The accumulation of false-negative results in the immunocytochemical investigation of vaginal swabs of nonsecretors was our reason for checking ABH blood-group labeling in female genitalia in situ (11 sections each from 51 different autopsies). The epithelia and endothelia showed positive staining in the blood group concerned. The epithelia of the secretors were strongly marked in the fallopian tube, vagina, urinary bladder, and urethra, whereas in the other sections the staining was irregular. The superficial layers of the vaginal epithelium of nonsecretors were not marked, which explains the

Advances in spatial proteomics have fundamentally transformed the analysis of complex tissues by enabling the simultaneous interrogation of protein expression and spatial organization within intact biological samples. Early immunohistochemical approaches were limited by low multiplexing capacity, restricting the ability to capture cellular heterogeneity and microenvironmental interactions. Recent technological developments have markedly expanded multiplexing power, allowing the parallel detection of dozens of protein markers while preserving tissue architecture and spatial context. Multiplexed

We present a simple, low-cost, and efficient protocol for fluorescent staining of Bacillus spp. endospores using the thioflavin T (ThT) dye. The article provides step-by-step procedures for culturing Bacillus paranthracis, Bacillus thuringiensis (2 strains), and Bacillus subtilis, purifying endospores from pure cultures, and staining them with ThT. ThT-stained endospores retain fluorescence for at least 2 years when stored at 4°C in the dark and remain fluorescent following formaldehyde fixation. Importantly, ThT staining does not impair spore viability, and stained endospores germinate normal

Immunoreagents, such antibodies, must often be functionalized before being used for a specific application, and this modification can strongly affect their biophysical characteristics. In contrast, recombinant antibody fragments can be engineered with elevated precision at selected residues to obtain customized reagents. Several alternative strategies can be used that exploit different principles and require different effort to reach this objective. Direct labeling can be obtained by fusing the antibody fragment to a fluorescent protein; otherwise, tag sequences can be appended to the binders

Meiosis assures formation of both male and female gametes. However, cytological studies of female meiotic chromosome behavior in plants are scarce, mainly due to the comparatively low number and inaccessibility of female meiotic cells. We present Female Meiotic cell IDentification (FeM-ID), an approach for cytological identification of female meiotic cells in Arabidopsis. By employing ASY1-eYFP-TurboID, female meiotic cells (chromatin and cytoplasm) are specifically labeled by biotin. This facilitates their rapid microscopic identification and detailed assessment of female meiotic chromosome b

While phenotypes affecting the development of the craniofacial region, axial skeleton, and limbs can sometimes be inferred from observations alone, revealing the composition of the skeleton of Xenopus tadpoles and froglets can often add more detailed and useful information. An estimated 11% of recognized rare genetic diseases are classified as skeletal disorders, with the latest nosology linking 552 genes. Both commonly used species of Xenopus, as tetrapod vertebrates, are highly amenable to genetic manipulation and have been used to study skeletal development as well as craniofacial and skele

The most widely accepted technique for the confirmation of semen presence in forensic biology is the microscopic identification of sperm cells. The process of manual microscopic examination, however, can often be time consuming, particularly where sperm numbers are low or where sperm cells are absent, particularly when in the presence of numerous non-sperm cells. The findings of this study demonstrate that automated microscopy utilising Deep Neural Network (DNN) allows for an improvement of the efficiency of examination times associated with microscopy. Additionally, the use of instrument and

Stress tolerant slow- or non-growing bacterial subpopulations represent a key factor for chronic, recurrent, antibiotic-tolerant infections and necessitate prolonged antibiotic therapy. Consequently, new tools that facilitate investigation of these small subpopulations are highly needed. To characterize slow-/non-growing populations of mycobacteria, we here utilized Vybrant DiD, a lipophilic, fluorescent organic probe that detected pathogenic and non-pathogenic species in a rapid, specific and non-invasive manner. It enabled accurate, direct quantification of mycobacterial replication rate in
