Microgravity as a translational model for metabolic dysfunction: implications for obesity, insulin resistance, steatotic liver and pancreatic diseases.

PURPOSE: The long-term effects of physical inactivity, whether due to microgravity in spaceflight or an inactive lifestyle on Earth, can lead to a range of metabolic problems. This review examines the physiological response to reduced gravity and contemporary inactivity, with a focus on obesity, insulin resistance, type 2 diabetes, and hepatic and pancreatic steatosis, all of which are associated with metabolic dysfunction (MASLD, MASPD). METHODS: A narrative review approach was used, integrating evidence from spaceflight research, bed rest studies, and clinical trial data to examine the meta

Microgravity-induced muscle atrophy in rodent and human models.

Exposure to microgravity leads to severe skeletal muscle atrophy, particularly in muscles that act to maintain posture against gravity, and results in significant reductions in muscle mass, cross-sectional area, and contractile force. This review examines the physiological and cellular response of microgravity in both rodent and human models, emphasizing shifts in muscle fiber type, altered contractile properties, and molecular adaptations. Specifically, rodents exhibit rapid atrophy, predominantly in slow-twitch fibers, whereas human studies suggest greater intersubject variability, with both

Skeletal muscle adaptation to microgravity: how altered mechanical cues drive catabolic signaling.

Exposure to microgravity induces rapid and profound skeletal muscle atrophy, yet the mechanisms by which reduced mechanical loading is sensed and translated into coordinated cellular and tissue-level responses remain incompletely understood. Accumulating evidence indicates that muscle adaptation to microgravity arises from disruption of mechanosensing pathways that regulate membrane signaling, cytoskeletal organization, and extracellular matrix (ECM) structure. Here, we integrate findings from spaceflight, bed rest, immobilization, and in vitro models to examine how reduced mechanical loading

Approaching ocular risks during spaceflight with 3D printing: Technical strategies to protect astronaut vision.

Long-duration spaceflight poses significant risks to ocular health due to prolonged microgravity exposure and space environmental stressors, contributing to conditions such as Spaceflight-Associated Neuro-ocular Syndrome (SANS) and Spaceflight-Associated Dry Eye Syndrome (SADES). These conditions, along with radiation exposure and risk of ocular trauma in resource-limited environments, necessitate development of innovative countermeasures to safeguard astronauts' vision, particularly for future planetary missions such as Mars. Traditional ophthalmic care depends on specialized equipment and ma

Artificial Gravity Attenuates the Transcriptomic Response to Spaceflight in the Optic Nerve and Retina.

The development of eye pathology is a serious concern for astronauts who spend time in deep space. Microgravity is a major component of the spaceflight environment which could have adverse effects on ocular health. The use of centrifugation to exert forces that partially or fully mimic Earth-level gravity in space is a possible countermeasure to mitigate the effects of microgravity on the eye. Therefore, we subjected mice on the International Space Station (ISS) to microgravity (0 G) or artificial gravity by centrifugation at 0.33 G, 0.67 G, and 1 G, and then performed RNA sequencing (RNA-seq)

Adaptation to full weight-bearing following disuse in rats: The impact of biological sex on musculoskeletal recovery.

With the technological advances made to expand space exploration, astronauts will spend extended amounts of time in space before returning to Earth. This situation of unloading and reloading influences human physiology, and readaptation to full weight-bearing may significantly impact astronauts' health. On Earth, similar situations can be observed in patients who are bedridden or suffer from sport-related injuries. However, our knowledge of male physiology far exceeds our knowledge of female's, which creates an important gap that needs to be addressed to understand the sex-based differences re

Modelled-Microgravity Reduces Virulence Factor Production in Staphylococcus aureus through Downregulation of agr-Dependent Quorum Sensing.

Bacterial contamination during space missions is problematic for human health and damages filters and other vital support systems. Staphylococcus aureus is both a human commensal and an opportunistic pathogen that colonizes human tissues and causes acute and chronic infections. Virulence and colonization factors are positively and negatively regulated, respectively, by bacterial cell-to-cell communication (quorum sensing) via the agr (accessory gene regulator) system. When cultured under low-shear modelled microgravity conditions (LSMMG), S. aureus has been reported to maintain a colonization

[Characteristics of local human skeleton reactions to microgravity and drug treatment of osteoporosis in clinic].

Analysis of the results of long-term investigations of bones in cosmonauts flown on the orbital station MIR and International space station (n = 80) was performed. Theoretically predicted (evolutionary predefined) change in mass of different skeleton bones was found to correlate (r = 0.904) with position relatively the Earth's gravity vector. Vector dependence of bone loss ensues from local specificity of expression of bone metabolism genes which reflects mechanic prehistory of skeleton structures in the evolution of Homo erectus. Genetic polymorphism is accountable for high individual variabi

Effect of microgravitation on the human equilibrium.

The 38-year period of human space exploration has gained for us a lot of experience in the problem of space motion sickness. The authors collected some causes of space sickness from the literature and from their research results. We collaborated with the Soviet-Russian space researchers and elaborated those vestibular research methods that were carried out by human space missions in the Soviet-Russian space station, on Earth, and after flight: spontaneous eye movement, foveal and peripheral optokinetic nystagmus, and vestibular stimulation. Using an earthly model of the effect of microgravitat

The "Space Cycle" Self Powered Human Centrifuge: a proposed countermeasure for prolonged human spaceflight.

