Regulated storage and subsequent transformation of spermatozoa in the fallopian tubes of an Australian marsupial, Sminthopsis crassicaudata.

The disposition of spermatozoa in the female tract of the dasyurid marsupial, Sminthopsis crassicaudata, was examined before and after ovulation, by transillumination using differential interference optics. At 6-30 h after mating in females with maturing follicles, spermatozoa were restricted to special storage crypts that define the isthmus of the Fallopian tube. These spermatozoa were mostly immotile and were spear shaped, except for 50-200 vanguard spermatozoa within anterior crypts that were T shaped due to a rotation of the head on the tail. After ovulation and arrival of eggs in the narr

Novel role of proton-secreting epithelial cells in sperm maturation and mucosal immunity.

Epithelial cells are immune sensors and mediators that constitute the first line of defense against infections. Using the epididymis, a model for studying tubular organs, we uncovered a novel and unexpected role for professional proton-secreting 'clear cells' in sperm maturation and immune defense. The epididymal epithelium participates in the maturation of spermatozoa via the establishment of an acidic milieu and transfer of proteins to sperm cells, a poorly characterized process. We show that proton-secreting clear cells express mRNA transcripts and proteins that are acquired by maturing spe

Star1 gene mutation reveals the essentiality of 11-ketotestosterone and glucocorticoids for male fertility in Nile Tilapia (Oreochromis niloticus).

Steroidogenic acute regulatory protein (Star) plays an essential role in the biosynthesis of corticosteroids and sex steroids by mediating the transport of cholesterol from the outer to the inner membrane of mitochondria. Two duplicated Star genes, namely star1 and star2, have been identified in non-mammalian vertebrates. To investigate the roles of star genes in fish steriodogenesis, we generated two mutation lines of star1-/- and star1-/-/star2-/- in Nile tilapia (Oreochromis niloticus). Previous studies revealed that deficiency of star2 gene caused delayed spermatogenesis, sperm apoptosis a

Deficiency in the Rab25 gene leads to a decline in male fertility and testicular injury: Impact on the regulation of germ cell proliferation and apoptosis.

BACKGROUND: Rab25 is a member of the Rab family, functioning as a regulatory molecule in intracellular transport. Although its involvement in cellular functions and disease development is well-established, its precise roles in male reproductive physiology remain elusive. METHODS: To explore the specific roles of Rab25 in testicular development and spermatogenesis, we established the Rab25-/- mouse model and Rab25 knockdown germ cell line (GC-2). We compared the fertility, sperm analysis, and testicular tissues between Rab25-/- and wild-type male mice. To delve deeper into potential mechanisms

Actin-related protein ACTL7B ablation leads to OAT with multiple morphological abnormalities of the flagellum and male infertility in mice†.

The formation of fertilisation-competent sperm requires spermatid morphogenesis (spermiogenesis), a poorly understood program that involves complex coordinated restructuring and specialised cytoskeletal structures. A major class of cytoskeletal regulators are the actin-related proteins (ARPs), which include conventional actin variants, and related proteins that play essential roles in complexes regulating actin dynamics, intracellular transport, and chromatin remodeling. Multiple testis-specific ARPs are well conserved among mammals, but their functional roles are unknown. One of these is acti

Extenders for alpaca epididymal spermatozoa: Comparison of INRA96 and andromed.

Artificial insemination would be a useful technique for alpaca breeders to use as an aid to breeding to increase fleece quality. The technique, however, is not well developed in alpacas, partly because of the viscous nature of their seminal plasma. Castration conducted for husbandry purposes can provide a source of epididymal spermatozoa to test semen extenders or handling regimens, thus circumventing the problem of the viscous ejaculate. In this experiment, two semen extenders (Andromed and INRA96) developed for other species (bovine and equine, respectively) were tested with alpaca spermatoz

Null mutation of the transcription factor inhibitor of DNA binding 3 (id3) affects spermatozoal motility parameters and epididymal gene expression in mice.

