Pathogenic variants in USP7 cause a neurodevelopmental disorder with speech delays, altered behavior, and neurologic anomalies.

PURPOSE: Haploinsufficiency of USP7, located at chromosome 16p13.2, has recently been reported in seven individuals with neurodevelopmental phenotypes, including developmental delay/intellectual disability (DD/ID), autism spectrum disorder (ASD), seizures, and hypogonadism. Further, USP7 was identified to critically incorporate into the MAGEL2-USP7-TRIM27 (MUST), such that pathogenic variants in USP7 lead to altered endosomal F-actin polymerization and dysregulated protein recycling. METHODS: We report 16 newly identified individuals with heterozygous USP7 variants, identified by genome or ex

How Effectively Do People Remember Voice Disordered Speech? An Investigation of the Serial-Position Curve.

We examined how well typical adult listeners remember the speech of a person with a voice disorder (relative to that of a person without a voice disorder). Participants (n = 40) listened to two lists of words (one list uttered in a disordered voice and the other list uttered in a normal voice). After each list, participants completed a free recall test, in which they tried to remember as many words as they could. While the total number of words recalled did not differ between the disordered voice condition and the normal voice condition, an investigation of the serial-position curve revealed a

Metacognition in speech and language therapy for children with social (pragmatic) communication disorders: implications for a theory of therapy.

BACKGROUND: Metacognition is a significant component of complex interventions for children who have developmental language disorders. Research into how metacognition operates in the content or process of developmental language therapy delivery is limited. Identification and description of proposed active therapy components, such as metacognition, may contribute to our understanding of how to deliver complex communication interventions in an optimal manner. AIMS: To analyse aspects of metacognition during therapy derived from a manualized speech and language intervention (the Social Communicat

An exploratory study of the influence of load and practice on segmental and articulatory variability in children with speech sound disorders.

This exploratory treatment study used phonetic transcription and speech kinematics to examine changes in segmental and articulatory variability. Nine children, ages 4 to 8 years old, served as participants, including two with childhood apraxia of speech (CAS), five with speech sound disorder (SSD) and two who were typically developing. Children practised producing agent + action phrases in an imitation task (low linguistic load) and a retrieval task (high linguistic load) over five sessions. In the imitation task in session one, both participants with CAS showed high degrees of segmental and a

A multi-dimensional approach to gradient change in phonological acquisition: a case study of disordered speech development.

The purpose of the current study is to document phonological change from a multidimensional perspective for a 3-year-old boy with phonological disorder by comparing three measures: (1) accuracy of consonant productions, (2) dynamic assessment, and (3) acoustic analysis. The methods included collecting a sample of the targets /s, [symbol: see the text], t[symbol: see the text], and d[symbol: see the text]/ produced in single words and repeated over time. The samples were analysed using phonetic transcription and acoustic measures of duration, spectral mean, and spectral variance. A dynamic asse

Intragenic rearrangements in NRXN1 in three families with autism spectrum disorder, developmental delay, and speech delay.

NRXN1 is highly expressed in brain and has been shown recently to be associated with ASD, schizophrenia, cognitive and behavioral abnormalities, and alcohol and nicotine dependence. We present three families, in whom we identified intragenic rearrangements within NRXN1 using a clinical targeted oligonucleotide array CGH. An approximately 380 kb deletion was identified in a woman with Asperger syndrome, anxiety, and depression and in all four of her children affected with autism, anxiety, developmental delay, and speech delay but not in an unaffected child. An approximately 180 kb tandem duplic

Speech-sound memory consolidation over a daytime nap in adults with and without developmental language disorder.

INTRODUCTION: Sleep plays a critical role in memory consolidation, yet its contribution to the formation of perceptual-linguistic representations remains only partially understood. In the current study, we examined sleep spindles and slow oscillations (SOs) in relation to offline changes in speech-sound memory in adults across a wide range of language ability. METHODS: Twenty-nine monolingual English-speaking participants completed a perceptual training task contrasting retroflex /ɖ/ and dental /d̪/ consonants, followed by a polysomnography-monitored nap. RESULTS: Slow spindle density was po

What Is in a Label? The Importance of Clinical Specificity and Challenges of Diagnostic Evolution in Progressive Speech-Language Disorders.

