BMPR2 Variants Underlie Nonsyndromic Oligodontia.

Oligodontia manifests as a congenital reduction in the number of permanent teeth. Despite the major efforts that have been made, the genetic … ranscripts were highly expressed in the dental papilla and adjacent inner enamel epithelium in mice tooth germs, suggesting that BMPR2 may play important roles in tooth development. Our findings broaden the genetic spectrum of oligodontia and provide clinical and genetic evidence supporting the importance of BMPR2 in nonsyndromic oligodontia.

Bilateral ectopic third molars in maxillary sinus associated with dentigerous cyst identified with ophthalmic, nasal and maxillary complication: A rare case report.

Etopic tooth eruption is the occurrence of the tooth germ in a nonanatomical position. It can be associated with dentigerous cyst, which is the second most seen in the development of odontogenic cyst commonly presented in mandibular region and seen in single form. It is usually accompanied with mandibular third molar followed by canine. Bilateral and multiple dentigerous cysts in nonsyndromic patients are fairly rare phenomenon and barely documented in the literatüre. We report a rare case of bilateral dentigerous cysts associated with ectopic third molars in the maxillary sinuses. Computerize…

[A statistic analysis on absence of third molar germs in orthodontic patients].

OBJECTIVE: To examine the absence of the third molar germs in orthodontic patients and to evaluate the relationship between third molar germs and malocclusion. METHODS: The subjects comprised 234 patients (male 92, female 142) from the orthodontic clinic whose ages were 14-18. The assessments of the third molar germs were made from panoramic radiographs, and the assessments of ANB angle were made from lateral cephalograms. All the data were analyzed by statistic chi2 test. RESULTS: The percentage of male who missed one or more third molar gems (37.0%) was higher than that of female (24.6%).

In vitro effects of calcitonin and/or parathyroid hormone on odontogenesis of mouse embryonic molars.

Mandibular first molars of mouse embryos were cultivated for examination of the effects of calcitonin (CT) and/or parathyroid hormone (PTH) on the odontogenesis of the molars, and for determination of whether and how CT, which is a PTH antagonist, has an influence on the effect of PTH on odontogenesis. On the second day, the inner enamel epithelium in the control group had already differentiated into pre-ameloblasts. Typical odontoblasts had secreted a layer of predentin. On the fourth day of culture, the pre-ameloblasts achieved terminal differentiation into secretory ameloblasts, and enamel

Specific binding sites for transferrin on ameloblasts of the enamel maturation zone in the rat incisor.

During enamel maturation in rodents, an iron-containing pigment is deposited into the surface layer of the enamel. Maturation zone ameloblasts presumably are responsible for this deposition. The presence of large amounts of ferritin in the cytoplasm of these cells suggests that they receive iron, presumably from circulating transferrin. An in vivo radioautographic binding assay using iodinated transferrin was used to determine if indeed maturation ameloblasts possess transferrin receptors at their cell surfaces. Experimental rats received systemic injections of labeled transferrin while contro

Iatrogenic displacement of an impacted third maxillary molar in the parapharyngeal space: A case report.

Accidental iatrogenic displacement of third molars in peripheral soft tissues is a major intraoperative complication, but is considered rare. A young women reported discomfort during mandibular lateral movements since she underwent the extraction her impacted third molars under general anesthesia, 2 years before. The clinical examination was not specific except for a laterality limitation of the right mandibular movements. The germ of 28 appeared radiologically encapsulated in the peripheral muscle tissues in the left parapharyngeal space and showed no signs of ankylosis. After measurement of

Otodental syndrome: a case report and genetic considerations.

A 5-year-old boy presented with otodental syndrome. His maxillary and mandibular incisors were within normal limits. The premolar/molar areas in all quadrants were occupied by markedly macrodontic teeth showing globular shape. The canines had a similar rounded shape. Two canines and 1 of the other abnormal teeth demonstrated areas of yellow hypoplastic enamel. Radiographs revealed that some of the abnormal teeth had bifurcated pulp chambers and pulp stones. Premolar tooth germs were absent. Audiometry demonstrated that the child had a marked bilateral sensorineural hearing loss for frequencies…

Tooth agenesis: from molecular genetics to molecular dentistry.

Tooth agenesis may originate from either genetic or environmental factors. Genetically determined hypodontic disorders appear as isolated features or as part of a syndrome. Msx1, Pax9, and Axin2 are involved in non-syndromic hypodontia, while genes such as Shh, Pitx2, Irf6, and p63 are considered to participate in syndromic genetic disorders, which include tooth agenesis. In dentistry, artificial tooth implants represent a common solution to tooth loss problems; however, molecular dentistry offers promising solutions for the future. In this paper, the genetic and molecular bases of non-syndrom

Osteopetrosis: a case report including gross and microscopic findings in the mandible at autopsy.

