Male Infertility and Oxidative Stress: A Focus on the Underlying Mechanisms.

Reactive oxygen species (ROS) play a critical role in defining the functional competence of human spermatozoa. When generated in moderate amounts, ROS promote sperm capacitation by facilitating cholesterol efflux from the plasma membrane, enhancing cAMP generation, inducing cytoplasmic alkalinization, increasing intracellular calcium levels, and stimulating the protein phosphorylation events that drive the attainment of a capacitated state. However, when ROS generation is excessive and/or the antioxidant defences of the reproductive system are compromised, a state of oxidative stress may be in…

Distribution, crypticity, stability, and localization of α-L-fucosidase of mouse cauda epididymal sperm.

Sperm-associated and semen-specific isoforms of α-L-fucosidase are thought to function in fertilization in numerous organisms. Here, we report the localization, distribution, crypticity, and stability of this enzyme in mouse cauda epididymal sperm and cauda fluid. Western analysis revealed that the sperm-associated α-L-fucosidase is present as two isoforms (Mr ∼49 and 56 kDa), whereas the cauda fluid α-L-fucosidase shows a single band at 50 kDa. α-L-Fucosidase activity was detected using the fluorogenic substrate 4-MU-FUC. Of the total α-L-fucosidase activity recovered in the cauda epididymal

The addition of calcium ion chelator, EGTA to thawing solution improves fertilizing ability in frozen-thawed boar sperm.

In our previous study, seminal plasma effectively suppressed the induction of sperm to capacitation-like status and acrosome loss during the thawing process. However, because boar seminal plasma is contaminated with various kinds of bacteria and/or viruses, it is necessary to develop a thawing solution without animal-derived materials. In this study, we focused on the increase of intracellular Ca(2+) ([Ca(2+)](i)) in sperm after thawing and the negative effects of sperm qualities. After thawing, the fluorescent intensity of [Ca(2+)](i) indicator, Fluo-3/AM, and the level of phosphorylated tyro

SPACA4 regulates the structure and molecular basis of spermatid maturation and ultimately affects sperm quality in mice.

Sperm quality is crucial for sperm function and can even affect embryo quality and offspring health. Spermatid maturation is extremely compl … n and affects sperm quality in mice. Abnormal sperm quality in Spaca4 -/- mice results in decreased sperm capacitation and a decreased acrosome response, ultimately affecting the fertility of male mice.

The anatomy, movement, and functions of human sperm tail: an evolving mystery.

Sperms have attracted attention of many researchers since it was discovered by Antonie van Leeuwenhoek in 1677. Though a small cell, its every part has complex structure and different function to play in carrying life. Sperm tail is most complicated structure with more than 1000 proteins involved in its functioning. With the advent of three-dimensional microscopes, many studies are undergoing to understand exact mechanism of sperm tail movement. Most recent studies have shown that sperms move by spinning rather than swimming. Each subunit of tail, including axonemal, peri-axonemal structures,

Assessments of sperm quality integrating morphology, swimming patterns, bioenergetics and cell signalling.

Spermatozoa are diverse in form and function and these differences impact on their fertilizing capacity. Because of considerable inter-male and inter-species differences in sperm traits, assessments of sperm quality demand that we consider variations at different levels. We should thus pay attention not only to average values but also intra- and inter-sperm population variations and subpopulation structure. Sperm shape and size evolve in reponse to postcopulatory sexual selection. Assessments of morphological variation, with conventional microscopy or with computer-assisted systems, should bea

Cholesterol-Loaded Cyclodextrin Increases the Cholesterol Content of Goat Sperm to Improve Cold and Osmotic Resistance and Maintain Sperm Function after Cryopreservation.

The success of semen cryopreservation depends on sperm membrane integrity and function after thawing. Cholesterol-loaded cyclodextrin (CLC) is used for in vitro incorporation of cholesterol to protect cells against cold temperatures. We hypothesized that CLC treatment also enhances sperm cholesterol content to increase tolerance to osmotic shock and cryoresistance, thereby improving fertility. We confirmed the fact that treatment of goat semen with 3 mg/ml CLC increases sperm cholesterol content using both the Liebermann-Burchard approach and filipin III labeling of membrane cholesterol. Sperm

Extracellular vesicles from different regions of the female reproductive tract promote spermatozoa motility and support capacitation.

Extracellular vesicles (EVs) have been proven to have a crucial role in intercellular communication and have attracted significant attention in the physiology of reproduction because of their multiple functions in physiological processes essential for reproduction including gametogenesis, fertilization and embryo-endometrial cross-talk. Although EVs from the male reproductive tract have been extensively studied for their role in sperm maturation, research on female reproductive tract-derived EVs in humans is still emerging and supported by only a few studies to date. In vitro study was perform

The role of Mitogen activated protein kinase (MAPK) in sperm functions.

The generation of mature spermatozoa in the epididymis includes the activation of the MAPK cascade in a complex manner. MAPKs are thought to be involved in the regulation of transcription and ectoplasmic specialization (ES) in the testis. MAPKs also regulate mature spermatozoa flagellar motility, hyperactivation and the acrosome reaction. Here we review the current data regarding the functions of MAPKs in spermatogenesis and in mature spermatozoa.

