Detection of residues of chlorine-containing pesticides on tomato fruits by machine learning evaluation of laser-induced breakdown spectroscopy data.

In this pilot analytical study, we propose the machine learning evaluation of laser-induced breakdown spectroscopy (LIBS) data as a new, fast screening methodology for the detection of pesticides on tomato fruits. Five pesticides (acetamiprid, chlorpyrifos, λ-cyhalothrin, tebuconazole, tefluthrin) at three concentration levels down to 0.01 m/v% corresponding to their real application (spraying) concentrations were tested on fully ripe fruits of tomato (Solanum lycopersicum cv. Money maker). Direct (on the fruit peel) and indirect (sampling-based) analytical approaches were both tested. Linear …

A novel leucine-rich repeat receptor-like kinase gene in potato, StLRPK1, is involved in response to diverse stresses.

A potato gene, StLRPK1 (Solanum tuberosum L. leucine-rich-repeat receptor-like protein kinases 1), encoding a protein belonging to leucine-rich repeat receptor-like kinases (LRR-RLKs) was identified. It encodes 796 amino acids with 88% of identity to SRF3 of Arabidopsis thaliana and contains a signal peptide, five LRR motifs, a transmembrane domain, two proline-rich regions and a serine/threonine protein kinase domain. The transcripts were present at high levels in flowers and young leaves, while low in other tested organs. The mRNA of StLRPK1 was inducible in potato leaves by Phytophthora inf…

Screening of plants used by Southern African traditional healers in the treatment of dysmenorrhoea for prostaglandin-synthesis inhibitors and uterine relaxing activity.

Plants used by Southern African traditional healers for the treatment of menstrual pains were screened for prostaglandin-synthesis inhibitor … ctivities were obtained with ethanolic extracts of Siphonochilus aethiopicus, Cenchrus ciliaris and Solanum mauritianum. Generally ethanolic extracts gave higher activity than the aqueous extracts. None of the ethanolic plant extracts were able to relax or reduce the contractions of the precontracted guinea pig uterus.

Single-Tube Mass Spectrometry-Based Workflow for Multi-Omics Profiling of Diverse Biomolecules.

Advances in mass spectrometry-based omics technologies enable detailed exploration of biological processes. However, robust detection of multiple biomolecule types from the same biological sample remains challenging. We developed a multi-omics platform for the simultaneous detection of metabolites (polar and semi-polar), lipids, proteins, and native peptides, all detected in a high-throughput manner from a single tube. As a proof-of-concept for the platform, we profiled the profound and dynamic molecular changes occurring in tomato during fruit development. This included optimizing sample coll

Effects of Air Fryer Processing Conditions on Pesticide Residue Changes in Potatoes and Baby Bok Choy.

Air frying is increasingly used as a healthier cooking method, yet its effectiveness in reducing pesticide residues has not been investigated. Processing factor (PF) values ranged from 0.18-2.36 for potato strips, 0.06-2.30 for potato slices, and 0.37-2.13 for baby bok choy. For most pesticides, PF rose initially due to water loss, then fell with prolonged heating, except for volatile phoxim and dichlorvos, which declined continuously. Temperature (180-200 °C) had a significant effect on phoxim and dichlorvos, and the effectiveness depended on time. Removal efficiency also varied among vegetab

A novel Aureobasidium pullulans isolate for the biological control of Penicillium commune in potato decay and its optimization for enhanced antifungal activity using the Taguchi method.

Penicillium commune is a significant postharvest fungal pathogen responsible for blue mold rot in stored potatoes, resulting in substantial economic losses worldwide. In this study, we isolated and characterized a novel epiphytic strain of Aureobasidium pullulans SBAN-IAUF-1 from potato foliage and evaluated its antagonistic activity against Penicillium commune SBAN-IAUF-12. The new isolate exhibited strong inhibitory effects, reducing pathogen growth by 60% in vitro and 69.41% in vivo, outperforming values reported for most previously studied A. pullulans strains. Extracellular antifungal met

Genistein Mediates Beauveria bassiana-Induced Resistance Against Bemisia tabaci Reproduction in Tomato.

Endophytic colonization by entomopathogenic fungi Beauveria bassiana enhances plant resistance to herbivores; however, the underlying molecular mechanisms, particularly those involving hormonal disruption of pests, remain poorly understood. This study examined whether root inoculation of tomato with B. bassiana suppresses whitefly Bemisia tabaci fitness via flavonoid modulation. Metabolomic profiling of leaf tissues showed a 3.5-fold accumulation of the isoflavonoid genistein in B. bassiana-inoculated plants. KEGG enrichment analysis indicated significant upregulation of the isoflavonoid biosy

Role of Gut Bacteria in Tomato Leaf Miner Tuta absoluta Gaining Resistance to α-Tomatine by Converting It to Cholesterol.

Tuta absoluta is a worldwide invasive pest that causes severe losses in tomato industry in China, while its resistance mechanism to tomato's protective alkaloid of α-tomatine remains unclear. This study explores the role of its gut microbiota, and results showed that the abundance of the gut symbiont Enterococcus mundtii was significantly higher in T. absoluta feeding on wild-type tomatoes than that in those feeding on α-tomatine-deficient GAME2 mutant tomatoes. Isolated E. mundtii strain E14 degraded 72.2 ± 1.3% of α-tomatine in vitro; reinoculating it into antibiotic-treated T. absoluta rest

Hot-water immersion against Paracoccus marginatus: disinfestation efficacy on sprouting potato and effects on papaya fruit quality.

