The TASK-1 and TASK-3 activator JG-C3-98 attenuates cold and mechanical responses in primary somatosensory neurons.

K2P channels underlie background K+ currents that act as brakes on neuronal excitability. In the somatosensory system, the K2P channels TASK-1 and TASK-3 play a relevant role in cold sensing, mechanosensitivity, and pain. Combining molecular docking and molecular dynamic simulations, patch-clamp recordings of recombinant channels, Ca2+-imaging and patch-clamp analysis in cultured primary somatosensory neurons from trigeminal and dorsal root ganglia, extracellular recording of the nerve endings of trigeminal neurons at the corneal surface, and behavioral analysis in mice models of acute, irrita

Hemispheric lateralization in 47,XXY Klinefelter's syndrome boys.

Thirty-two boys with a 47,XXY karyotype were compared with chromosomally normal male controls in their performance on six tasks of hemispheric specialization. The results revealed that the 47,XXY subjects had smaller asymmetries on left hemisphere tasks and larger asymmetries on right hemisphere tasks than controls. Analyses of individual right and left side scores revealed that the atypical lateral asymmetries of the 47,XXYs were due to a shift toward greater right hemisphere involvement on four of the six measures. It was postulated that the slower fetal growth rates of the extra X chromosom

Assessing Specific Cognitive Deficits Associated with Dementia in Older Adults with Down Syndrome: Use and Validity of the Arizona Cognitive Test Battery (ACTB).

BACKGROUND: Down syndrome is associated with specific cognitive deficits. Alongside this, older adults with Down syndrome are a high risk group for dementia. The Arizona Cognitive Test Battery (ACTB), a cognitive assessment battery specifically developed for use with individuals with Down syndrome, has been proposed for use as outcome measures for clinical trials in this population. It has not been validated in older adults with Down syndrome. This study aims to assess the use and validity of the ACTB in older adults with Down syndrome. METHODS: Participants with Down syndrome aged 45 and ove

ERP correlates of form and rhyme letter tasks in impaired reading children: a critical evaluation.

Event-related potentials (ERPs) were obtained from 15 electrode sites in six average and six impaired reading children, 12 years of age, during visual letter discrimination tasks. Subjects responded to target letters with an enclosed area in the form task and to letters that rhymed with "e" in the rhyme task. Response accuracy was similar between the groups. Reaction time was relatively longer for the impaired group during the rhyme task. At lateral sites, condition differences were evident as greater negative shifts in the rhyme task than the form at 170 and 470 ms as well as a delayed late p

Modulation of Speech Motor Learning with Transcranial Direct Current Stimulation of the Inferior Parietal Lobe.

The inferior parietal lobe (IPL) is a region of the cortex believed to participate in speech motor learning. In this study, we investigated whether transcranial direct current stimulation (tDCS) of the IPL could influence the extent to which healthy adults (1) adapted to a sensory alteration of their own auditory feedback, and (2) changed their perceptual representation. Seventy subjects completed three tasks: a baseline perceptual task that located the phonetic boundary between the vowels /e/ and /a/; a sensorimotor adaptation task in which subjects produced the word "head" under conditions o

The effect of frontoparietal paired associative stimulation on decision-making and working memory.

Previous single-site neurostimulation experiments have unsuccessfully attempted to shift decision-making away from habitual control, a fast, inflexible cognitive strategy, towards goal-directed control, a flexible, though computationally expensive strategy. We employed a dual-target neurostimulation approach in 30 healthy participants, using cortico-cortical paired associative stimulation (ccPAS) to target two key nodes: lateral prefrontal cortex (LPFC) and intraparietal sulcus (IPS), to test whether decision-making can be artificially shifted from habitual toward goal-directed control. Partic

Auditory attentional shifts in reading-disabled students: quantification of attentional effectiveness by the Attentional Shift Index.

A controversy has existed for some years regarding auditory attentional skills in reading-disabled children. Data have suggested highly developed attentional skills in groups of reading-disabled students, but reduced attentional shifts have also been documented in equivalent groups. Attentional shifts in dichotic listening with forced or directed attention are usually inferred from a significant interaction between attentional task and ear. However, this procedure cannot be used to evaluate individual test performance, and the interaction does not give a useful measure of attentional shifts in

Effects of instructed focus and task difficulty on concurrent walking and cognitive task performance in healthy young adults.

Dual task paradigms can be used to examine the interactions between cognition and the control of posture and gait. Measuring and interpreting changes in dual task performance is challenging, however, because many factors can influence performance. This study examined the effects of instructed focus and walking task difficulty, and the interaction between these factors, on dual task performance in healthy young adults. Fifteen participants performed a cognitive task while walking with either a usual base or a narrow base of support. Participants were instructed to focus on either the cognitive

A behavioral integration mechanism underlies action timing.

Animals often display stereotyped behavioral patterns during timekeeping tasks. However, it remains unclear whether these behaviors are simply incidental-used by the animals to kill time-or whether they serve a functional role as part of the internal clock that enables animals to keep time. Here, using optogenetic stimulation of various brainstem, thalamic, and dopaminergic cell populations in mice trained on an action-timing task, we manipulated actions preceding the timing decision to causally test these hypotheses. Our findings show that bidirectional regulation of actions during the waitin

The selective cognitive benefits of long-term Tai Chi practice on executive function of students: a study on young adults.

