Construction and Immunogenicity of Recombinant Vaccinia Virus Vaccine Against Japanese Encephalitis and Chikungunya Viruses Infection in Mice.

Japanese encephalitis virus (JEV) is recognized as a public health risk by the World Health Organization. In Asia, each year, ∼70,000 people become infected with JEV, which results in ∼10,000 deaths. Chikungunya virus (CHIKV) is an RNA virus, whose infection mainly causes fever, myalgia, and skin rash. Although the mortality rate is low, it seriously affects daily life. JEV and CHIKV infect humans through mosquitoes; therefore, a recombinant vaccinia virus coexpressing JEV E and CHIKV E1 proteins was constructed to prevent their concurrent infection. In this study, after mice first immunizatio

Efficacy of a single dose of a novel topical combination product containing eprinomectin to prevent heartworm infection in cats.

Cats may be infected by heartworm, Dirofilaria immitis, through mosquito bites. They can develop severe heartworm disease when infective D. immitis larvae migrate and develop into adults in the pulmonary vasculature or other tissues. As there is no curative treatment for feline heartworm infection, the monthly administration of preventative treatment is recommended in endemic areas. Three controlled, blinded laboratory studies were conducted to evaluate the preventative efficacy of BROADLINE(®), a novel combination of fipronil, (S)-methoprene, eprinomectin, and praziquantel against D. immitis

Protection of red-legged partridges (Alectoris rufa) against West Nile virus (WNV) infection after immunization with WNV recombinant envelope protein E (rE).

West Nile virus (WNV) is maintained in nature in an enzootic transmission cycle between birds and mosquitoes, although it occasionally infects other vertebrates, including humans, in which it may result fatal. To date, no licensed vaccines against WNV infection are available for birds, but its availability would certainly benefit certain populations, as birds grown for restocking, hunting activities, or alimentary purposes, and those confined to wildlife reservations and recreation installations. We have tested the protective capability of WNV envelope recombinant (rE) protein in red-legged pa

Acyl Carrier Protein is Essential for Apicoplast Biogenesis in Malaria Parasites Independent of Fatty Acid Synthesis.

Acyl carrier protein (ACP) and its 4-phosphopantetheine prosthetic group canonically function as the soluble scaffold for acyl chain assembly and elongation during type-II fatty acid synthesis (FASII). Plasmodium malaria parasites retain a FASII pathway in the apicoplast organelle that has been the subject of considerable scrutiny and confusion. Although apicoplast FASII is essential for P. falciparum growth within mosquitoes and the human liver, this pathway is dispensable and largely inactive in blood-stage parasites that can scavenge host fatty acids. In contrast to FASII enzymes that can b

Comparative mitogenomic and phylogenetic insights from four newly sequenced tick mitochondrial genomes.

Ticks, recognized as the second most significant vector of pathogens after mosquitoes, are of considerable interest in medical research. Although mitochondrial genomes are commonly employed in the phylogenetic studies of insects and arthropods, investigations into tick mitochondrial genomes are relatively scarce. The evolutionary and phylogenetic relationships at the family and genus levels remain unresolved. In this study, the mitochondrial genomes of Haemaphysalis warburtoni, Ixodes crenulatus, Rhipicephalus bursa, and Rhipicephalus pumilio were first sequenced and annotated based on the Ill

Impact of male-sterilizing doses and mating status on blood feeding rates and longevity in Aedes aegypti, Aedes albopictus, and Anopheles arabiensis females.

The irradiation of female mosquitoes has been shown to influence blood feeding behavior, depending on dose, life stage at irradiation, and mating status. In sterile male releases during the sterile insect technique (SIT) programs, a small number of sterile females can be released accidentally, which may pose a manageable but nonnegligible risk regarding disease transmission. In this study, female Aedes aegypti, Aedes albopictus, and Anopheles arabiensis were exposed to gamma irradiation at male-sterilizing doses and the blood feeding rates and longevity of mated versus unmated sterile females

Pickpocket315 affects male mating behavior in the yellow fever mosquito Aedes aegypti.

The molecular basis of mating behavior in the important disease vector mosquito, Aedes aegypti, remains poorly characterized. We investigated the functional role of a pickpocket gene, ppk315, in male mating behavior using both RNAi-mediated knockdown and CRISPR/Cas9 approaches. Behavioral assays revealed that RNAi-treated males (dsPPK315) made fewer mating attempts, were less responsive to female acoustic cues, and were less likely to achieve copulation, though their latency to initiate contact when attempts were made was comparable to controls. Males with a CRISPR/Cas9-induced disruption to p

Triacylglycerol metabolism is a novel target to combat West Nile virus infection.

West Nile virus (WNV) is a zoonotic Orthoflavivirus transmitted by mosquitoes that is responsible for outbreaks of meningitis and encephalitis worldwide. Driven by climate change, WNV has expanded as a global public health concern, particularly in temperate regions. However, there are still no specific approved therapies, reinforcing the need for antiviral development. Previous works have documented that WNV multiplication strictly depends on certain cellular lipids. To identify novel lipid-related therapeutic targets, we analyzed the infection-driven alterations in the CNS lipidome, the prima

Microhaplotype deep sequencing assays to capture Plasmodium vivax infection lineages.

