In vivo Administration of Atorvastatin Does Not Impair Mitochondrial Function or Cellular Viability in the Prefrontal Cortex of Mice.

Atorvastatin is a widely prescribed statin used for its cholesterol-lowering effects; however, it also exerts known pleiotropic effects, including antioxidant and anti-inflammatory properties. Atorvastatin´s impact on mitochondrial function remains controversial, with reports of toxicity in skeletal muscle and liver, while exhibiting beneficial effects on the brain. Atorvastatin has a neuroprotective effect, modulating glutamatergic transmission and reducing oxidative stress. Additionally, we previously showed that atorvastatin treatment in vivo increases mitochondrial capacity in the mouse hi

Hormonal Regulation of Physiology, Innate Immunity and Antibody Response to H1N1 Influenza Virus Infection During Pregnancy.

In 2009, the H1N1 swine flu pandemic highlighted the vulnerability of pregnant women to influenza viral infection. Pregnant women infected with influenza A virus were at increased risk of hospitalization and severe acute respiratory distress syndrome (ARDS), which is associated with high mortality, while their newborns had an increased risk of pre-term birth or low birth weight. Pregnant women have a unique immunological profile modulated by the sex hormones required to maintain pregnancy, namely progesterone and estrogens. The role of these hormones in coordinating maternal immunotolerance in

Particulate Air Pollutants, Brain Structure, and Neurocognitive Disorders in Older Women.

INTRODUCTION: An increasing number of studies have suggested that exposure to particulate matter (PM) may represent a novel - and potentially amendable - environmental determinant of brain aging. The current longitudinal environmental epidemiological study addressed some important knowledge gaps in this emerging field, which combines the study of air pollution and neuroepidemiology. The investigators hypothesized that long-term PM exposure adversely influences global brain volume and brain regions (e.g., frontal lobe or hippocampus) that are critical to memory and complex cognitive processing

Enlarged cavum septum pellucidum as a neuroimaging signature of head impact exposure.

Cavum septum pellucidum (CSP) is commonly observed upon neuroimaging examination in individuals exposed to repetitive head impacts (RHI) and post-mortem in cases with chronic traumatic encephalopathy. Consequently, CSP has been proposed as a potential biomarker for RHI-related neurodegeneration, yet prevalence estimates of CSP across other neurodegenerative diseases and its clinical implications are largely unknown. We assessed CSP prevalence and clinical correlates in individuals with RHI exposure, a history of traumatic brain injury (TBI), a neurodegenerative disease (i.e. Alzheimer's diseas

Bidirectional Associations Between Adiposity and Cognitive Function and Mediation by Brain Morphology in the ABCD Study.

IMPORTANCE: Most epidemiologic studies examine the brain as an outcome in relation to adiposity (ie, the brain-as-outcome perspective), but it is also a potential risk factor associated with adiposity accumulation over time (ie, the brain-as-risk factor perspective). The bidirectionality hypothesis has not been fully explored in adolescent samples previously. OBJECTIVE: To assess bidirectional associations between adiposity and cognitive function in youth and test mediational pathways through brain morphology (specifically the lateral prefrontal cortex [LPFC]), lifestyle behaviors, and blood

Genetic and environmental influence on resting state networks in young male and female adults: a cartographer mapping study.

We propose a unique, minimal assumption, approach based on variance analyses (compared with standard approaches) to investigate genetic influence on individual differences on the functional connectivity of the brain using 65 monozygotic and 65 dizygotic healthy young adult twin pairs' low-frequency oscillation resting state functional Magnetic Resonance Imaging (fMRI) data from the Human Connectome Project. Overall, we found high number of genetically-influenced functional (GIF) connections involving posterior to posterior brain regions (occipital/temporal/parietal) implicated in low-level pro

Targeting metabolic reprogramming to enhance adoptive immunotherapy: emerging mechanisms and translational perspectives.

BACKGROUND: Adoptive cell therapy (ACT) has achieved breakthrough advances in the treatment of malignancies, demonstrating particularly remarkable efficacy in hematologic cancers. However, the therapeutic efficacy of ACT remains limited by multiple factors, including low tumor antigen expression, the limited in vivo persistence of infused immune cells, and the immunosuppressive characteristics of the tumor microenvironment (TME). Meanwhile, metabolic reprogramming, a hallmark of cancer, plays a pivotal role in immune cells by directly regulating their differentiation, functional states, and lo

Performance of clay pigeon shooters in multiple object tracking under different target loads: skill level and gender differences.

OBJECTIVE: To examine the effect of target load on the multiple object tracking performance of clay pigeon shooters, and to investigate differences in skill level (professional vs. amateur) and gender in this context. The attentional patterns in clay pigeon shooting differ fundamentally from those in ball sports. It remains unclear whether expert advantages exist in multiple object tracking among clay pigeon shooters, and whether long-term training can eliminate gender-based cognitive differences. METHODS: Forty clay pigeon shooters (20 professionals, 20 amateurs; 20 males, 20 females) comple

How much does the reduced electroencephalographic montage matter for seizure detection? A large-cohort simulation study.

OBJECTIVE: Subscalp electroencephalographic (EEG) systems with few channels have emerged as promising solutions for ultra-long-term seizure monitoring, but the impact of montage configuration on automated seizure detection is unclear. We compared automated detection performance between full-scalp and simulated reduced montages approximating published subscalp devices, and assessed variation by epilepsy type, lateralization, and localization. METHODS: We conducted a retrospective cross-sectional study of consecutive epilepsy monitoring unit (EMU) admissions from January 2017 to December 2024 a

Cellular immune responses 12 months after fractional or standard dose BNT162b2 booster vaccination in Mongolian adults.

