Risk factors for nausea and vomiting requiring the daily administration of 5-HT3 receptor antagonists in radiotherapy combined with temozolomide for high-grade glioma.

Radiotherapy combined with temozolomide (TMZ+RT) is the primary treatment for high-grade glioma. TMZ is classified as a moderate emetic risk agent and, thus, supportive care for nausea and vomiting is important. In Nagoya University Hospital, all patients are treated with a 5-hydroxy-tryptamine 3 receptor antagonist (5-HT3RA) for the first 3 days. The daily administration of 5-HT3RA is resumed after the 4th day based on the condition of patients during TMZ+RT. Therefore, the present study investigated risk factors for nausea and vomiting in patients requiring the daily administration of 5-HT3R…

BACKGROUND: GBM is one of the most common and most aggressive brain tumors in adults, and upfront standard of care treatment has limited efficacy. Immune checkpoint inhibitor strategies have significantly improved outcomes in various solid tumors but have not proven effective in GBM, suggesting other strategies may be needed to realize their full potential. METHODS: GBM patients were treated with upfront standard of care chemoradiation with temozolomide and pembrolizumab, followed by adjuvant temozolomide and pembrolizumab for six nine-week cycles. Depending on production of sufficient vaccin…

Short-course radiotherapy with or without temozolomide in elderly patients with newly diagnosed glioblastoma: a time-to-event meta-analysis of overall and progression-free survival.

BACKGROUND: Glioblastoma is the most common and aggressive primary brain tumor in adults, with elderly patients facing particularly poor outcomes due to frailty and treatment intolerance. Short-course radiotherapy (SCRT) is a pragmatic approach, but the added benefit of temozolomide (TMZ) to SCRT is supported by limited evidence. METHODS: We conducted a search of PubMed, SCOPUS and Embase for studies published from inception to July 9, 2025. Eligible studies directly compared SCRT to SCRT + TMZ in elderly adults (≥ 65years) with newly diagnosed, histologically confirmed glioblastoma, and provi…
