Familial risk of chronic rhinosinusitis with and without nasal polyposis: genetics or environment.

BACKGROUND: Chronic rhinosinusitis (CRS) is a highly prevalent inflammatory condition, with significant effects on morbidity and quality of life, yet little is known about its pathogenesis. Preliminary evidence suggests there is a heritable component to the multifactorial etiology of CRS; however, our understanding of this genetic susceptibility is limited. METHODS: Using an extensive genealogical database linked to medical records, the risk of CRS with nasal polyps (CRSwNP) and without polyps (CRSsNP) was calculated for relatives and spouses of adult probands (1638 CRSwNP and 24,200 CRSsNP p

IDO1 and IDO2 non-synonymous gene variants: correlation with crohn's disease risk and clinical phenotype.

BACKGROUND: Crohn's disease (CD) is a chronic inflammatory disease of the gastrointestinal tract. Genetic polymorphisms can confer CD risk and influence disease phenotype. Indoleamine 2,3 dioxygenase-1 (IDO1) is one of the most over-expressed genes in CD and mediates potent anti-inflammatory effects via tryptophan metabolism along the kynurenine pathway. We aimed to determine whether non-synonymous polymorphisms in IDO1 or IDO2 (a gene paralog) are important either as CD risk alleles or as modifiers of CD phenotype. METHODS: Utilizing a prospectively collected database, clinically phenotyped

Identification of novel mutations in Chinese Hans with autosomal dominant polycystic kidney disease.

BACKGROUND: Autosomal dominant polycystic kidney disease (ADPKD) is the most common inherited renal disease with an incidence of 1 in 400 to 1000. The disease is genetically heterogeneous, with two genes identified: PKD1 (16p13.3) and PKD2 (4q21). Molecular diagnosis of the disease in at-risk individuals is complicated due to the structural complexity of PKD1 gene and the high diversity of the mutations. This study is the first systematic ADPKD mutation analysis of both PKD1 and PKD2 genes in Chinese patients using denaturing high-performance liquid chromatography (DHPLC). METHODS: Both PKD1

Genotyping Giardia duodenalis isolates from dogs: lessons from a multilocus sequence typing study.

Giardiasis is a common infection of dogs, and the occurrence of both zoonotic and host-adapted assemblages of Giardia duodenalis is well documented in this host. In the current study, G. duodenalis isolates from dogs collected in Croatia from both private owners (n=44) and kennels (n=52) were analyzed at four genetic loci: the ITS1-5.8S-ITS2 (ITS), the glutamate dehydrogenase (gdh), the triosephosphate isomerase (tpi), and the beta-giardin (bg). Both generic and assemblage D specific primers were used for the amplification of the tpi gene. All data were stored in a dedicated database, and anal

Gene trap mutagenesis: a functional genomics approach towards reproductive research.

We have entered a new era of genomics in biomedical research with the availability of genome-wide sequences and expression data, resulting in the identification of a huge number of novel reproductive genes. The challenge we are facing today is how to determine the function of those novel and known genes and their roles in normal reproductive physiology, such as gamete production, pregnancy and fertilization, and the disease physiology such as infertility, spontaneous abortion and gynecological cancers. Mouse genetics has contributed tremendously to our understanding of the genetic causes of hu

Genome-Wide Association Study of Pterygium Versus Pinguecula Identifies Loci Associated with Lesion-Type Divergence.

PURPOSE: Both pterygium and pinguecula are ocular surface lesions associated with shared environmental risk factors, including ultraviolet (UV) radiation exposure. However, most pingueculae do not progress to pterygium, and while prior genetic studies suggest a genetic contribution to pterygium susceptibility, the difficulty of controlling for shared UV exposure has been a limiting factor. Here, we use array-based data and whole genome sequencing (WGS)-based data from the multiancestry All of Us Research Platform to perform a case-case genome-wide association study directly comparing individua

A Case Report: Treatment, follow-up, and literature review of type A insulin resistance syndrome caused by a De Novo INSR c.3328G>C mutation.