A case of juvenile osteopetrosis in a 10-year-old girl, including the skeletal, hematologic, and neurologic features, is described. Ankylosis of cementum to bone was evident in histologic sections, providing a possible explanation for the delayed or impaired eruption of teeth. A summary of the literature on the pathogenesis of osteopetrosis is presented.

Intramural calcifying epithelial odontogenic tumor.

The calcifying epithelial odontogenic tumor (CEOT) is a rare lesion of the jaws. It accounts for about 1% of all odontogenic tumors. The CEOT occurs primarily in the molar-premolar region of the mandible, and 52% of cases are associated with an unerupted tooth. This report describes an unusual case in a 37-year-old woman. The tumor arose in the molar area of the right mandible, appeared radiographically as a radiolucent lesion, and was thought to be a dentigerous cyst in association with an impacted first molar. The lesion was enucleated. Microscopic examination showed it to be a dental sac, w

SH3BP2-encoding exons involved in cherubism are not associated with central giant cell granuloma.

Central giant cell granuloma (CGCG) is a benign lesion with unpredictable biological behaviour ranging from a slow-growing asymptomatic swelling to an aggressive lesion associated with pain, bone and root resorption and also tooth displacement. The aetiology of the disease is unclear with controversies in the literature on whether it is mainly of reactional, inflammatory, infectious, neoplasic or genetic origin. To test the hypothesis that mutations in the SH3BP2 gene, as the principal cause of cherubism, are also responsible for, or at least associated with, giant cell lesions, 30 patients wi

[Orthodontic and oral surgery therapy in cleidocranial dysplasia].

A cleidocranial dysplasia is an autosomal dominant inherited condition consisting of generalized skeletal disorder. Associated dental signs are present in 93,5%; failure of tooth eruption with multiple supernumerary teeth, dilaceration of roots, crown germination, microdontia, high arched palate, midface hypoplasia, high gonion angle. The molecular- genetic analysis revealed a missense mutation in the CBFA1 gene located on chromosome 6p21, which is considered to be etiological factor for CCD. Orthodontic and oral surgery therapy of a 13 year-old child with CCD was performed due to aesthetic an

Comparison between two HNK-1-related antibodies immunoreactivity (HNK-1-anti-leu 7 and anti-HNK-1/N-CAM) during rat cephalogenesis.

The fixation sites of two antibodies, HNK-1-anti-leu 7 and anti- HNK-1/N-CAM, were studied during visceral cephalogenesis in the rat. The labelling patterns of both antibodies were different. Anti-leu 7 staining was restricted to trunk neural crest cells, the peripheral nervous system, myotomes and premuscular cells, the eye rudiment and the olfactory vesicle. On the other hand, anti- HNK-1/N-CAM extended to the otocyst, several epithelia (including tooth germs) and precartilages. Anti-HNK-1/N-CAM staining of somites and cranial nerve ganglia was more precocious than anti-leu 7 labelling. We c…

[Lysosomal storage diseases, genetic or drug-induced? effect of glycosaminoglycan and sphingolipid disorders on dental tissues].

In vivo studies were carried out on dental tissues of rat incisor after a single injection of suramin, a drug which induces mucopolysaccharidosis-like disease. Accumulation of lysosome-like structures was seen in secretory ameloblasts and odontoblasts. In vitro studies on embryonic tooth germ buds showed similar changes when they were cultured in presence of suramin. Anti-phospholipid immunolabelling revealed a developmentally regulated temporo-spatial pattern. Radiolabeling with 3H-suramin indicated cytosolic and nuclear incorporation. The drug acting as polyanion interacted directly with pre…

Developmental stages of the third molar in Israeli children.

PURPOSE: The purpose of this study was to examine the developmental stages of the third molar in Israeli children, compare its development in boys and girls, in the maxilla and mandible, right to left sides in various age groups, and relate the developmental stage to chronological age. METHODS: Panoramic radiographs of 693 healthy children and adolescents (328 boys and 365 girls) ages 7 to 16 were analyzed using a modified 6-stage method. Since the 16-year-old group was too small for statistical analysis, it was later excluded from the study. Radiographs were obtained from patient files. RES

Extraoral Root-End Resection May Promote Pulpal Revascularization in Autotransplanted Mature Teeth-A Retrospective Study.

Tooth germ autotransplantation of open apices of the teeth exhibits high pulpal healing rates, whereas that of mature permanent teeth normally causes irreversible pulpal necrosis. Extraoral root-end resection (EORER) during transplantation may promote pulpal revascularization (PRV) in transplanted mature teeth and reduce endodontic treatment requirement. This study compared the primary outcomes of survival rates, PRV, and root resorption and determined relevant confounders in autotransplanted mature and immature teeth. The medical charts of consecutive patients who underwent tooth autotranspla…
