Blood cyanide (HCN) and thiocyanate (SCN) concentrations were measured at intervals in anaesthetized dogs given bolus doses of sodium nitroprusside (SNP) 1 mg kg-1 or potassium cyanide 1.07 mg kg-1 and in animals infused with SNP 1.5 mg kg-1 for 1 h. Cyanide appeared rapidly in the red cells to give peak concentrations which accounted for more than 90% of the total blood HCN. Of thirty-five patients with various types of epilepsy treated with sodium valproate, 15 achieved complete seizure control on that drug alone, 12 other patients benefited and eight failed to improve on the drug. Excellent results were more likely in those with petit mal epilepsy and in those whose epilepsy was controlled with other drugs at the expense of side effects. The distribution of sodium salicylate was investigated in the fetuses of 18 mothers in the fifth or sixth month of pregnancy which had to be interrupted for medical reasons. Ninety-six min after drug administration, the salicylic acid levels in mother and fetus blood are almost equal. The highest drug levels in fetus tissue were found in the kidney, the lowest in the brain. In amniotic fluid, only traces of salicylic acid were estimated. A limited number of suitable cases made the exact statistical evaluation impossible.

An uncontrolled trial of sodium valproate in 25 severe epileptics uncontrollable by conventional antiepileptic drugs is presented. Excellent control was achieved in petit mal, myoclonic and minor motor seizures. No serious side effects were encountered, but hyperactivity may be aggravated and interaction with other anticonvulsants does occur.

The effect of sodium valproate, a drug which has been demonstrated to increase gamma-aminobutyric acid levels in the CNS, on tardive dyskinesia and psychiatric symptoms was investigated in a double-blind cross-over study on 32 chronic psychiatric patients. The oro-facial dyskinesias were totally or significantly relieved in 17 cases. During the active treatment period the involuntary movements of the extremities and dystonic spasms were also significantly relieved in 7 out of 9 patients. We have now investigated the effects of sodium periodate which is thought to oxidize the sialic acid of glycoproteins on cell surfaces and has been shown to affect the functions of other cells. In the dog, the emetic ED50 for sodium salicylate was 256+/-113 mg/kg for the intravenous route with a mean latency of 10.4+/-3.96 min, and for the oral route 228+/-68 mg/kg with a mean latency of 18.2+/-2.63 min.
