Hepatic organelle interaction. IV. Mechanism of succinate enhancement of formaldehyde accumulation from endoplasmic reticulum N-dealkylations.

Further evidence for organelle interaction during drug metabolism by the liver is presented. The apparent stimulation by succinate of formaldehyde accumulation in the medium, which was reported to occur with liver slices and homogenates as well as with mitochondria plus microsomes, has been shown to be the result of succinate inhibition of mitochondrial aldehyde dehydrogenase. The mechanism of succinate inhibition is shown to be by reverse electron transport, and an increase in the NADH to NAD+ ratio in the mitochondria; the aldehyde dehydrogenase requires the oxidized form of the pyridine nuc

Hypoxia Reprograms a Ciliary Scaffold into a Metabolic-Epigenetic Trafficking Hub via a Succinate-Driven DNMT3L-ASXL2 Axis.

Environmental hypoxia is a major yet mechanistically unresolved driver of male infertility. While the canonical hypoxia-inducible factor (HIF) and reactive oxygen species (ROS) pathways dominate current models, how chronic oxygen deprivation persistently rewires spermatogenic epigenetics and flagellar architecture remains unknown. Here, we identify CEP162-traditionally defined as a static ciliary transition zone scaffold-as a dynamic oxygen-sensitive signaling hub that orchestrates a pathogenic metabolic-epigenetic-trafficking axis. In human asthenozoospermia and murine hypoxia models, CEP162

Itaconation and Succination as Metabolite-Derived Cysteine Modifications in Inflammatory Signalling: Biochemical Mechanisms, Chemoproteomic Mapping, and Therapeutic Opportunities.

Inflammatory activation rewires cellular metabolism and generates electrophilic metabolites that can modify reactive cysteine residues. This review focuses on two metabolite-derived cysteine modifications: itaconation, driven by aconitate decarboxylase 1 (ACOD1)-dependent itaconate production, and succination, driven by fumarate accumulation. Although both involve cysteine engagement, they differ in chemical stability, metabolite source, cellular distribution, glutathione competition, and functional outcome. Itaconation is linked to Kelch-like ECH-associated protein 1 (KEAP1)-nuclear factor er

Role of Succinate Accumulation in Prolonged Febrile Seizure-Induced Neuronal Injury and Epileptic Seizures in Sprague-Dawley Rats.

BACKGROUND: Though succinate accumulation is a vital contributor to overexcitation and reactive oxygen species (ROS) production, whether it contributes to febrile seizures (FS) is unclear. We sought to mechanistically explore the role of succinate in prolonged FS. METHODS: Prolonged FS were induced by hyperthermic treatment in Sprague-Dawley pups. Inhibitors of succinate dehydrogenase (SDH), purine nucleotide cycle (PNC), and malate/aspartate shuttle (MAS) were administered, respectively. Succinate levels and oxidative stress damage were assessed, and epileptic seizures and learning and memor

Development and Validation of a Stability-Indicating HPLC-UV Assay for Quantification of Tafenoquine Succinate in a Novel Paediatric Antimalarial Formulation.

Tafenoquine, an 8-aminoquinoline antimalarial, represents a promising advancement in combating malaria, but its bitter taste and lack of paediatric formulations hinder effective treatment for children. In response, we have formulated a chocolate-based chewable tafenoquine tablet for paediatric use. A rapid, stability-indicating high-performance liquid chromatography method with ultraviolet detection was developed and validated for tafenoquine succinate quantification. Tablet samples were extracted with 85% methanol and analysed on a Waters XBridge C18 (150 × 4.6 mm, 5 μm) column using an isocr

Discovery of Novel Diphenyl-Oxime-Ether Derivatives as Potent Succinate Dehydrogenase Inhibitors via Fragment Combination Strategy.

Succinate dehydrogenase inhibitors (SDHIs) have emerged as one of the fastest-growing fungicides, yet their structural conservation and overuse have triggered widespread fungal resistance. Herein, we designed and synthesized a series of novel diphenyl-oxime-ether pyrazole-carboxamide derivatives by employing the fragment combination strategy. Among them, compound 8r exhibited potent SDH inhibition with an IC50 of 0.056 μM, which was approximately a 24-fold improvement over penthiopyrad (IC50 = 1.320 μM). In greenhouse trials, compounds 8t and 8z showed 50% and 40% disease control efficacy agai

Succinate and lactate produced as conserved biomarkers through chronic and transient substrate-level phosphorylation: from microorganisms to cancer.

ATP is the primary energy currency required by living organisms. Mitochondrial oxidative phosphorylation (OxPhos) produces most of the ATP in quiescent and differentiated cells. OxPhos interruption results in analogous bioenergetic adaptations across divergent evolutionary taxa, yet this adaptation is poorly recognized. Oxygen availability is a major determinant of the source of ATP generation across most eukaryotic cell types. Acute oxygen deprivation, mitochondrial dysfunction, high energy demand, or other metabolic cues can shift relative ATP production from OxPhos to high-throughput fermen

The succinate prodrug NV354 prevents brain lesions and late-stage motor dysfunction in mitochondrial complex I deficiency.

Leigh syndrome is a fatal pediatric neurodegenerative disease caused by mitochondrial dysfunction, which can be modeled in the Ndufs4 KO mouse with mitochondrial respiratory chain complex I (CI) deficiency. This study explores NV354, a prodrug of succinate with enhanced oral bioavailability and brain uptake, as a potential therapy to counteract this devastating condition. NV354 modulated whole-body respiration and metabolic flexibility, prevented late-stage motor dysfunction, delayed clinical ataxia scores, and improved body weight development, but had otherwise minimal effect on neurobehavior

Identification of a pathological feed-forward loop linking succinate, HIF1α signalling and vascular remodelling in abdominal aortic aneurysm.

