In this study, the immunoregulation of IgG and IgM synthesis by synovial fluid lymphocytes (SFL) in rheumatoid arthritis (RA) was investigated. A group of 15 sero-positive rheumatoids with a level of lymphocytes in the joint effusion fluids greater than 40% were selected. In 20 experiments, Con A stimulated peripheral blood lymphocytes (PBL) suppressed Ig synthesis by autologous SFL, tested in the presence and absence of pokeweed mitogen. Enrichment of murine antigen-specific T cells was achieved by stimulation of primed lymph node cells with macrophages containing the immunizing antigen. The percentage of T lymphocytes that form rosettes (RFC) with sheep erythrocytes (E rosettes), cutaneous delayed-type hypersensitivity, and in vitro lymphocyte transformation was measured in 15 patients with primary intracranial tumors. The ability of T lymphocytes to form E rosettes was significantly impaired. These patients also exhibited impaired ability to become sensitized to dinitrochlorobenzene and humoral suppression of PHA-induced lymphocyte blastogenesis. Idiopathic CD4+ T lymphocytopenia (ICL) has been defined as a cause of immunodeficiency with a variable clinical course and an unknown etiology.