PURPOSE: This viewpoint addresses the complexity of the nomenclature of progressive speech-language disorders, which often exists at the intersection of descriptive symptoms, clinical syndromes, and underlying neuropathology. Using primary progressive apraxia of speech (PPAOS) as an example, we explore how overlapping diagnostic frameworks employed by speech-language pathologists, neurologists, and other clinicians and researchers can lead to different diagnostic labels that may evolve throughout disease progression. These discrepancies and changes can complicate patient understanding, clinica

Disorders of the Basal Ganglia and Cerebellum: A Comprehensive Review of Speech Sensorimotor Symptoms and Responses to Feedback Perturbations.

Situated within critical cortical-subcortical sensorimotor circuits, the basal ganglia and cerebellum are thought to play important roles in both feedforward and feedback mechanisms of speech sensorimotor control. Motor speech impairments and speech-related sensory deficits are indeed common in disorders involving these two structures such as Parkinson's disease, dystonia, and cerebellar degeneration. Recently, several studies have also investigated the impact of these disorders on the feedforward and feedback components of speech sensorimotor control by examining immediate compensation to une

Exposure to online hate speech is positively associated with post-traumatic stress disorder symptom severity.

Post-traumatic stress disorder (PTSD) after traumatic events is prevalent and can lead to negative consequences. While social media use has been associated with PTSD, little is known about the specific association of online hate speech on social media networks and PTSD, and whether such association is stronger among those with difficulties in emotion regulation, who may have a harder time coping with hate speech. In a general population sample of Jewish adults (aged 18-70) in Israel (N = 3,998), assessed about two months after the wide-scale terror attacks of October 7, 2023, regression analys

Communicative activity and participation in children with speech sound disorder: a case series using the ICF framework.

The everyday lives of young children with speech sound disorder (SSD) are not well described, with few studies taking a holistic view with interaction in focus. The aim of this study was to investigate how SSD affects communicative activity and participation in everyday life of four children in a Swedish context. The study involved 4 triads: 4 children with SSD aged 4 to 7, their parents (n = 4), grandmothers (n = 2) and pedagogues, i.e. preschool teachers (n = 2). The research design was a case series, including speech language tests, questionnaires and structured interviews. The Internationa

Acquisition of Speech Prosody in a Non-native Tone Language by Children With and Without Autism Spectrum Disorder.

PURPOSE: Children with autism spectrum disorder (ASD) often show abnormal speech prosody. Tonal languages can pose more difficulties as speakers need to use acoustic cues to make lexical contrasts while encoding the focal function, but the acquisition of speech prosody of non-native languages, especially tonal languages has rarely been investigated. METHODS: This study aims to fill in the aforementioned gap by studying prosodic focus-marking in Mandarin by native Cantonese-speaking children with ASD (n = 25), in comparison with their typically developing (TD) peers (n = 20) and native Mandari

Acoustic features from speech as markers of depressive and manic symptoms in bipolar disorder: A prospective study.

INTRODUCTION: Voice features could be a sensitive marker of affective state in bipolar disorder (BD). Smartphone apps offer an excellent opportunity to collect voice data in the natural setting and become a useful tool in phase prediction in BD. AIMS OF THE STUDY: We investigate the relations between the symptoms of BD, evaluated by psychiatrists, and patients' voice characteristics. A smartphone app extracted acoustic parameters from the daily phone calls of n = 51 patients. We show how the prosodic, spectral, and voice quality features correlate with clinically assessed affective states and

Whole Genome Analysis in Consanguineous Families Reveals New Loci for Speech Sound Disorder (SSD).