BACKGROUND: Succinate is a metabolite with roles beyond energy production, including regulation of metabolic pathways, signalling and protein modifications. Abdominal aortic aneurysm (AAA) is a disease characterized by deleterious remodelling of the aortic wall, increasing the risk of rupture and sudden death. Although succinate has been linked to vascular diseases, its regulation within the aorta and potential involvement in AAA remain unclear. AIMS AND RESULTS: In this study, we combined analyses of human plasma and aortic tissue with cell culture experiments to help understand the role of

Gut microbiota-induced elevation of succinate exacerbates diabetic myocardial ischemia/reperfusion injury by promoting macrophage polarization.

INTRODUCTION: Patients with diabetes exhibit increased susceptibility to myocardial ischemia/reperfusion (IR) injury. Emerging evidence highlights gut microbiota-derived microbial metabolites as critical modulators of the gut-heart axis. This study examined the pathological role of succinate, a crucial metabolite derived from the microbiota, in the progression of diabetic myocardial IR injury. METHODS: The gut microbiota composition and succinate levels in diabetic mice were analyzed. Changes in succinate levels were subsequently assessed following the depletion of the gut microbiota induced

Stress Hyperglycemia Drives CD4+ T Cell PANoptosis and Postoperative Organ Injury via Monocyte-Derived Succinate.

Perioperative stress hyperglycemia is a transient but frequent metabolic disturbance strongly linked to postoperative organ injury and mortality; however, the immunometabolic mechanisms driving this association remain largely undefined. In a two-center cohort of patients undergoing total aortic arch replacement, we identify stress hyperglycemia as an independent determinant of poor postoperative outcomes that associates strongly with CD4+ T cell loss. Hyperglycemia induces inflammatory PANoptosis in CD4+ T cells from patients in response to surgical trauma. This results from elevated glucose d

Correlations Between Serum Vitamin D, Synovial Succinic Acid Levels and Disease Activity in Patients with Rheumatoid Arthritis.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune synovitis characterized by progressive joint destruction. Vitamin D exerts immunomodulatory and anti-inflammatory effects, and its insufficiency is highly prevalent in RA and closely related to exacerbated inflammatory status. Synovial succinate acts as a pro-inflammatory metabolic mediator that aggravates intra-articular inflammation. However, the interactive associations among vitamin D, synovial succinate, and RA disease activity remain poorly defined. This cross-sectional study aimed to elucidate their correlations in RA patient

An integrated bioinformatic investigation of succinic acid metabolism related genes in ccRCC followed by preliminary validation of SLC25A4 in tumorigenesis.

The defective function of succinate dehydrogenase promotes the development and progression of kidney renal clear cell renal cell carcinoma (ccRCC). However, the molecular mechanism of succinate metabolism-related genes (SMRGs) has not been extensively studied in KIRC. First, differentially expressed SMRGs (DE-SMRGs) were screened among genesets. Next, functional enrichment analysis was performed to investigate the functions of DE-SMRGs. The univariate Cox algorithm, LASSO, and multivariate Cox analysis were performed to obtain biomarkers and build a prognostic model. Afterward, enrichment and

Dehydroandrographolide succinate attenuates dexamethasone-induced skeletal muscle atrophy by regulating Akt/GSK3β and MuRF-1 pathways.

Andrographis paniculata (AGPA) is known for its wide-ranging biological activities, including antiviral, antipyretic, and anticancer properties. However, its effects on muscle atrophy have not been well understood. This study investigates the impact of andrographolide (AD) and dehydroandrographolide succinate (DAS), key components of AGPA, on skeletal muscle atrophy using in vitro and in vivo models. We employed dexamethasone (DEX)-treated mice and C2C12 myotubes as models of skeletal muscle atrophy. While DAS and AD did not reverse the DEX-induced reduction in body weight, both compounds sign

Synthesis of Psilocin, Psilocybin and 5-MeO-DMT Succinate, All Labelled With Carbon-14 at the Indole 2-Position.

Three novel 14C-labelled isotopologues of the psychoactive agents psilocin, psilocybin and 5-methoxy-N,N-dimethyltryptamine (5-MeO-DMT) were synthesised, all labelled at the 2-position of the indole. The syntheses involved incorporating the 3-dimethylaminoethyl substituent common to all three substances onto a 4- or 5-substituted indole intermediate via successive treatments with oxalyl chloride, dimethylamine and reduction with lithium aluminium hydride. Psilocybin-2-14C with a specific activity of 234 μCi/mg exhibited limited stability, but a 5.5-fold radio dilution with unlabelled psilocybi

Clonal Parabacteroides from Gut Microfistulous Tracts as Transmissible Cytotoxic Succinate-Commensal Model of Crohn's Disease Complications.

Crohn's disease (CD) has been traditionally viewed as a chronic inflammatory disease that cause gut wall thickening and complications, including fistulas, by mechanisms not understood. By focusing on Parabacteroides distasonis (presumed modern succinate-producing commensal probiotic), recovered from intestinal microfistulous tracts (cavernous fistulous micropathologies CavFT proposed as intermediate between 'mucosal fissures' and 'fistulas') in two patients that required surgery to remove CD-damaged ilea, we demonstrate that such isolates exert pathogenic/pathobiont roles in mouse models of CD
