Gallbladder aspiration for acute cholecystitis in high-surgical-risk patients.

In humans, prolonged sedations with propofol or using high doses have been associated with propofol infusion syndrome. The main objective of this study was to evaluate the effects of prolonged high-dose administration of a specific propofol emulsion (Propofol Lipuro) and an improved lipid formulation (SMOFlipid) in liver mitochondrial bioenergetics and oxidative stress of rabbits, comparatively to a saline control. Twenty-one male New Zealand white rabbits were randomly allocated in three groups that were continuously treated for 20 h. Each group of seven animals received separately: NaCl 0.9

[Regeneration of auto- and isografts of submandibular glands in the brain of laboratory mice. I. Morphologic evaluation of the healed graft using light microscopy].

The mouse granular convoluted tubules of submandibular gland of laboratory mouse are releasing a range of biologically active peptides (renin, neural growth factor and others) into both the saliva and blood circulation, the males producing it in a larger extent than females. Recently, several from respective peptides were identified to be endogenous ones and brain-related. The present work was aimed to utilise submandibular gland/SMG auto- and isotransplants regenerating in murine brain as a possibly local source of peptides. Experimentally, the newborn and juvenile matured white A breeded mic

Oxidatively denatured proteins are degraded by an ATP-independent proteolytic pathway in Escherichia coli.

E. coli contains a soluble proteolytic pathway which can recognize and degrade oxidatively denatured proteins and protein fragments, and which may act as a "secondary antioxidant defense." We now provide evidence that this proteolytic pathway is distinct from the previously described ATP-dependent, and protease "La"-dependent, pathway which may degrade other abnormal proteins. Cells (K12) which were depleted of ATP, by arsenate treatment or anaerobic incubation (after growth on succinate), exhibited proteolytic responses to oxidative stress which were indistinguishable from those observed in c

Responses of the L5178Y tk+/tk- mouse lymphoma cell forward mutation assay: III. 72 coded chemicals.

Seventy-two chemicals were tested for their mutagenic potential in the L5178Y tk+/- mouse lymphoma cell forward mutation assay, using procedures based upon those described by Clive and Spector (Mutat Res 44:269-278, 1975) and Clive et al. (Mutat Res 59:61-108, 1979). Cultures were exposed to the chemicals for 4 hr, then cultured for 2 days before plating in soft agar with or without trifluorothymidine (TFT), 3 micrograms/ml. The chemicals were tested at least twice. Significant responses were obtained with allyl isothiocyanate, p-benzoquinone dioxime, benzyl acetate, 2-biphenylamine HCl, bis(2

High-intensity intermittent training promotes adipose tissue browning via the IL-27/p38 MAPK-PGC-1α signaling pathway in diet-induced obese rats.

OBJECTIVES: This study investigated the effects of high-intensity intermittent training (HIIT) versus moderate-intensity aerobic training (MAIT) on IL-27 signalling and adipose tissue browning in obese rats. METHODS: Forty male Sprague-Dawley rats were randomly divided into two groups: standard diet (C, n = 10) and high-fat diet (HFD, n = 30). After 8 weeks of HFD feeding, 24 obese rats were further randomised into three subgroups: HFD (H, n = 8), HFD + moderate-intensity training (HMT, n = 8), and HFD + HIIT (HHT, n = 8). The HMT and HHT groups underwent 8 week training interventions (six se

Plasma proteomics stratification identifies phospholamban R14del carriers at risk for disease progression.

AIMS: Incomplete penetrance is common in genetic cardiomyopathy, but poorly understood. Here, we investigate the circulating molecular signature in a cohort of patients with one specific phospholamban (PLN) p.Arg14del (R14del, R14Δ/+) pathogenic variant underlying R14Δ/+ cardiomyopathy and its association with disease variability and progression. METHODS AND RESULTS: Targeted proteomics, metabolomics, and lipidomics were performed on plasma from 87 R14Δ/+ carriers across the disease spectrum. Unsupervised clustering of plasma proteomics classified R14Δ/+ carriers into clusters, which were eva

A Novel Hypothermic Preservation Formulation Containing SUL-138 Enables Long-Term Hypothermic Storage of Clinical-Grade CAR-T Cells.