BACKGROUND: Paracoccus marginatus is a regulated, highly invasive pest that poses a significant risk to papaya production and international trade, necessitating effective phytosanitary disinfestation measures. Hot-water treatment (HWT) is widely recognized as a residue-free alternative to chemical fumigation; however, its operational schedules require alignment between pest thermal tolerance and fruit quality. In this study, the thermal tolerance of developmental stages of P. marginatus was quantified, and candidate HWT regimes were evaluated based on thermal tolerance determined on a surrogat

Moderate gamma irradiation enhances the biostimulant activity of humic acid in tomato: Integrated effects on growth, physiology, yield, and fruit quality.

Humic acid is widely used as a plant biostimulant, but its effectiveness depends strongly on its physicochemical characteristics and application rate. Gamma irradiation may modify the structure and biological activity of humic substances, although its effects on horticultural crops remain insufficiently characterized. This study evaluated the effects of non-irradiated and gamma-irradiated humic acid on the growth, physiological performance, mineral nutrition, antioxidant status, yield, and fruit quality of tomato (Solanum lycopersicum L.). The humic-acid treatments were arranged factorially us…

Friends or Foes? The Indispensable Role of Gut Microbiota in Plant-Fungus-Herbivore Interactions.

Plants interact complexly with herbivores and diverse microbial communities, both plant-associated and insect-associated. A recent study investigated how the endophytic fungus Trichoderma, colonizing tomato plants, affects the herbivore Spodoptera littoralis. The study revealed that Trichoderma-enhanced plant resistance significantly reduced larval performance by profoundly changing the insect's gut microbial community composition. This work demonstrates that the effects of plant fungal colonization on insect herbivores are critically mediated by the insect's gut microbiota, highlighting a key

Elucidating the modes of incorporation of the ferulic acid amides feruloyltyramine and feruloyloctopamine into the lignin-suberin fraction of potato periderms.

Ferulic acid amides are naturally present in the cell walls of potato (Solanum tuberosum) periderms. In this study, we investigated their modes of incorporation into the periderm cell wall polymers. A lignin/suberin-enriched fraction was isolated and analyzed by GPC, DFRC, and 2D-NMR. The analyses revealed that the lignin domain of this fraction was predominantly composed of G-lignin units, with an H:G:S ratio of 2:70:28 (S/G ratio of 0.40). More importantly, the data also indicated the presence of two ferulic acid amides, feruloyltyramine and feruloyloctopamine, that are incorporated into the…

Evolutionary genomics of three agricultural pest moths reveals rapid evolution of host adaptation and immune-related genes.

BACKGROUND: Understanding the genotype of pest species provides an important baseline for designing integrated pest management (IPM) strategies. Recently developed long-read sequence technologies make it possible to compare genomic features of nonmodel pest species to disclose the evolutionary path underlying the pest species profiles. Here we sequenced and assembled genomes for 3 agricultural pest gelechiid moths: Phthorimaea absoluta (tomato leafminer), Keiferia lycopersicella (tomato pinworm), and Scrobipalpa atriplicella (goosefoot groundling moth). We also compared genomes of tomato leafm

Potato starch/naringenin complexes for high-stability Pickering emulsions: Structure, properties, and emulsion stabilization mechanism.

In this study, potato starch (PS)/naringenin (NAR) complex was prepared, and its properties and emulsification behavior were evaluated. The experimental results demonstrated that NAR successfully formed a complex with PS molecules through hydrogen bonds and other non-covalent interactions. The emulsifying capacity (ROV) of PS/NAR complex with 16 % composite ratio was 0.9999, which was higher than PS (ROV = 0.3329) (p < 0.05). Based on particle property analysis and molecular dynamics simulation, the mechanism of improving the emulsification performance might be the action of the benzene ring o

Response of tomatoes to inactivated endophyte LSE01 under combined stress of high-temperature and drought.

Endophytes can assist crops in adapting to high temperatures and drought conditions, thereby reducing agricultural losses. However, the mechanism through which endophytes regulate crop resistance to high temperatures and drought stress remains unclear, and concerns regarding safety and stability exist with active endophytes. Thus, heat-treated endophytic bacteria LSE01 (HTB) were employed as a novel microbial fertilizer to investigate their effects on plant adaptation to high temperatures and drought conditions. The results indicated that the diameter and weight of tomatoes treated with HTB un

The predatory mite Pronematus ubiquitus curbs Aculops lycopersici damage under greenhouse conditions.

BACKGROUND: The tomato russet mite, Aculops lycopersici, is a major worldwide pest infesting tomato crops for which only few control methods are available. At present, no commercialized beneficial organism has proven to be an effective biological control agent of the pest. As there is a strong need to develop alternatives to synthetic insecticides, we assessed the efficacy of an iolinid mite, Pronematus ubiquitus, as a preventive method against A. lycopersici in comparison with a curative treatment in a replicated experiment in the greenhouse. RESULTS: After pre-establishment of P. ubiquitus

Micro-magnetic resonance imaging of ex vivo mouse embryos with potato starch suspension.

Potato starch suspension (PSS) holds promise as a solution to issues, such as air bubbles and specimen motion, associated with micro-magnetic resonance imaging (micro-MRI) of ex vivo embryos. Here, we present a protocol for using PSS when scanning specimens with micro-MRI. We describe steps for preparing samples and potato starch with phosphate-buffered saline. We then detail steps for specimen immersion and micro-MRI scanning. This protocol will enable micro-MRI of not only embryos but also other specimens, such as insects. For complete details on the use and execution of this protocol, pleas
