Nanoengineered extrusion-aligned tract bioprinting enables functional repair of spinal cord injuries.

Spinal cord repair demands biomaterials that replicate the aligned axonal architecture and mechanical softness of native tissue. However, most current scaffolds fail to support three-dimensional alignment and neuronal differentiation of human neural stem cells (hNSCs) in hydrated, low-stiffness environments. Here, we present NEAT (nanoengineered extrusion-aligned tract), a shear-stress-driven 3D bioprinting strategy that utilizes norbornene-functionalized collagen (NorCol) to generate highly aligned, mechanically stable hydrogels without post-processing. NEAT preserves the native triple-helica

A Review of the Potential Use of Antioxidants in Spinal Cord Injuries.

Spinal cord injury (SCI) is a debilitating neurological condition marked by primary mechanical damage followed by a complex secondary injury cascade, in which oxidative stress plays a central role. Mitochondrial dysfunction, ionic imbalance, and inflammatory responses drive excessive generation of reactive oxygen and nitrogen species, leading to lipid peroxidation, protein and DNA damage, apoptosis, and progressive neurological impairment. Antioxidant-based therapies have emerged as promising neuroprotective strategies, with compounds such as A91 peptide, curcumin, edaravone, ginsenosides, and

Numerical and Experimental Assessment of Poly-Pyrrole Used in Spinal Cord Injuries.

Some common conductive polymers are polyfuran, polyacetylene, polythiophene, and poly-pyrrole. Since their discovery, many researchers have been exploring and evaluating their conductive and electronic properties. Various applications have been developed for conductive materials. Their biocompatibility offers a new alternative for studying and solving complex problems, such as cellular activity, or, more recently, for use as neural implants and as an alternative to spinal cord regenerative tissue. This is particularly true for the use of poly pyrrole. The main obstacle lies in estimating some

Imaging Demyelinated Axons After Spinal Cord Injuries with PET Tracer [18F]3F4AP.

Spinal cord injuries (SCIs) often lead to lifelong disability. Among the various types of injuries, incomplete and discomplete injuries, where some axons remain intact, offer potential for recovery. However, demyelination of these spared axons can worsen disability. Demyelination is a reversible phenomenon, and drugs such as 4-aminopyridine (4AP), which target K+ channels in demyelinated axons, show that conduction can be restored. Yet, accurately assessing and monitoring demyelination after SCI remains challenging because of the lack of suitable imaging methods. In this study, we introduce a …

Increases of peripheral blood pressure in men with cervical spinal cord injuries.

Patients with cervical spinal cord injuries (CSCI) experience hypotension mainly caused by attenuated total peripheral resistance. Some physicians and nurses believe that this peripheral blood pressure (PBP) decrease in patients with CSCI results in the formation of pressure injuries. We hypothesized that the measurement of PBP, such as arteriole and capillary pressure, could be a simple and efficient means of detecting skin and deep tissue damage in patients with CSCI. However, PBP in the skin has not been measured in patients with CSCI. The purpose of this study was to investigate PBP in pat…

Empowering Women's Health after Spinal Cord Injuries: A Timely and Practical Update.

Women encompass about 20% of all the traumatic spinal cord injury (SCI) population and there is increased incidence and prevalence of women with SCIs. The most recent data estimate that approximately 40,000-45,000 women with SCI are living in the United States, increasing from previous years, with the most common cause still being motor vehicle collisions. Throughout their lifespan, women with SCI present with unique healthcare needs compare to the male population. Therefore we present a comprehensive summary to empower and equip all providers with awareness of these needs and recommendations

Combination therapy using nanomaterials and stem cells to treat spinal cord injuries.

As a part of the central nervous system, the spinal cord (SC) provides most of the communications between the brain and other parts of the body. Any damage to SC interrupts this communication, leading to serious problems, which may remain for the rest of their life. Due to its significant impact on patients' quality of life and its exorbitant medical costs, SC injury (SCI) is known as one of the most challengeable diseases in the world. Thus, it is critical to introduce highly translatable therapeutic platforms for SCI treatment. So far, different strategies have been introduced, among which u

How to generate graded spinal cord injuries in swine - tools and procedures.

Spinal cord injury (SCI) is a medically, psychologically and socially disabling condition. A large body of our knowledge on the basic mechanisms of SCI has been gathered in rodents. For preclinical validation of promising therapies, the use of animal models that are closer to humans has several advantages. This has promoted the more-intensive development of large-animal models for SCI during the past decade. We recently developed a multimodal SCI apparatus for large animals that generated biomechanically reproducible impacts in vivo. It is composed of a spring-load impactor and support systems

QUAD fever: beware of non-infectious fever in high spinal cord injuries.

A case of cervical spinal cord injury and quadriparesis with prolonged fever is being described. Initially, the patient received treatment for well-documented catheter-related bloodstream infection. High spiking fever returned and persisted with no obvious evidence of infection. The usual non-infectious causes too were carefully excluded. QUAD fever or fever due to spinal cord injury itself was considered. The pathogenetic basis of QUAD fever is unclear but could be attributed to autonomic dysfunction and temperature dysregulation. Awareness of this little known condition could help in avoidin

Caring for reproductive-aged women with spinal cord injuries: a case report.

In 2008 there were an estimated 259,000 people living in the USA with spinal cord injuries (SCI). The majority of these people are in their reproductive years, and over 19% are estimated to be women. Advances in medical management have allowed many women to live successfully with congenital defects or injuries resulting in SCI that even a few years ago would have been fatal. Although many of these women may be classified as 'disabled', fertility is not usually affected in SCI and many of these women desire children of their own. It is important to counsel these women regarding the range of iss…