ID3 is a transcription factor that acts as a dominant-negative regulator of other transcription factors by sequestering them, thus rendering them unable to bind DNA. We have shown previously that ID3 is expressed in a unique, region-specific manner along the epididymis, a highly specialized tissue of the male reproductive tract that functions in the transport and maturation of spermatozoa. The goal of these studies was to test the hypothesis that ID3 plays a role in the epididymis in the region-specific regulation of gene expression that is responsible for establishing the microenvironment req

Morphology of the urogenital papilla of the male marine teleost Black Rockfish, Sebastes schlegelii (Hilgendorf, 1880), and its role in internal fertilization.

There are few detailed descriptions of the morphology of the male external genitalia, the urogenital papilla (UGP), of the Black Rockfish (Sebastes schlegelii Hilgendorf, 1880). The purpose of this study was to evaluate this organ histologically and to determine the time of development of the UGP in Black Rockfish. Twelve adult males, three adult females and around 500 juveniles were used in the experiment. The juveniles were divided into normal developmental and androgen groups. The androgen group was exposed to methyltestosterone (100 μg/L) for 2 h daily for 38 days. Samples (N = 10 per samp

A review of sperm cryopreservation in the domestic dog and cat: part I, from science to clinic.

Semen cryopreservation is widely applied in the breeding of several domestic animal species. In the domestic dog, artificial insemination with cryopreserved semen is now routinely performed, whereas in the domestic cat the technique is more challenging. Achieving acceptable pregnancy rates requires high post-thaw semen quality. Unfortunately, sperm cells are susceptible to damage caused by temperature reduction. At temperatures below - 130 °C harmful reactions that can damage spermatozoa are essentially halted. Therefore, spermatozoa can be stored in liquid nitrogen at -196 °C for virtually un

Gene-knockout by iSTOP enables rapid reproductive disease modeling and phenotyping in germ cells of the founder generation.

Cytosine base editing achieves C•G-to-T•A substitutions and can convert four codons (CAA/CAG/CGA/TGG) into STOP-codons (induction of STOP-codons, iSTOP) to knock out genes with reduced mosaicism. iSTOP enables direct phenotyping in founders' somatic cells, but it remains unknown whether this works in founders' germ cells so as to rapidly reveal novel genes for fertility. Here, we initially establish that iSTOP in mouse zygotes enables functional characterization of known genes in founders' germ cells: Cfap43-iSTOP male founders manifest expected sperm features resembling human "multiple morpho

Protective Role of Spirulina platensis Against Bifenthrin-Induced Reprotoxicity in Adult Male Mice by Reversing Expression of Altered Histological, Biochemical, and Molecular Markers Including MicroRNAs.

: The potential reprotoxicity of bifenthrin remains unclear if only the common clinical indicators of reproductive disease are examined. The present study aimed to investigate the efficacy of Spirulina platensis, a microalga rich in antioxidant compounds, against bifenthrin-induced testicular oxidative damage in male mice. At the first, we demonstrate that administration of bifenthrin resulted in a decline of testosterone level and in deterioration of sperm quality that was correlated with significant transcription changes of some specific mRNA and microRNA involved in cholesterol transport, t

Sperm migration and selection in the reproductive tract of female mice is mostly affected by male genotype.

The aim of the present study is to assess the influence of male and female genotypes on the transport of sperm to the site of fertilization. We mated B10.BR, B10.BR-Ydel and BALB/c males with B10.BR and BALB/c females and then analyzed the quality and quantity of spermatozoa found five hours post coitus in three successive parts of the female reproductive tract. We found that B10.BR and B10.BR-Ydel spermatozoa are very effectively selected by the uterotubaljunction and other barriers of the female genital tract. On the contrary, severely deformed BALB/c spermatozoa appeared to be able to cross

The importance and potential of artificial insemination in CANDES (companion animals, non-domestic, endangered species).

Artificial insemination (AI) is the least invasive assisted reproductive technology, and is therefore of great interest to breeders of companion animals, non-domestic, and endangered species (CANDES). This most fundamental artificial breeding technique circumvents physical or behavioral impediments to natural mating and provides the means for genetic exchange between populations without transfer of live animals. In addition, because oocytes grow, mature and are fertilized in vivo and embryos are not subjected to in vitro culture conditions, AI eliminates the epigenetic effects on the female ga

HE1/NPC2 status in human reproductive tract and ejaculated spermatozoa: consequence of vasectomy.

