Patients treated with hemodialysis (HD) repeatedly undergo intradialytic low arterial oxygen saturation and low central venous oxygen saturation, reflecting an imbalance between upper body systemic oxygen supply and demand, which are associated with increased mortality. Abnormalities along the entire oxygen cascade, with impaired diffusive and convective oxygen transport, contribute to the reduced tissue oxygen supply. HD treatment impairs pulmonary gas exchange and reduces ventilatory drive, whereas ultrafiltration can reduce tissue perfusion due to a decline in cardiac output. In addition to

Here, we summarize current understanding of functional involvement of microglial cells in the most common neurodegenerative disease to affect humans, which is sporadic or late-onset Alzheimer's disease (LOAD). Our review narrowly focuses on insights obtained from post-mortem neuropathological examinations of human brains paying particular attention to microglia as these cells have long been implicated as pivotal players in the cellular processes that lead to AD-type neurodegeneration. Although complete understanding of the roles played by microglia in AD neurodegeneration remains elusive, our

Visceral adipose tissue (VAT) inflammation plays a central role in longevity and multiple age-related disorders. Cellular senescence (SEN) is a fundamental aging mechanism that contributes to age-related chronic inflammation and organ dysfunction, including VAT. Recent studies using heterochronic parabiosis models strongly suggested that circulating factors in young plasma alter the aging phenotypes of old animals. Our study investigated if young plasma rescued SEN phenotypes in the VAT of aging mice. With heterochronic parabiosis model using young (3 months) and old (18 months) mice, we found…

Aging and psychosocial stress are associated with increased inflammation and disease risk, but the underlying molecular mechanisms are unclear. Because both aging and stress are also associated with lasting epigenetic changes, a plausible hypothesis is that stress along the lifespan could confer disease risk through epigenetic effects on molecules involved in inflammatory processes. Here, by combining large-scale analyses in human cohorts with experiments in cells, we report that FKBP5, a protein implicated in stress physiology, contributes to these relations. Across independent human cohorts

Collapse of proteostasis and accumulation of damaged macromolecules have been recognized as hallmarks of aging and age-related diseases. The proteasome is the major cellular protease responsible for intracellular protein degradation, having an impaired function during aging. We have previously shown that proteasome activation through overexpression of β5 proteasome subunit delays replicative senescence and confers resistance to oxidative stress in primary fibroblasts. Herein, we have investigated the impact of enhanced proteasome function on organismal longevity and aggregation-related patholo

BACKGROUND/AIMS: We have reported the involvement of deregulated autophagy and subsequent cellular senescence in biliary epithelial lesions in primary biliary cirrhosis (PBC). Given that mitochondria are a major target of autophagy, we hypothesized that deregulated autophagy of mitochondria may be involved in autoimmune pathogenesis in PBC.
METHODS: We examined immunohistochemically the expression of pyruvate dehydrogenase complex-E2 component (PDC-E2) and cytochrome c oxidase, subunit I (CCO), in livers taken from patients with PBC (n = 42) and control livers (n = 76). The colocalization of …

Telomerase, a cellular reverse transcriptase, adds telomeric repeats to chromosome ends. In normal human somatic cells, telomerase is repressed and telomeres progressively shorten, leading to proliferative senescence. Introduction of the telomerase (hTERT) cDNA is sufficient to produce telomerase activity and immortalize normal human cells, suggesting that the repression of telomerase activity is transcriptional. The telomerase transcript has been shown to have at least six alternate splicing sites (four insertion sites and two deletion sites), and variants containing both or either of the del

Intervertebral disc degeneration, the leading cause of chronic low back pain, remains incurable with traditional conservative therapies limited to symptomatic alleviation. We present an ECM-mimetic injectable hydrogel (HPTC) synthesized via dynamic crosslinking of hyaluronic acid-phenylboronic acid (HA-PBA) and tannic acid-cerium(III) metal-polyphenol networks (TA-Ce³⁺ MPNs), which faithfully recapitulates native nucleus pulposus ECM to enable functional tissue regeneration. In vitro, HPTC presented broad-spectrum reactive oxygen species scavenging and downregulated pro-inflammatory cytokine e