INTRODUCTION: This study investigates the cognitive effects of long term (years of practice) Tai Chi practice, revealing mixed outcomes for executive functions. This study equally addresses the gap in research regarding the long term effects of Tai Chi, emphasizing its influence on the executive functions of young adults. METHODS: Using the BRIEFA scale and tasks like the Flanker task, More Odd Shift Task, and Nback tasks, significant differences emerged between Tai Chi students and nonstudents. RESULTS: In the results, Tai Chi students demonstrated superior inhibitory control and working me

Sensorimotor environment but not task rule reconfigures population dynamics in rhesus monkey posterior parietal cortex.

Primates excel at mapping sensory inputs flexibly onto motor outcomes. We asked if the neural dynamics to support context-sensitive sensorimotor mapping generalizes or differs between different behavioral contexts that demand such flexibility. We compared reaching under mirror-reversed vision, a case of adaptation to a modified sensorimotor environment (SE), with anti reaching, a case of applying an abstract task rule (TR). While neural dynamics in monkey posterior parietal cortex show shifted initial states and non-aligned low-dimensional neural subspaces in the SE task, remapping is achieved

Age-related changes in the effects of induced positive affect on executive control in younger and older adults-evidence from a task-switching paradigm.

Positive affect has been shown to promote task-switching performance in healthy young adults. Given the well-documented age-related decline in executive functioning, we asked whether induced positive affect also helps to improve task-switching performance in older adults. Sixty-eight younger and older adults performed a switching task before and after they had watched cartoon clips (positive affect group) or documentaries (neutral affect group). Positive affect was associated with reduced error rates across all trial types in both age groups. In older adults, the increase in accuracy came at t

Ferulic acid eicosyl ester enhances cognitive flexibility and modulates arousal-related neural circuits in mice.

Cognitive deficits are a major contributor to disability in numerous neuropsychiatric and neurodegenerative disorders, yet effective pharmacological treatments remain limited. Ferulic acid eicosyl ester (FAE-20), a natural constituent of the plant Rhodiola rosea, has previously been identified as an enhancer of simple forms of Pavlovian conditioning in flies, bees, and mice. Here, we investigated whether FAE-20 has further potential to enhance cognitive flexibility, working memory, or spatial learning in mice, and explored potential neurobiological mechanisms underlying such enhancement. How Healthy Older Adults Enact Lateral Maneuvers While Walking.

BACKGROUND: Walking requires frequent maneuvers to navigate changing environments with shifting goals. Humans accomplish maneuvers and simultaneously maintain balance primarily by modulating their foot placement, but a direct trade-off between these two objectives has been proposed. As older adults may rely more on foot placement to maintain lateral balance, they may be less able to adequately adapt stepping to perform lateral maneuvers. RESEARCH QUESTION: How do older adults adapt stepping to enact lateral lane-change maneuvers, and how do physical and perceived ability influence their task

What Drives Task Performance During Animal Fluency in People With Alzheimer's Disease?

BACKGROUND: Animal fluency is a widely used task to assess people with Alzheimer's disease (AD) and other neurological disorders. The mechanisms that drive performance in this task are argued to rely on language and executive functions. However, there is little information regarding what specific aspects of these cognitive processes drive performance on this task. OBJECTIVE: To understand which aspects of language (i.e., semantics, phonological output lexicon, phonological assembly) and executive function (i.e., mental set shifting; information updating and monitoring; inhibition of possible

Dopaminergic influences on executive function and impulsive behaviour in impulse control disorders in Parkinson's disease.

The development of impulse control disorders (ICDs) in Parkinson's disease (PD) may arise from an interaction among cognitive impairment, impulsive responding and dopaminergic state. Dopaminergic state may be influenced by pharmacologic or genotypic (catechol-O-methyltransferase; COMT) factors. We sought to investigate this interaction further by comparing those with (n = 35) and without (n = 55) ICDs on delay-discounting in different pharmacologic conditions (ON or OFF dopaminergic medication) and on response inhibition as well as aspects of executive functioning in the ON state. We then unde

Nurse-led interventions for hypertension management in Sub-Saharan Africa: A scoping review.

BACKGROUND: Sub-Saharan Africa exhibits the lowest global rates of hypertension detection, treatment, and control. Task shifting from traditionally physician-led care to other healthcare team members, mainly nurses, has been considered a viable solution to address the hypertension burden in this region. OBJECTIVE: This study aimed to map the available evidence on nurse-led hypertension management in Sub-Saharan Africa. METHODS: We followed the JBI guidelines for evidence synthesis in scoping reviews. A systematic database search was conducted across PubMed, Embase, Scopus, CINAHL Complete, W…

Task-evoked functional connectivity exhibits novel and strengthened relationships with executive function relative to the resting state.

Executive functioning in children has been linked to intrinsic brain network organization assessed during the resting state, as well as to brain network organization during the performance of cognitive tasks. Prior work has established that task-based brain networks are stronger predictors of behavior than resting state networks, yet it is unclear if tasks only strengthen relationships that exist weakly at rest or if tasks also evoke unique relationships. A lack of discernment regarding how tasks and the resting state commonly and uniquely support executive functions precludes a holistic under

Changes in oxygen sensitivity of TASK in carotid body glomus cells during early postnatal development.

A post-natal increase in carotid body (CB) hypoxia responsiveness occurs at the level of carotid sinus nerve activity, intracellular calcium, cell membrane depolarization and hypoxic inhibition of O(2)-sensitive background K(+) conductance. TASK-1, TASK-1/3 and TASK-3 are functionally expressed in CB glomus cells, with TASK-1/3 providing the major part of the O(2)-sensitive TASK-like background K(+) conductance. Here we report the effects of graded hypoxia on TASK-like channel activity in CB glomus cells from rats aged 0 to 1, 6 to 7 and 16 to 18 days; the time frame of postnatal CB functional
