The molecular basis of susceptibility to childhood malignant hemopathy remains largely unknown. An excess of skeletal congenital anomalies has been reported among children with hematological malignancy and points towards involvement of developmental genes, like those belonging to the HOX gene family. In addition to their role in embryogenesis, HOX transcription factors are known to be regulators of proliferation and differentiation of hematopoietic cells. We aimed to explore the possibility that germline alterations of HOX genes might be involved in childhood acute lymphoid malignancies. A coh

The primary infection of Epstein-Barr virus (EBV) may result in fatal infectious mononucleosis or hemophagocytic syndrome (HPS) in 2 diseases; that is, X-linked lymphoproliferative disorder (XLP) and hemophagocytic lymphohistiocytosis (HLH). XLP is linked to mutations of the SAP/SH2D1A gene with dysregulated T-cell activation in response to EBV infection. Patients with sporadic HLH, however, usually have no mutation of the SAP/SH2D1A gene, and EBV latent membrane protein-1 (LMP1) can up-regulate Th1 cytokines in EBV-infected T cells. Since both diseases share common manifestations of HPS, it i

A broad array of antibodies directed against the variable (V) region of the T-cell receptor (TCR) beta (V beta) chain has become available in a directly conjugated multicolor format that permits assessment of 19 of 25 V beta families, covering 70% of the normal circulating T-cell repertoire. These antibodies were used to detect expanded T-cell populations in 43 peripheral blood samples submitted for suspected T-cell malignancy. Of 43 samples, 27 were diagnosed as follows: T-cell large granular lymphocyte leukemia, 14 samples; Sézary syndrome, 4 samples; T-cell prolymphocytic leukemia, 5 sample

Clinical and pathologic features of 24 patients with large cell lymphomas that expressed the activation antigen Ki-1 are described. Phenotypic and/or genotypic studies characterized these neoplasms as T-cell (16 cases), B-cell (six cases), or null cell (two cases) type. Males predominantly were affected. Age of patients ranged from 19 to 73 years, with a bimodal distribution, with peaks in the third and seventh decades. Lymphadenopathy was present in nearly all patients. Extranodal involvement, including skin, soft tissue, bone, central nervous system, lung, or small intestine was observed in

This report describes a patient with a large granular lymphocyte leukaemia (CD8 + lymphoproliferative disease) and severe neutropenia (less than 0.5 x 10(9)/l) in whom exercise resulted in a marked lymphocytosis, a phenomenon which has not previously been recorded. The lymphocyte count at rest was within normal limits (2.2 x 10(9)/l), then fell to the resting level within 15 min of cessation of exercise. The peripheral blood mononuclear cells showed the morphology of large granular lymphocytes (LGL) by light and electron microscopy both at rest (30%) and to a much greater extent during exercis

BACKGROUND: Malignancy is a leading cause of mortality after liver transplantation (LT), yet its long-term incidence and outcomes remain incompletely defined.
METHODS: We performed a retrospective single-center cohort study of 1841 adult LT recipients transplanted between 1990 and 2025 to characterize the spectrum, and outcomes post-transplant malignancy. Competing-risk methods accounting for non-cancer mortality were used to estimate cumulative incidence, and Fine-Gray regression identified independent predictors.
RESULTS: Over a median follow-up of 10.6 years, the 25-year cumulative incide

Human herpesvirus 8-associated multicentric Castleman disease (HHV-8-associated MCD) is a rare lymphoproliferative disorder that typically occurs in immunocompromised patients with HIV infection. HHV-8-associated MCD is characterized by systemic inflammatory manifestations driven by viral interleukin-6, where neurological complications are uncommon. This contrasts with POEMS-associated MCD, which is HHV-8 negative and frequently complicated by polyneuropathy. A 56-year-old HIV-negative man presented with prolonged high-grade fever, severe anemia, multiple mildly enlarged lymph nodes, hepatospl

BACKGROUND We report a 43-year-old man with ulcerative lesions of the right arm without pulmonary symptoms that was diagnosed as cutaneous lymphomatoid granulomatosis, a rare Epstein Barr virus-associated angiocentric and angiodestructive lymphoproliferative disorder that most commonly involves the lungs. Cutaneous involvement usually occurs in association with pulmonary disease, while isolated cutaneous disease is rare and most often presents with papules, nodules, or plaques. Necrotizing ulcers are uncommon and can closely mimic other dermatologic conditions. CASE REPORT A 43-year-old man pr