Plasmodium vivax elimination is challenged by dormant liver stages (hypnozoites) that can reactivate months after initial infection resulting in relapses. Relapsing infections confound antimalarial clinical efficacy trials due to the inability to distinguish between recurrences arising from blood-stage treatment failure (recrudescence), reinfection or relapse. Genetic relatedness of paired parasite isolates, measured by identity-by-descent (IBD), can provide important information on whether individuals have had single or multiple mosquito inoculations, thus informing on recurrence origin. We d

Zika virus and the fetal-maternal interface: deciphering the mechanisms of placental infection and implications for pregnancy outcomes.

Zika virus (ZIKV) is an emerging flavivirus primarily transmitted by Aedes mosquitoes. It has gained significant attention due to its potential teratogenic effects during pregnancy, particularly the increased risk of severe congenital outcomes such as microcephaly and fetal mortality. ZIKV's ability to breach the fetal-maternal interface and compromise placental function distinguishes it from other arboviruses. This mini-review explores how ZIKV targets key placental cells, including cytotrophoblasts, extravillous trophoblasts, syncytiotrophoblasts, and Hofbauer cells, to establish infection i

Evaluating the General Public's Knowledge of Malaria: A Nationally Representative Cross-Sectional Study.

Malaria is a life-threatening disease caused by Plasmodium, transmitted through the bites of infected female Anopheles mosquitoes. Despite global efforts, malaria remains a major health burden in developing countries. In Tanzania, cultural beliefs and misconceptions often delay treatment, especially in rural areas. This article aims to evaluate the level of malaria knowledge among the Tanzanian population and identify demographic factors associated with disparities in awareness. This cross-sectional study used secondary data from the 2021-2022 Tanzania Demographic and Health Survey, analyzing

Investigating the role of vitamin D in the prevention and control of dengue virus vectors and related diseases: a systematic review study.

Dengue fever, caused by the dengue virus (DENV) and transmitted by Aedes aegypti mosquitos, remains a global health concern with no definitive treatment. Given the challenges in vaccine development and disease management, micronutrients such as vitamin D have emerged as potential adjunctive therapies because of their immunomodulatory properties. The effectiveness of vitamin D in the prevention and control of dengue fever was assessed in this systematic review by evaluating vitamin D's impact on immune responses, viral replication, and clinical outcomes. Following Preferred Reporting Items for

A DNA vaccine candidate provides protection against Rift Valley Fever virus in sheep under natural field conditions.

Rift Valley Fever virus (RVFV) is a mosquito-borne zoonotic pathogen, that causes significant morbidity and mortality in livestock, including high abortion rates in pregnant animals and elevated case fatality in neonates, representing a major threat to both animal and human health. Vaccination is the most effective countermeasure to reduce RVFV's impact. In this study, we designed a veterinary DNA vaccine encoding a consensus RVFV glycoprotein precursor (GPC), optimized for expression in sheep. The construct was evaluated for immunogenicity in mice and sheep and for protective efficacy in shee

β2-tubulin and its promoter in the brown planthopper: A versatile tool for genetic control strategies.

At present, the application of CRISPR/Cas9 technology for genetic manipulation in insects is predominantly concentrated on Diptera model species, including Drosophila and mosquitoes. In contrast, non-model insects such as the brown planthoppers (BPH, Nilaparvata lugens), a major insect pest of rice, have received less attention in genetic manipulation due to insufficient tools. Here, the analysis of spatiotemporal expression patterns revealed that β2-tubulin in BPH (NlB2t) was predominantly concentrated in male adults and male testis, exhibiting high expression levels. Knockdown of NlB2t expre

Breakthroughs and insights: A comprehensive review of yellow fever vaccine breakthrough infection across 8 decades.

The yellow fever vaccine 17D is one of the most successful live-attenuated vaccines ever developed, controlling mosquito-borne yellow fever virus and yellow fever disease worldwide. Introduced in 1937, 17D never underwent rigorous phase III clinical trials to evaluate safety or efficacy, and while protection in the field was quickly established, no prospective evaluation of vaccine efficacy has ever been conducted. One important measure of vaccine efficacy is breakthrough infection resulting from vaccine failure. Yellow fever breakthrough infection was previously formally evaluated in a policy

Molecular Identification of Species Belonging to Culex vishnui Subgroup (Diptera: Culicidae), Vectors of Japanese Encephalitis Virus, in Taiwan.

Classification of mosquitoes with overlapping features remains problematic when using traditional morphological identification alone. In this study, we used molecular methods to elucidate the taxonomic status of Culex tritaeniorhynchus, Culex annulus, and Culex pseudovishnui species as vectors of the Japanese encephalitis virus belonging to the Culex vishnui subgroup and gene flow among them. In this study, 76, 59, and 3 samples of Cx. annulus, Cx. tritaeniorhynchus, and Cx. pseudovishnui, respectively, were collected around Taiwan. Phylogenetic analysis and genetic divergence were based on ge