Triple-negative breast cancer (TNBC) poses significant therapeutic challenges due to its aggressive metastasis and immunosuppressive microenvironment. Herein, we developed a tumor-targeting biomimetic nanosystem (MTB@LM) integrating BRD4-targeting PROTAC (dBET6), Mn-TCPP-based sonosensitizers, and a tumor cell membrane-fusogenic liposome hybrid membrane coating. MTB@LM efficiently accumulates in tumors via homologous targeting and bypasses lysosomal degradation through membrane fusion-mediated delivery. Under ultrasound activation, the nanosystem triggers a multimodal cascade: (1) Mn-enhanced

INTRODUCTION: Ischemia/Reperfusion (I/R) and the increasing age of patients are coexisting challenges in today's cardiovascular medicine. Vascular I/R leads to endothelial dysfunction and is partially driven by inflammation. With increasing age, a profile of biomolecules known as the senescence-associated secretory phenotype (SASP) also increases. The SASP correlates with decreased resilience and intrinsic capacity to withstand various stressors and contains pro-inflammatory cytokines, likely fueling I/R injury. Senomorphics block SASP secretion. We investigated the effect of ruxolitinib on en

Osteoarthritis (OA) is a prevalent degenerative joint disease characterized by cartilage destruction, inflammation, and chondrocyte senescence, yet no disease-modifying therapies are currently available. Poly(ADP-ribose) polymerase-1 (PARP-1) has been implicated in inflammation and aging, but its role in OA pathogenesis remains incompletely understood. In this study, we investigated the effects of PARP-1 inhibition on OA progression using both in vitro and in vivo rat models. In vitro, rat chondrocytes were stimulated with interleukin-1β (IL-1β) to establish an OA-like phenotype. PARP-1 expres

Ulcerative colitis (UC), a principal subtype of inflammatory bowel disease, is characterized by chronic inflammation of the intestinal mucosa. In previous studies, intestinal senescence has been shown to exacerbate UC symptoms by promoting inflammation, indicating a potential mechanistic connection between intestinal senescence and UC. In this study, we established DSS-induced senescence models in colonic epithelial cells (NCM460) and UC mouse models. Through an anti-senescence compound screening, we identified the topical antibiotic Retapamulin (RET) as a novel agent that concurrently mitigat

BackgroundSystemic lupus erythematosus is a common autoimmune disease. Studies have suggested that defective stem cells could be involved in the pathogenesis of systemic lupus erythematosus, which leads to changes in the function of immune cells. By observing the cell morphology, autophagy, and senescence of bone marrow mesenchymal stem cells (BMSCs) from lupus mice and normal controls, this study investigated the role of IL-6 in autophagy and senescence of BMSCs and explored relevant mechanisms.MethodFemale MRL/lpr and C57 mice with similar weights and sizes at 20-22 weeks old were selected.

Chronic myeloid leukemia (CML) is a hematopoietic malignancy driven by the fusion gene BCR::ABL1. Drug resistance to tyrosine kinase inhibitors (TKIs), due to BCR::ABL1 mutations and residual leukemia stem cells (LSCs), remains a major challenge in CML treatment. Here, we revealed the requirement of the vitamin D receptor (VDR) in the progression of CML. VDR was upregulated by BCR::ABL1 and highly expressed in CML cells. Interestingly, VDR knockdown inhibited the proliferation of CML cells driven by both BCR::ABL1 and TKI-resistant BCR::ABL1 mutations. Mechanistically, VDR transcriptionally re

Obesity is a worldwide epidemic that predisposes individuals to many age-associated diseases, but its exact effects on organ dysfunction are largely unknown1. Hair follicles-mini-epithelial organs that grow hair-are miniaturized by ageing to cause hair loss through the depletion of hair follicle stem cells (HFSCs)2. Here we report that obesity-induced stress, such as that induced by a high-fat diet (HFD), targets HFSCs to accelerate hair thinning. Chronological gene expression analysis revealed that HFD feeding for four consecutive days in young mice directed activated HFSCs towards epidermal
