Quantification of cortisol in human eccrine sweat by liquid chromatography - tandem mass spectrometry.

Cortisol has long been recognized as the "stress biomarker" in evaluating stress related disorders. Plasma, urine or saliva are the current source for cortisol analysis. The sampling of these biofluids is either invasive or has reliability problems that could lead to inaccurate results. Sweat has drawn increasing attention as a promising source for non-invasive stress analysis. A sensitive HPLC-MS/MS method was developed for the quantitation of cortisol ((11β)-11,17,21-trihydroxypregn-4-ene-3,20-dione) in human eccrine sweat. At least one unknown isomer that has previously not been reported an…

Sweat test practice in pediatric pulmonology after introduction of cystic fibrosis newborn screening.

UNLABELLED: The influence of the generalization of cystic fibrosis newborn screening (CFNBS) in France on sweat test (ST) prescription is unknown. In this French retrospective, descriptive, and multicenter study, we studied the indications, number, methods, and results of STs prescribed by a pediatric pulmonologist in children who had a negative CFNBS and an ST for respiratory symptoms in 2012. We included 502 children with 523 STs, performed with four different methods. The main indication was asthma (71.3 %), then chronic cough (52.4 %), atypical lower airway infections (42.2 %), and bronchi…

Sweat gland regeneration after burn injury: is stem cell therapy a new hope?

Stem cells are the seeds of tissue repair and regeneration and a promising source for novel therapies. The skin of patients with an extensive deep burn injury is repaired by a hypertrophic scar without regeneration of sweat glands and therefore loses the function of perspiration. Stem cell therapy provides the possibility of sweat gland regeneration. In particular, recent studies have reported the reprogramming of mesenchymal stromal cells into sweat gland-like (SGL) cells. We present an overview of recent researches into sweat gland regeneration with stem cells. Difficulties of sweat gland re…

Endocannabinoids regulate growth and survival of human eccrine sweat gland-derived epithelial cells.

The functional existence of the emerging endocannabinoid system (ECS), one of the new neuroendocrine players in cutaneous biology, is recently described in the human skin. In this study, using human eccrine sweat gland-derived immortalized NCL-SG3 model cells and a wide array of cellular and molecular assays, we investigated the effects of prototypic endocannabinoids (anandamide, 2-arachidonoylglycerol) on cellular functions. We show here that both endocannabinoids dose-dependently suppressed proliferation, induced apoptosis, altered expressions of various cytoskeleton proteins (e.g., cytokera…

Blood, urine, and sweat (BUS) study: monitoring and elimination of bioaccumulated toxic elements.

There is limited understanding of the toxicokinetics of bioaccumulated toxic elements and their methods of excretion from the human body. This study was designed to assess the concentration of various toxic elements in three body fluids: blood, urine and sweat. Blood, urine, and sweat were collected from 20 individuals (10 healthy participants and 10 participants with various health problems) and analyzed for approximately 120 various compounds, including toxic elements. Toxic elements were found to differing degrees in each of blood, urine, and sweat. Serum levels for most metals and metalloi…

Exercise-induced asthma may be associated with diminished sweat secretion rates in humans.

BACKGROUND: Muscarinic receptor agonists increase water secretion from the acinar cells of respiratory, sweat, salivary, and lacrimal glands. Mice lacking the gene for aqueous water channel aquaporin (Aqp) 5 exhibit methacholine-induced bronchiolar hyperreactivity when compared to normal mice. Individuals with asthma also have enhanced airway responsiveness to methacholine and diminished airway hydration. Because Aqp5 in humans is also expressed in respiratory, sweat, salivary, and lacrimal glands, we hypothesized that those individuals with exercise-induced asthma and excessive bronchiolar re…

Skin adnexal neoplasms--part 2: an approach to tumours of cutaneous sweat glands.