BACKGROUND: Type A insulin resistance syndrome (TAIRS) is a rare inherited disorder caused by mutations in the insulin receptor (INSR) gene. It is characterized by severe insulin resistance, hyperandrogenism, and acanthosis nigricans. Its clinical manifestations overlap with those of polycystic ovary syndrome (PCOS), making it prone to misdiagnosis, particularly in adolescent females. Currently, the diagnosis of TAIRS relies on clinical evaluation and genetic confirmation, while long-term management strategies remain limited and challenging. CASE PRESENTATION: We report a 12-year-old female p

Prevalence of genes encoding carbapenem-resistance in Klebsiella pneumoniae recovered from clinical samples in Africa: systematic review and meta-analysis.

BACKGROUND: The potential of Klebsiella pneumoniae (K. pneumoniae) to acquire and spread carbapenem-resistant genes is the most concerning characteristic of the bacteria. In hospitals and other healthcare settings, multidrug-resistant K. pneumoniae can be prevalent and cause severe infections, posing significant challenges to patient management. Studying genetic variants and drug-resistant mutations in pathogenic bacteria of public health importance is essential. Therefore, this study aimed to assess the overall prevalence of carbapenemase-encoding genes in K. pneumoniae across Africa. METHOD

Molecular epidemiology and population structure of Providencia stuartii obtained from humans and other sources.

Providencia stuartii is an emerging opportunistic pathogen in humans and belongs to a genus that has shown great genomic diversity. P. stuartii has shown relevant traits in antimicrobial resistance because of its intrinsic resistance to multiple antibiotics and its ability to acquire resistance genes that limit therapeutic options. The present study describes the molecular epidemiology of P. stuartii based on the multilocus sequence typing (MLST) scheme developed for this purpose. This non-PCR-based scheme uses seven complete housekeeping genes. In total, 518 genomes obtained from public datab

Plasmodium knowlesi clinical isolates from Malaysia show extensive diversity and strong differential selection pressure at the merozoite surface protein 7D (MSP7D).

BACKGROUND: The high proportion of human cases due to the simian malaria parasite Plasmodium knowlesi in Malaysia is a cause of concern, as they can be severe and even fatal. Merozoite surface protein 7 (MSP7) is a multigene family which forms a non-covalent complex with MSP-1 prior to receptor-ligand recognition in Plasmodium falciparum and thus an important antigen for vaccine development. However, no study has been done in any of the ortholog family members in P. knowlesi from clinical samples. This study investigates the level of polymorphism, haplotypes, and natural selection acting at th

Causative Genes of Homologous Recombination Deficiency (HRD)-Related Breast Cancer and Specific Strategies at Present.

Recently, homologous recombination deficiency (HRD) has become a new target for hereditary cancers. Molecular-based approaches for hereditary cancers in the clinical setting have been reviewed. In particular, the efficacy of the PARP inhibitor has been considered by several clinical trials for various kinds of hereditary cancers. This indicates that the PARP inhibitor can be effective for any kind of BRCA mutated cancers, regardless of the organ-specific cancer. Homologous recombination deficiency (HRD) has become a new target for hereditary cancers, indicating the necessity to confirm the sta

Comparison on epidemiology, tumor location, histology, and prognosis of intracranial germ cell tumors between Mayo Clinic and Japanese consortium cohorts.

OBJECTIVE: Central nervous system (CNS) germ cell tumors (GCTs) are rare malignant neoplasms that arise predominantly in adolescents and young adults. CNS GCTs demonstrate characteristic trends in national associations, with implications for both tumor incidence and genetics. Although the incidence of CNS GCTs is markedly higher in East Asia than Western countries, direct comparative analyses between these CNS GCT populations are limited. METHODS: In Japan, to facilitate the genomic analyses of CNS GCTs, the Intracranial Germ Cell Tumor Genome Analysis Consortium was established in 2011, and