Speech is the most common means of communication in humans. Any defect in accurate speech production ability results in the development of speech sound disorder (SSD), a condition that can significantly impair an individual's academic performance, social interactions, and relationships with peers and adults. This study investigated the genetic basis of SSD in three Pakistani families. We performed family-based genome-wide parametric linkage analysis and homozygosity mapping in three consanguineous families with SSD from the Punjab province of Pakistan. The Test for Assessment of Articulation a

Genetic aetiologies for childhood speech disorder: novel pathways co-expressed during brain development.

Childhood apraxia of speech (CAS), the prototypic severe childhood speech disorder, is characterized by motor programming and planning deficits. Genetic factors make substantive contributions to CAS aetiology, with a monogenic pathogenic variant identified in a third of cases, implicating around 20 single genes to date. Here we aimed to identify molecular causation in 70 unrelated probands ascertained with CAS. We performed trio genome sequencing. Our bioinformatic analysis examined single nucleotide, indel, copy number, structural and short tandem repeat variants. We prioritised appropriate v

CDK13-related disorder: a deep characterization of speech and language abilities and addition of 33 novel cases.

Speech and language impairments are central features of CDK13-related disorder. While pathogenic CDK13 variants have been associated with childhood apraxia of speech (CAS), a systematic characterisation of communication has not been conducted. Here we examined speech, language, non-verbal communication skills, social behaviour and health and development in 41 individuals with CDK13-related disorder from 10 countries (male = 22, median-age 7 years 1 month, range 1-25 years; 33 novel). Most participants used augmentative and alternative communication (AAC) in early childhood (24/41). CAS was com

ZNF142 mutation causes neurodevelopmental disorder with speech impairment and seizures: Novel variants and literature review.

The ZNF142 gene on chromosome 2q35 contains ten exons and encodes a zinc finger protein 142 with 31 C2H2-type zinc fingers domain. Pathogenic variants in ZNF142 result in an autosomal recessive neurodevelopmental disorder with impaired speech and developmental delay. Here, we report two novel variants (NM_001105537: c.25C > T/c.1741C > T, p.Gln9*/p.Arg581Cys) in ZNF142 in an Iranian family identified by Whole-Exome sequencing and confirmed by Sanger sequencing. These variants are categorized as "pathogenic" and "variant of unknown significance" based on the standards for the interpretation of

The Association of Dyslexia and Developmental Speech and Language Disorder Candidate Genes with Reading and Language Abilities in Adults.

Reading and language abilities are critical for educational achievement and success in adulthood. Variation in these traits is highly heritable, but the underlying genetic architecture is largely undiscovered. Genetic studies of reading and language skills traditionally focus on children with developmental disorders; however, much larger unselected adult samples are available, increasing power to identify associations with specific genetic variants of small effect size. We introduce an Australian adult population cohort (41.7-73.2 years of age, N = 1505) in which we obtained data using validat

Aminoglycoside-associated nonsyndromic deafness and speech disorder in mitochondrial A1555G mutation in a family: A case report.

RATIONALE: Mitochondrial DNA mutations have been associated with many maternal inherited diseases. A1555G mutation in mtDNA effects the gene code for rRNA, resulting in the structural change of human ribosome rending it susceptible to binding of the common antibiotic, aminoglycosides. Such mutation has linked with non-syndromic hearing loss and is one of the most common mtDNA mutations in Asian populations. PATIENT CONCERNS: A 50-year-old Taiwanese female visited our neurology department with concern for multiple members with hearing loss in her family, including herself. DIAGNOSES: Physical

The DYX2 locus and neurochemical signaling genes contribute to speech sound disorder and related neurocognitive domains.

A major milestone of child development is the acquisition and use of speech and language. Communication disorders, including speech sound disorder (SSD), can impair a child's academic, social and behavioral development. Speech sound disorder is a complex, polygenic trait with a substantial genetic component. However, specific genes that contribute to SSD remain largely unknown. To identify associated genes, we assessed the association of the DYX2 dyslexia risk locus and markers in neurochemical signaling genes (e.g., nicotinic and dopaminergic) with SSD and related endophenotypes. We first per