Renal hilar Castleman disease is a rare lymphoproliferative disorder characterized by non-specific radiological manifestations. It is easily misdiagnosed as primary renal or adrenal malignant tumors in clinical practice, which may consequently lead to unnecessary radical nephrectomy. A 51-year-old female with a history of right breast cancer treated with mastectomy and adjuvant endocrine therapy 7 years prior presented with right nipple erosion. Biopsy confirmed Paget's disease, and preoperative staging with contrast-enhanced computed tomography (CT) incidentally revealed a solitary soft-tissu

BACKGROUNDS: Castleman disease (CD) is a rare lymphoproliferative disorder. While its general clinical and pathological features are well-described, detailed data on retroperitoneal CD-a diagnostically challenging location-remain scarce. This study characterizes the clinicopathological profile, management, and long-term outcomes of retroperitoneal CD. The aim of this study is to clarify the clinical and therapeutic characteristics of retroperitoneal CD, and to provide practical evidence for its accurate diagnosis and individualized management in clinical practice.

BACKGROUND: Human immunodeficiency virus-associated multicentric Castleman disease (HIV-MCD), a human herpesvirus 8 (HHV-8)-driven lymphoproliferative disorder affecting individuals infected by HIV, typically exhibits a relapsing-remitting course and can be fatal. The incidence of HIV-MCD is on the rise while that of Kaposi's sarcoma, another HHV-8-associated disease, has been declining. Although an increasing number of studies have reported relapses of human herpesvirus-associated diseases and HIV-negative MCD following coronavirus disease 2019 (COVID-19), to the best of our knowledge there a

Idiopathic multicentric Castleman disease (iMCD) is a rare lymphoproliferative disorder characterized by systemic inflammation and lymphadenopathy. Two major clinical subtypes, idiopathic plasmacytic lymphadenopathy (iMCD-IPL) and iMCD with thrombocytopenia, anasarca, fever, renal dysfunction/reticulin fibrosis, and organomegaly (iMCD-TAFRO), have distinct pathophysiological mechanisms. While interleukin-6 (IL-6) is known to be elevated in iMCD, differences in the sources of IL-6 production between subtypes remain unclear. We examined the source of IL-6 production and its transcriptional regul

BACKGROUND: Although dual-drug immunosuppression is standard for rejection prophylaxis in pediatric heart transplant (HT), single-drug immunosuppression (monotherapy) may be appropriate for certain patients. We describe the experience with monotherapy in select patients at a single institution.
METHODS: Retrospective review of HT patients at a pediatric transplant center who underwent HT between 1/2001-12/2020 and were treated with calcineurin-inhibitor monotherapy for > 1 month. Patients were transitioned to monotherapy due to concerns for over-immune suppression, including recurrent infecti

OBJECTIVES: Castleman disease (CD) is a very rare B-cell lymphoproliferative disorder marked by the abnormal enlargement of lymph node tissue. It can present as either unicentric (UCD) or multicentric, with the former often appearing in intrathoracic regions, although its presence within the lungs is uncommon.
METHODS: We report the case of a 42-year-old woman who underwent resection of an 11 cm intrapulmonary UCD. Additionally, we conducted a systematic review of the demographics, clinical presentation, diagnosis, and treatment approaches for intrapulmonary UCD.
RESULTS: Our review identifi

Systemic chronic active Epstein-Barr virus disease (sCAEBV) is a chemotherapy-resistant, EBV-positive T- or NK-cell lymphoproliferative disorder characterized by persistent systemic inflammation driven by the activation of EBV-infected cells. In this study, we explored BCL2, an anti-apoptotic factor implicated in various hematopoietic malignancies, as a potential therapeutic target for sCAEBV, focusing on the effects of its inhibitor, venetoclax. We confirmed BCL2 expression in EBV-positive T- and NK-cell lines and peripheral blood mononuclear cells (PBMCs) from sCAEBV patients using western b