[In vitro assessment of semen for the prediction of fertility].

The prediction of fertility is a primary goal in the field of reproductive medicine. The aim of the present paper is to describe the value of conventional and modern sperm analysis systems considering the process of fertilization. The classical assessment of motility and morphology enables the rough estimation of semen quality in order to select ejaculates for the use in artificial insemination. Recent methods for sperm diagnosis, such as fluorescent marking for the detection of sperm plasma membrane integrity, the hypoosmotic swelling test, and computer assisted semen analysis allow for the e

Increased reproductive outcomes after optimized sperm preparation.

A key factor to the success of in vitro fertilization (IVF) is the preparation of human sperm, a critical step that directly impacts the efficacy of the procedure. This proof-of-concept study evaluated the effect of HyperSperm, a novel sperm preparation technique designed to enhance sperm function, on fertilization, embryo development, and pregnancy outcomes in both a mouse model and a first-in-human trial following IVF. In mice, HyperSperm treatment significantly increased hyperactivated motility (p < 0.05), leading to improved fertilization and blastocyst development (p < 0.05), as well as h

TMEM232 is required for the formation of sperm flagellum and male fertility in mice.

Asthenoteratozoospermia is a major cause of male infertility. Thus far, the identified related genes can explain only a small share of asthe … l sheath. Proteomic analysis revealed altered expression of proteins related to flagellar motility, sperm capacitation, the integrity and stability of sperm structure, especially an upregulated expression of multiple ribosome components in TMEM232-deficient spermatids. Additionally, TMEM232 was observed to be involved in autophagy by interacting with autophagy-relat …

CRISPR-Cas9-mediated mutation revealed BSPH2 protein is dispensable for male fertility.

Members of the Binder of SPerm (BSP) superfamily have been identified in both human and mouse epididymis. These proteins are known to bind sperm membrane and promote sperm capacitation. Studies suggest that BSPH2 might play a different role in sperm functions from its counterparts; however, the role of BSPH2 remains mainly unexplored. To investigate whether the absence of one member of the BSP family could affect fertility, mice lacking Bsph2 expression were generated using clustered regularly interspaced short palindromic repeats (CRISPR) associated 9 (Cas9) technology. Knockout (KO) male mic…

Uterine flushings from women treated with levonorgestrel affect sperm functionality in vitro.

Levonorgestrel (LNG), a synthetic 19 nor-testosterone derivative, is widely used for emergency contraception. It is well known that LNG prev … ified GdA significantly decreased sperm-ZP binding. The data were compatible with changes affecting sperm capacitation, motility and interaction with the ZP. These results may offer evidence on additional mechanisms of action of LNG as an emergency contraceptive.

Fallopian tubes influences sperm selection and fertilization success.

This study investigates the proteomic profile of the fallopian tube following exposure to human sperm, with a focus on its role in sperm capacitation, final sperm maturation, successful fertilization, and early embryonic development. Twenty reproductive-age women undergoing hysterectomy during the luteal phase were divided into two groups. The control group was instructed to abstain from intercourse for at least one week prior to surgery. The case group was instructed to have intercourse 24 to 48 h before surgery, ensuring intravaginal ejaculation. A 1 cm segment of the ampullary region of the…

Spermatogenic cell-specific type 1 hexokinase (HK1S) is essential for capacitation-associated increase in tyrosine phosphorylation and male fertility in mice.

Hexokinase (HK) catalyzes the first irreversible rate-limiting step in glycolysis that converts glucose to glucose-6-phosphate. HK1 is ubiquitously expressed in the brain, erythrocytes, and other tissues where glycolysis serves as the major source of ATP production. Spermatogenic cell-specific type 1 hexokinase (HK1S) is expressed in sperm but its physiological role in male mice is still unknown. In this study, we generate Hk1s knockout mice using the CRISPR/Cas9 system to study the gene function in vivo. Hk1s mRNA is exclusively expressed in testes starting from postnatal day 18 and continuin

Seminal Plasma and Extracellular Vesicles as Molecular Gatekeepers: Oxidative Stress, Endocrine Crosstalk, and Biomarker Discovery in Male Infertility.

Conventional semen analysis fails to capture the molecular determinants underlying impaired reproductive function. Emerging evidence positions seminal plasma (SP) and extracellular vesicles (EVs) as dynamic regulators of sperm physiology, rather than passive transport components. SP, enriched with proteins, metabolites, hormones, and antioxidants, modulates sperm motility, capacitation, acrosome reaction, and immune tolerance. Complementarily, EVs, including prostasomes, epididymosomes, and testicular vesicles, deliver proteins, lipids, and small RNAs that remodel sperm membranes, protect agai

Membrane Potential Determined by Flow Cytometry Predicts Fertilizing Ability of Human Sperm.

Infertility affects 10 to 15% of couples worldwide, with a male factor contributing up to 50% of these cases. The primary tool for diagnosing male infertility is traditional semen analysis, which reveals sperm concentration, morphology, and motility. However, 25% of infertile men are diagnosed as normozoospermic, meaning that, in many cases, normal-appearing sperm fail to fertilize an egg. Thus, new information regarding the mechanisms by which sperm acquire fertilizing ability is needed to develop a clinically feasible test that can predict sperm function failure. An important feature of sper