We have previously demonstrated that the amount of HE1/NPC2 mRNA and protein expressed in the human epididymis is decreased under vasectomy. In this study, western blot analyses showed that many vasovasostomized men are characterized by high HE1/NPC2 levels in spermatozoa when compared with fertile donors. HE1/NPC2 association with sperm from vasovasostomized men was not related to low motility per se as spermatozoa from asthenospermic men have HE1/NPC2 levels similar to those in normal fertile semen samples. Spermatozoa from vasovasostomized men with high amount of HE1/NPC2 are characterized

The complex relationships between cystic fibrosis and congenital bilateral absence of the vas deferens: clinical, electrophysiological and genetic data.

Congenital bilateral absence of the vas deferens (CBAVD) is found in 1-2% of infertile males and in most male cystic fibrosis (CF) patients. CF and some of the CBAVD cases were found to share the same genetic background. In this study, 21 males with CBAVD had extensive physical and laboratory testing for symptoms of CF. Possible defective cellular chloride transport was measured by interstitial current measurement of rectal suction biopsies. Cystic fibrosis transmembrane conductance regulator (CFTR) gene mutation analysis was performed for 10 common CFTR mutations. CF-related symptoms were fou

Differential molecular mechanisms of heparin, BSA, and Ca2+-Induced boar sperm capacitation revealed by transcriptomics.

Sperm capacitation is a prerequisite for successful fertilization. In vitro, this process can be induced by various agents such as heparin, bovine serum albumin (BSA), and calcium ions (Ca2+), yet their underlying regulatory mechanisms remain incompletely understood. This study employed RNA sequencing (RNA-seq) to comparatively investigate the transcriptomic profiles and regulatory mechanisms during sperm capacitation induced by heparin, BSA, or Ca2+. Phenotypic analysis confirmed that all three inducers significantly promoted capacitation, albeit with differential effects on motility paramete

Effect of conditioned medium from equine Wharton's jelly mesenchymal stem/stromal cells on cool-stored stallion semen.

Artificial insemination in horses largely relies on cooled transported semen, but stallion spermatozoa are highly sensitive to cold stress, limiting storage duration. Mesenchymal stem/stromal cell-derived conditioned medium (CM) has shown protective effects on sperm in other species, yet its impact on equine semen remains unexplored. This study investigated the effects of equine Wharton's jelly-derived MSC-CM (eWJ-MSC-CM), on the quality of cooled stallion semen. Semen from five stallions of proven fertility was extended in INRA96 supplemented with 0, 5, or 15% eWJ-MSC-CM at 5 °C for 0, 24 and

Zinc alleviates cadmium-induced reproductive toxicity via regulating ion homeostasis, metallothionein expression, and inhibiting mitochondria-mediated apoptosis in the freshwater crab Sinopotamon henanense.

Cadmium (Cd) is a carcinogenic environmental pollutant that harms male reproductive systems by lowering sperm quality, impairing spermatogenesis, and causing apoptosis. Although zinc (Zn) has been reported to alleviate Cd toxicity, the underlying mechanisms have not been fully elucidated. The aim of this work was to investigate the mitigating effects of Zn on Cd-induced male reproductive toxicity in the freshwater crab Sinopotamon henanense. Cd exposure not only resulted in its accumulation but also in Zn deficiency, decreased sperm survival rate, poor sperm quality, altered ultrastructure, an

LLG2/3 Are Co-receptors in BUPS/ANX-RALF Signaling to Regulate Arabidopsis Pollen Tube Integrity.

In angiosperms, two sperm cells are transported and delivered by the pollen tube to the ovule to achieve double fertilization. Extensive communication takes place between the pollen tube and the female tissues until the sperm cell cargo is ultimately released. During this process, a pollen tube surface-located receptor complex composed of ANXUR1/2 (ANX1/2) and Buddha's Paper Seal 1/2 (BUPS1/2) was reported to control the maintenance of pollen tube integrity by perceiving the autocrine peptide ligands rapid alkalinization factor 4 and 19 (RALF4/19). It was further hypothesized that pollen-tube