Despite the role of booster doses in sustaining protection against emerging SARS-CoV-2 variants, global uptake remains low, highlighting the need for dose-sparing strategies that maintain durable immunity. We conducted a randomized controlled trial in Mongolian adults to examine long-term cellular immune responses to standard and fractional doses of BNT162b2 given as a third dose. A total of 601 participants, primed with ChAdOx1-S, BBIBP-CorV, or Gam-COVID-Vac, were randomized (1:1) to receive 15 μg (fractional dose) or 30 μg (standard dose) of BNT162b2. A subset of participants (N = 256) were

CURE: a phase-based therapeutic framework for chronic inflammation.

BACKGROUND: Chronic inflammation represents a central mechanism underlying the persistence of immune-mediated diseases. Unlike acute inflammation, which resolves through coordinated regulatory checkpoints and active resolution programs, chronic inflammation stabilizes in pathological states maintained by cytokine redundancy, tissue-resident immune memory, and self-reinforcing feedback loops that resist termination. CONCEPTUAL FRAMEWORK: Here, we introduce the CURE framework (CONTROL - UNLOAD and RESET - EQUILIBRATE), a phase-based therapeutic model grounded in systems immunology and nonlinear

Long-term misdiagnosis of Niemann-Pick disease type C as Wilson disease: A case report.

RATIONALE: Niemann-Pick disease type C (NPC) is a rare autosomal recessive lysosomal lipid storage disorder caused by pathogenic variants in NPC1 or NPC2. Adult-onset NPC may manifest with progressive ataxia, dysarthria, cognitive impairment, psychiatric symptoms, and splenomegaly, features that may overlap with Wilson disease (WD). Abnormal copper metabolism in NPC can further complicate the differential diagnosis. PATIENT CONCERNS: A 39-year-old man was admitted with a more than 15-year history of slowly progressive dysarthria and gait instability, accompanied by intermittent choking when d

The neuro-immune axis in preeclampsia: from the maternal-fetal interface to systemic dysregulation.

Preeclampsia (PE) is a complex hypertensive disorder of pregnancy characterized by new-onset maternal hypertension and multi-organ dysfunction. Although placental maladaptation and immune activation are well-established features of PE, growing evidence indicates that dysregulated neuro-immune-vascular integration critically contributes to disease initiation, progression, and long-term sequelae. Normal pregnancy requires coordinated immune and neural adaptations, particularly at the maternal-fetal interface, to support successful placentation. While placental pathology, angiogenic imbalance, an

Modeling NRTIs and PIs class drug therapy on the dynamics of HIV infection with real patient data analysis and optimized control strategy.

HIV/AIDS remains a major global health concern, necessitating advanced therapeutic strategies to enhance immune response and suppress viral replication. This research formulates and examines a novel fractional-order mathematical model that integrates Nucleoside Reverse Transcriptase Inhibitors (NRTIs) and Protease Inhibitors (PIs) to evaluate their collective effectiveness in managing HIV infection. Unlike existing models, our framework incorporates memory effects via fractional calculus, explicitly accounts for macrophage-derived viral load as an external reservoir, and employs optimal contro

Neurosteroid mitigation of developmental neurological dysfunction, long-term epileptic biomarkers, chronic neuroinflammation, and neurodegeneration in a pediatric rat model of organophosphate exposure.

Children are particularly susceptible to the neurotoxic effects of organophosphates, which can lead to developmental neuronal deficits and associated dysfunction, including cognitive disabilities, epilepsy, and associated comorbidities. Anticonvulsants like benzodiazepines fail to prevent the lasting neurobehavioral and neuropathological effects of organophosphate exposure, emphasizing the need for new anticonvulsants to address these effects. This study evaluated the efficacy of the synthetic neurosteroid ganaxolone (GX) in combating persistent behavioral deficits, electrographic abnormalitie

Reduced morbidity and mortality of cGVHD in patients who received treatment with mesenchymal stromal cells for steroid-resistant aGVHD: long-term follow-up of a randomized phase 3 trial.

BACKGROUND: Our open-label, multicenter, randomized, phase 3 trial showed that the incidence and severity of chronic graft-versus-host disease (cGVHD) reduced in steroid-resistant acute graft-versus-host disease (aGVHD) patients who underwent mesenchymal stromal cells (MSCs) treatments, but survival benefit was not received. Here, we present a post-hoc analysis of the 5-year follow-up to explore long-term survival and its underlying mechanism. METHODS: This long-term follow-up trial included steroid-resistant aGVHD patients, who were randomly assigned (1:1) to receive MSCs (MSC group) (1 × 10

[Study on effects of 40 Hz light flicker stimulation on spatial working memory in rats and its neural mechanism].

Alzheimer's disease (AD) is a neurodegenerative disease characterized by cognitive impairment, with the predominant clinical diagnosis of spatial working memory (SWM) deficiency, which seriously affects the physical and mental health of patients. However, the current pharmacological therapies have unsatisfactory cure rates and other problems, so non-pharmacological physical therapies have gradually received widespread attention. Recently, a novel treatment using 40 Hz light flicker stimulation (40 Hz-LFS) to rescue the cognitive function of model animals with AD has made initial progress, but

A Projectile Concussive Impact Model Produces Neuroinflammation in Both Mild and Moderate-Severe Traumatic Brain Injury.

Traumatic brain injury (TBI) is a major cause of death and disability and is experienced by nearly 3 million people annually as a result of falls, vehicular accidents, or from being struck by or against an object. While TBIs can range in severity, the majority of injuries are considered to be mild. However, TBI of any severity has the potential to have long-lasting neurological effects, including headaches, cognitive/memory impairments, mood dysfunction, and fatigue as a result of neural damage and neuroinflammation. Here, we modified a projectile concussive impact (PCI) model of TBI to delive