Background/Objectives: Point-of-care (PoC) manufactured fresh chimeric antigen receptor (CAR)-T cells are typically formulated in hypothermic preservation formulations (HPFs) and stored under hypothermic conditions (2-8 °C) until administered to the patient. However, in current HPFs the shelf life of fresh CAR-T cells is short (~24-36 h) due to limited CAR-T cell stability, which poses significant time constraints on manufacturing procedures and logistics. The objective of this study was to improve the stability and extend the shelf life of fresh clinical-grade CAR-T cell drug products (DPs).

Rougan Tongluo Decoction Initiates Neuroprotection Against Cerebral Ischemia by Activating the Endogenous SLC6A8-Creatine-EARS2 Mitochondrial Pathway.

OBJECTIVE: The traditional Chinese medicine formula Rougan Tongluo Decoction (RGTL) was widely used to treat neurological injury after cerebral ischemia, though its specific underlying mechanisms remain unknown. This study investigates the mechanisms via which RGTL helps alleviate cerebral ischemic injury to provide theoretical support for its application in cerebral ischemia treatment. METHODS: A middle cerebral artery occlusion reperfusion (MCAO/R) rat model was established and treated with RGTL, N-acetylcysteine (NAC), creatine, or sh-EARS2. Network pharmacology and metabolomics were condu

Low-Thujone A. absinthium L. (Wormwood) Essential Oils and Extracts with Potential Antioxidative/Prooxidant Activity.

Nowadays, the global demand for medicinal plants, including A. absinthium L. (wormwood), has increased considerably, leading to significant pressure on their wild populations and the biodiversity of ecosystems. Consequently, the rates of exploitation may exceed those of natural regeneration. This destructive process can be reduced by cultivating plants with the desired secondary metabolites by transferring them from their natural habitats. The present study investigates phytochemistry and the potential antioxidative/prooxidant activity of low-thujone A. absinthium plants. The chemical composit

A stimuli-responsive gradient-structured membrane for dual controlled release of bioactive agents: application to chronic wound dressings.

Currently, asymmetric polymeric membranes exhibit numerous advantages for wound dressing applications. Usually, numerous systems allow the release of only one or several antibacterial drugs to fight against the bacteria present on the wound. However, to be efficient, it is necessary to disrupt biofilm formation in order to render the enclosed bacteria sensitive to the antibacterial agent. Here, we describe a system capable of achieving such goal via a double compartment asymmetric polymeric membrane designed for dual drug release. In view of elaborating our gradient-structured membrane for dif

Multifunctional and dual-targeting polymeric prodrug nanomicelles against tamoxifen-resistant breast cancer.

Tamoxifen (TAM) resistance remains a significant challenge in treating estrogen receptor (ER)-positive breast cancer. Overexpression of epidermal growth factor receptor (EGFR), variations in TAM metabolism, and increased expression of drug-resistant proteins (P-glycoprotein, P-gp) all contribute to the development of TAM-resistant breast cancer. The lack of specific drug distribution to tumors further complicates the treatment of TAM-resistant breast cancer. Simultaneously addressing these four factors—EGFR overexpression, TAM metabolic variations, P-gp-mediated drug efflux, and poor tumor-spe

Representative probiotic extracellular vesicles engineered with modified hyaluronic acid and tea polyphenol for enhancing bioactive stability and targeted cellular delivery.

Curcumin (Cur) and anthocyanin (Ant) are typical plant-derived bioactives with promising biological benefits, but poor stability and bioavailability limit their applications. To address these challenges, we engineered a targeted co-delivery platform by systematically comparing two representative, probiotic-derived extracellular vesicle carriers: outer membrane vesicles (OMVs) from Escherichia coli Nissle 1917 and extracellular vesicles (EVs) from Lactobacillus plantarum. Encapsulation efficiency (EE) of Cur was 41.6% for OMVs and 35.9% for EVs, whereas EE of Ant was 25.4% and 24.6%, respective

Soil texture controls bioplastic-induced greenhouse gas emissions and carbon-nitrogen cycling in agricultural soils.

Bioplastics (BPs) are increasingly used in agriculture, yet their behavior in soil and effects on carbon (C) and nitrogen (N) cycling depend strongly on soil conditions. This study evaluated the effects of three BPs (1% w/w; 400-700 μm) polyhydroxyalkanoates (PHA), polybutylene succinate (PBS), and polylactic acid (PLA) on soil C and N cycling, microbial activity, and greenhouse gas emissions in clayey and sandy agricultural soils during a 120-day incubation with four wetting-drying cycles (70-30% water-filled pore space). PHA produced the largest changes in soil biochemical processes includin