The Self Powered Human Centrifuge, or Space Cycle, is a countermeasure to the adverse physiologic effects of prolonged human exposure to spaceflight microgravity. This unique device simultaneously provides exercise, impact loading and gravity analogous acceleration to emulate conditions on Earth. One or two crewmembers pedal themselves about a shaft mounted to the space craft located "above" their heads. This creates a short arm centrifuge with a head-to-toe acceleration orientation. The potential advantages of the Space Cycle include: a) reversal of cephalad fluid shift, minimizing post fligh

Effects of microgravity on muscular explosive power of the lower limbs in humans.

The maximal explosive power of the lower limbs of one astronaut has been measured before launch, and 2, 6 and 11 days after re-entry from 31 days on the MIR Station (EUROMIR '94). The subject, sitting on the carriage-seat of a Multipurpose Ergometer-Dynamometer (MED) constructed ad hoc in our laboratory, pushed maximally with both feet on two force platforms (knees angle 110 degrees). The carriage was free to move backwards on two rails inclined 20 degrees upwards. The force (F) of the lower limbs and the speed of the carriage (v) were recorded and the instantaneous mechanical power (w) was ca

[Ultrasonic diagnosis of the changes in human tibia during 370-day antiorthostatic hypokinesia].

Nine volunteers, aged 27 to 41 years, were exposed to 370-day head-down tilting during which their tibia bones were examined by measuring acoustic parameters with the aid of exponential point concentrators located at a 15 mm distance. Although the bone acoustic profile showed individual variations, the decrease of the average ultrasound velocity in the middle part and the gradient of the ultrasound velocity in the distal part of the diaphysis occurred primarily in the first half of the bed rest study, thus reflecting changes in bone adaptation to mechanical loads (redistribution of elastic pro

European vestibular experiments on the Spacelab-1 mission: 4. Thresholds of perception of whole-body linear oscillation.

Thresholds for the detection of linear oscillatory motion at 0.3 Hz in the X, Y and Z body axes were determined during the flight of Spacelab-1 and on the ground pre- and post-flight, using the method of limits with a single staircase procedure. Pre-flight, Z axis thresholds (mean 0.077 ms-2) were significantly higher than X and Y thresholds (mean 0.029 ms-2). Measures obtained on three crew members in-flight exhibited thresholds greater, by a factor of 1.5-4.3, than those obtained pre-flight. Post-flight, two crew members had significantly elevated X and Y axis thresholds whereas the other tw

Problem of reproduction of food protein by autotrophic organisms in extraterrestrial conditions.

To supply a man during his prolonged stay in extraterrestrial space with dietary protein by storing it in space vehicles seems to be impractical. Protein reproduction could be realized at present only by biosynthesis, which is possible with the autotrophic organisms and is the most economic, universal and therefore quite possible for use in life-support systems. In the paper, experimental results are presented on the biological value of single-celled algae proteins. The results obtained suggested that algae biomass in human rations could not be used in quantities needed for turnover of the com

WISE 2005: altered cerebrovascular autoregulation after 60 day head-down bed rest.

We tested the hypothesis that 60 days of head-down bed rest (HDBR) would affect cerebrovascular autoregulation and that this change would be correlated with changes in tolerance to the upright posture. Twenty-four healthy women (32 +/- 4 yrs) participated in a 60-d bed rest study at the MEDES Clinic in Toulouse, France. End tidal CO2 (ETCO2), continuous blood pressure (BP), middle cerebral artery (MCA) velocity and time to presyncope (endpoint) were measured during an orthostatic tolerance test conducted before/after bed rest. Given the large range of change in tolerance even within assigned c

Spaceflight/microgravity inhibits the proliferation of hematopoietic stem cells by decreasing Kit-Ras/cAMP-CREB pathway networks as evidenced by RNA-Seq assays.

Exposure to spaceflight and microgravity causes physiologic and psychologic changes including bone loss, cardiovascular dysfunction, and immune dysfunction. Anemia and hematopoietic disorders are observed in astronauts after spaceflight. Hematopoietic stem and progenitor cells (HSPCs), which can self-renew and give rise to all blood cells, play vital roles in hematopoiesis and homeostasis; however, the molecular mechanisms responsible for the impacts of microgravity on the proliferation of HSPCs remain unclear. We maintained mouse bone marrow HSPCs in the presence of stem cell factor for 12 d

Effect of human head flexion on the control of peripheral blood flow in microgravity and in 1 g.

This study evaluated, in six healthy subjects, whether head flexion, which stimulates the vestibular system and the tonic neck receptors, interferes with cardiovascular regulation. Arterial parameters were measured continuously using a pulsed Doppler ultrasound probe during parabolic flights with subjects either in the supine craned-head position (control) or in the supine anterior neck flexion bent-neck position. Exposure to 0 g induced a fluid shift towards the head (stroke volume +8%, P<0.05). Compared to the control situation the mean (SD) blood flow in the femoral artery decreased [ -10 (
