques are

Respiratory viruses are a major public health burden across all age groups around the globe, and are associated with high morbidity and mortality rates. They can be transmitted by multiple routes, including physical contact or droplets and aerosols, resulting in efficient spreading within the human population. Investigations of the cell biology of virus replication are thus of utmost importance to gain a better understanding of virus-induced pathogenicity and the development of antiviral countermeasures. Light and fluorescence microscopy techniques have revolutionized investigations of the cel

The Ihara epileptic rat (IER) is a mutant model with limbic-like seizures whose pathology and causative gene remain elusive. In this report, via linkage analysis, we identified Down syndrome cell adhesion molecule-like 1(Dscaml1) as the responsible gene for IER. A single base mutation in Dscaml1 causes abnormal splicing, leading to lack of DSCAML1. IERs have enhanced seizure susceptibility and accelerated kindling establishment. Furthermore, GABAergic neurons are severely reduced in the entorhinal cortex (ECx) of these animals. Voltage-sensitive dye imaging that directly presents the excitatio

The First International Conference on Vascular Targeting focused on vascular targeting agents (VTAs) that occlude or destroy the pre-existing blood vessels of solid tumors. The VTAs cause a rapid shutdown in the blood supply to the tumor that kills tumor cells by depriving them of oxygen and nutrients. The VTAs are distinct from antiangiogenic agents, which prevent new blood vessel formation. Two major types of VTAs are being developed for cancer: the ligand-directed VTAs that use antibodies, peptides, and growth factors to deliver toxins, procoagulants, and proapoptotic effectors to tumor end
