Chaperone-mediated autophagy (CMA) is a selective autophagy pathway that targets specific proteins containing a KFERQ-like motif for lysosomal degradation. It has been shown by us and others that CMA decreases during physiological aging in most tissues, and its impairment is associated with increased incidence of age-related pathologies, such as cardiovascular disease, neurodegenerative disorders or sarcopenia. However, its involvement in age-related macular degeneration (AMD), a prevalent progressive maculopathy that leads to bilateral central vision loss, had not been explored. In the early

Neurodegenerative diseases, including Alzheimer's disease (AD) and Parkinson's disease (PD), are major causes of cognitive and motor decline, yet early diagnosis remains challenging due to asymptomatic phases and limited non-invasive biomarkers. This narrative review systematically synthesized studies on retinal imaging in AD and PD. Published studies were identified through searches of PubMed, MEDLINE, Google Scholar, and reference lists, focusing on Optical Coherence Tomography (OCT), OCT Angiography (OCTA), and Spectral-Domain OCT (SD-OCT) assessing retinal structural and vascular changes.

Retinal degenerative diseases range from rare inherited forms to common multifactorial disorders such as age-related macular degeneration, which is the leading cause of blindness in developed countries. Recent evidence identifies impaired autophagy as a key pathogenetic mechanism. In the disease process alterations of the outer retina start from the retinal pigment epithelium (RPE), to progress downstream in the inner retina leading to widespread whole retinal degeneration. Recent studies indicate that among autophagy-related proteins Beclin1 plays a relevant effect in sustaining retinal integ

Neurocutaneous melanosis (NCM) and Dandy-Walker malformation (DWM) are both uncommon congenital condition, involving the central nervous system (CNS). Combined NCM with DWM have been reported in a few studies. Potential consequences, including hydrocephalus and CNS malignancies make these illnesses extremely challenging to diagnose and treat. Occurrences of NCM and DWM together, can result in progressive neurological consequences and patients with this condition seems to have really poor prognosis. This case report describes a 5-day-old girl who was born with several large pigmented cutaneous

Malattia Leventinese (MAL) is an inherited macular degeneration disorder characterized by retinal drusen formation in adolescence, leading to vision loss. A mutation in the fibulin-3 gene (EFEMP1) causes MAL; however, the mechanisms underlying disease onset and drusen formation remain unclear. In this study, we generated induced pluripotent stem cell-derived retinal pigment epithelial (iPSC-RPE) cells from a patient with MAL to investigate disease mechanisms and potential therapies. MAL iPSC-RPE exhibited fibulin-3 and apolipoprotein E (ApoE) aggregation, increased endoplasmic reticulum stress

The retinal pigment epithelium (RPE) is essential for the health and function of the retina and, more specifically, the photoreceptors. Hence, the dysfunction of RPE leads to various retinal degeneration diseases. The rising prevalence of these disorders highlights the need for effective, large-scale cell-based therapies, particularly those utilizing stem cell-derived RPE cells. This study presents the development of a biodegradable, smooth surface microcarrier designed to facilitate the expansion and maturation of RPE cells derived from induced pluripotent stem cells (iPSCs). Ipomoea batatas, commonly known as sweet potato, is an increasingly valued functional food because of its vivid coloration and rich bioactive compounds, especially anthocyanins and carotenoids, such as ipomoeaxanthin. This review focuses on the bioavailability, mechanisms of action, and therapeutic potential of sweet potato-derived anthocyanins in diabetes and metabolic disorders. Anthocyanins, which are plant pigments, exhibit high antioxidant activity by scavenging free radicals and stimulating endogenous antioxidant enzymes such as catalase and superoxide dismutase, thereby protecting cellu

Microscopic cell classification is a fundamental challenge in both clinical diagnosis and biological research. However, existing methods still struggle with the complexity and morphological diversity of cellular images, leading to limited accuracy or high computational costs. To overcome these constraints, we propose an efficient classification method that balances strong feature representation with a lightweight design. Specifically, an Inception-Linear Attention-based Lightweight Vision Transformer (ILViT) model is developed for microscopic cell classification. The ILViT integrates two innov

INTRODUCTION: Tenosynovial giant cell tumor (TGCT), previously referred to as pigmented villonodular synovitis (PVNS), is a rare, locally aggressive proliferative disorder of the synovium. It typically affects large joints, most commonly the knee. Posterior compartment involvement in the knee is less common compared to anterior or suprapatellar involvement, and it presents unique diagnostic and therapeutic challenges due to the complex anatomy and proximity to neurovascular structures. While popliteal (Baker's) cysts may coexist with intra-articular TGCT, they are considered synovial herniatio

Pentosan polysulfate (PPS) maculopathy is a progressive, vision-threatening retinal disorder linked to prolonged use of PPS, a heparin-like sulfated polysaccharide prescribed for interstitial cystitis/bladder pain syndrome. Affected individuals often experience impaired dark adaptation and progressive central vision loss. Fundus imaging commonly reveals hyperpigmented macular clumps at the level of the retinal pigment epithelium (RPE), and a distinctive pattern of autofluorescence abnormality in the posterior fundus. This degenerative maculopathy may continue to progress even after drug cessat

To compare the efficacy and safety of picosecond lasers (PSLs) versus nanosecond lasers (NSLs) for treatment of hyperpigmented disorders and tattoos, we conducted a systematic review of literature. PubMed, Web of Science and Cochrane library (from inception to December 2024) were searched for articles. The primary outcome was the treatment success rate (≥ 75% clearance of pigment), and the secondary outcomes were the visual analogue scale (VAS) to assess pain and the rates of adverse events. We analyzed 20 randomized controlled trials and 1 retrospective comparative study comprising a total of

Stargardt disease type 1 is a genetic retinal disorder caused by mutations in the ABCA4 gene, leading to toxic bisretinoid accumulation in the retinal pigment epithelium (RPE). This study examines the retinal phenotype of Abca4-/-Rdh8-/- double knockout mice on a C57BL/6J background, excluding confounding variants such as the Rpe65 Leu450Met variant or the rd8 mutation in the Crb1 gene, to define the effect of combined Abca4 and Rdh8 deficiency. Double knockout mice-confirmed free of the rd8 mutation and to not carry the protective Met variant of the Rpe65 gene-were analyzed at 4 and 9 months

INTRODUCTION: The retinal pigment epithelium (RPE) plays essential roles in maintaining retinal functions as well as choroidal capillaries and can lead to visual disorders if dysfunctional. Transplantation of human-induced pluripotent stem cell-derived RPE (hiPSC-RPE) is a promising therapy for such RPE impaired conditions including age-related macular degeneration. The challenge with cell suspension transplantation is targeted delivery of graft cells and undesired cell reflux. Gelatin hydrolysate, a soluble variant with specific molecular weight distribution, is examined in this study for its