Tumours of cutaneous sweat glands are uncommon, with a wide histological spectrum, complex classification and many different terms often used to describe the same tumour. Furthermore, many eccrine/apocrine lesions coexist within hamartomas or within lesions with composite/mixed differentiation. In addition to the eccrine and apocrine glands, two other skin sweat glands have recently been described: the apoeccrine and the mammary-like glands of the anogenital area. In this review (the second of two articles on skin adnexal neoplasms), common as well as important benign and malignant lesions of …

Responsiveness to autologous sweat and serum in cholinergic urticaria classifies its clinical subtypes.

BACKGROUND: It has been reported that patients with cholinergic urticaria have a type 1 allergy to autologous sweat; however, the pathogenesis of that disorder has not been fully elucidated. OBJECTIVE: We investigated the responsiveness to autologous sweat and serum in patients with cholinergic urticaria in relation to their clinical characteristics. We further classified the clinical subtypes that are clearly characterized by responsiveness to in vivo and in vitro tests as well as their clinical features. METHODS: Intradermal tests with autologous sweat and serum were performed in 18 patien…

Sweat gland carcinoma with metastases to the skin: response to 5-fluorouracil chemotherapy.

A 68-year-old white man presented to the inpatient service at Columbia-Presbyterian Hospital with a chief complaint of a progressively enlarging tumor of the right infraorbital region for many years and multiple, pink papulonodules on his head, neck, and trunk. A biopsy specimen of the right infraorbital and back lesions demonstrated an infiltrating adenocarcinoma with prominent signet ring cells, and small lumen formation. Results of an extensive work-up revealed bone marrow metastases and no evidence of a primary malignancy. We present a case of primary sweat gland carcinoma with metastases …

[Contribution of monoclonal antibody D 47 in the study of sweat gland pathology].

D 47 is a monoclonal antibody (IgG2) reacting with a surface antigen of cortical thymocytes. On normal human skin, D 47 was found to react with a cytoplasmic antigen of the cells of the secretory portion of eccrine sweat glands (ESG). No reactivity with D 47 is detected on the excretory part of ESG, on apocrine glands and all other cutaneous structures. In this work we studied through an indirect immunofluorescence method on frozen skin sections the reactivity pattern of D 47 on a group of epithelial skin tumours of certain or alleged glandular differentiation. These consisted of: eccrine spir…

Integrated Multimodal Fusion Network and Multispectral Sensing Photoelectrochemical Platform for Sweat Glucose Monitoring.

Sweat glucose detection offers the advantages of being painless, noninvasive, and suitable for continuous monitoring, showing broad application prospects in diabetes management. However, a single sweat glucose detection method cannot meet the requirements of multiscenario applications. To address this challenge, we developed a multimodal sweat glucose sensing platform integrating a type-II heterojunction with rare-earth nanoparticles (RENPs), which achieves an ultralow limit of detection (LOD) and a wide linear range while enabling robust adaptation to diverse scenarios. First, a photoelectroc…

Fingertip Sweat Leucine Detection Based on Electrochemical MIP Sensors Integrated with Porous Hydrogels.

Leucine (Leu) is a promising biomarker for metabolic health and muscle growth, offering significant potential for assessing physical well-being. Sweat sensors for Leu detection eliminate the reliance on invasive blood analysis and the sophisticated, large-scale instrumentation. Current sweat sensors, however, are complex to fabricate, exhibit low sensitivity toward nonelectroactive Leu, require intense exercise or thermal/chemical stimulation to generate sweat, and lack reusability. This work reports a flexible, highly sensitive, and reusable sweat sensor based on molecularly imprinted polymer…

Preserving Sample, Interface, and Signal Fidelity in Wearable Sweat Electrolyte Monitoring During Exercise.

Sweat electrolytes, including Na+, K+, and Cl-, provide dynamic information on exercise-induced fluid and electrolyte regulation. Wearable ion-selective electrode (ISE) platforms now enable continuous, in situ monitoring of these ions, but performance in standard solutions does not guarantee reliable on-body data. During exercise, measurement distortion can arise before sweat contacts the electrode, at the electrochemical interface, and during signal acquisition and interpretation. This review synthesizes these coupled distortion pathways through a three-layer fidelity framework. Sample fideli…
