Morphological evolution of the mammalian jaw adductor complex.

The evolution of the mammalian jaw during the transition from non-mammalian synapsids to crown mammals is a key event in vertebrate history and characterised by the gradual reduction of its individual bones into a single element and the concomitant transformation of the jaw joint and its incorporation into the middle ear complex. This osteological transformation is accompanied by a rearrangement and modification of the jaw adductor musculature, which is thought to have allowed the evolution of a more-efficient masticatory system in comparison to the plesiomorphic synapsid condition. While oste

Recent research on Gulf War illness and other health problems in veterans of the 1991 Gulf War: Effects of toxicant exposures during deployment.

Veterans of Operation Desert Storm/Desert Shield - the 1991 Gulf War (GW) - are a unique population who returned from theater with multiple health complaints and disorders. Studies in the U.S. and elsewhere have consistently concluded that approximately 25-32% of this population suffers from a disorder characterized by symptoms that vary somewhat among individuals and include fatigue, headaches, cognitive dysfunction, musculoskeletal pain, and respiratory, gastrointestinal and dermatologic complaints. Gulf War illness (GWI) is the term used to describe this disorder. In addition, brain cancer

Early motor deficits in mouse disease models are reliably uncovered using an automated home-cage wheel-running system: a cross-laboratory validation.

Deficits in motor function are debilitating features in disorders affecting neurological, neuromuscular and musculoskeletal systems. Although these disorders can vary greatly with respect to age of onset, symptomatic presentation, rate of progression and severity, the study of these disease models in mice is confined to the use of a small number of tests, most commonly the rotarod test. To expand the repertoire of meaningful motor function tests in mice, we tested, optimised and validated an automated home-cage-based running-wheel system, incorporating a conventional wheel with evenly spaced r…

"I know I have arthritis but I don't know what type". Understanding and knowledge of this chronic condition.

BACKGROUND: "Arthritis" is a common musculoskeletal condition but the knowledge of what type of arthritis people have, may be limited but may have changed over time in response to campaigns, increased awareness and improved health literacy. This paper describes people who did not know what type of arthritis they had, by a range of relevant demographic and socioeconomic variables, and assesses changes over time in the proportion of people who report having arthritis but do not know what type, using representative population surveillance data. METHODS: Data were collected using the South Austra

Congenital malformations in births with orofacial clefts among 3.6 million California births, 1983-1997.

Few population-based epidemiologic data are available on malformation phenotypes that co-occur with orofacial clefts. We explored the occurrence of structural congenital malformations in offspring with cleft palate (CP) and in offspring with cleft lip with or without CP (CLP), using data from a population-based active surveillance system. Ascertainment was performed among 3548991 liveborn and 23239 stillborn offspring of California women who delivered in nonmilitary hospitals during the period, 1983-1997. Structural congenital malformations were found in 91888 births. Among them, 2343 had CP a

Reconceptualizing Cerebral Palsy as a Lifelong Condition Characterized by Systemic and Accelerated Biological Decline: Implications for Physiatry Residency Education.

Cerebral palsy (CP), the most common childhood-onset disability worldwide, affects an estimated 50 million individuals globally. Although most individuals with CP are adults, CP education and training remain heavily concentrated within pediatric subspecialties, contributing to substantial gaps in adult care. At the same time, emerging evidence demonstrates that CP is associated with progressive multisystem, metabolic, musculoskeletal, and cellular changes across the lifespan, despite the original brain injury being nonprogressive. Current educational models, however, continue to frame CP prima

ULTRA-MoCap: A Multimodal IMU and sEMG Dataset for Upper Body Joint Kinematics Analysis.

Human upper limb movement prediction from multi-modal wearable sensors has applications in rehabilitation, sports analysis, assistive device control, and human-computer interaction, offering less intrusive alternatives to camera-based motion capture (MoCap) systems. Yet, current datasets often lack comprehensive multi-modal data integrating inertial measurement unit (IMU) and surface electromyography (sEMG) sensors and have limited coverage of upper limb kinematics with insufficient fidelity to capture isolated single-joint elbow motions and coordinated multi-joint shoulder movements. To addre

Multiscale Modeling and Systems Biology in Microgravity Investigations.

Spaceflight imposes a unique combination of environmental stressors including ionizing radiation, microgravity, and prolonged isolation that disrupt biological systems from the genome to the proteome. These stressors contribute to a range of health effects such as musculoskeletal degradation, neuroimmune dysfunction, and psychological challenges. Advances in countermeasure development, including biomedical devices and therapeutics, have already yielded terrestrial benefits for conditions like osteoporosis and cardiovascular disease. To systematically characterize spaceflight-induced biological

From Invisible to Visible: Cutting-Edge Ultrasound Insights into Entheses of the Distal Extremities in Rheumatology.

Musculoskeletal ultrasound (MSUS) is a well-established and reliable tool for the evaluation of entheses and enthesitis, particularly at larger and accessible sites. Recent technological advances, including high- and ultra-high-frequency transducers, have expanded its potential, enabling detailed assessment of distal extremity entheses. This narrative review provides a focused and updated perspective on this evolving field, highlighting three key advances. First, the identification and characterization of previously underrecognized entheseal sites in the distal extremities, such as pulley syst

The Potential Role of a Whole Food Plant-Based Diet on Fibromyalgia Symptoms: A Review Article.

Fibromyalgia is a complex, chronic disorder characterized by widespread musculoskeletal pain, fatigue, sleep disturbances, and cognitive dysfunction. While pharmacologic treatments remain the mainstay of symptom management, their long-term efficacy is limited and often accompanied by undesirable side effects. As a result, there is growing interest in non-pharmacologic interventions, particularly dietary therapies. Emerging literature suggests that whole food plant-based (WFPB) diets, which emphasize fresh fruits, vegetables, legumes, and whole grains while eliminating animal products and proce

Preclinical Rheumatoid Arthritis: Pathogenesis, Risk Stratification, and Therapeutic Interception.

Rheumatoid arthritis (RA) has traditionally been managed after the onset of clinically apparent synovitis; however, accumulating evidence indicates that disease-related immune abnormalities precede clinical diagnosis by several years. This preclinical phase is characterized by systemic autoimmunity, early musculoskeletal symptoms, and subclinical inflammation in genetically and environmentally susceptible individuals. In this review, we summarize current concepts regarding the pathogenesis, risk stratification, and therapeutic interception of preclinical RA. Particular attention is given to th

Scrub Typhus-Associated Inflammatory Oligoarthritis Mimicking Tubercular Arthritis in an Endemic Setting.

Scrub typhus is a common cause of acute febrile illness in endemic regions; however, musculoskeletal involvement is usually limited to myalgia, and true inflammatory arthritis is rarely reported. We describe a 19-year-old previously healthy male military recruit who presented with a two-month history of progressive bilateral knee swelling and left ankle pain, accompanied by fever, weight loss, and lymphadenopathy. Clinical evaluation revealed inflammatory oligoarthritis with culture-negative synovial fluid. Given the chronicity of symptoms and systemic features, tubercular and septic arthritis

TFNet: A Temporal-Frequency Domain Model for Gait Biomechanical Signal Prediction.

Precise prediction of upcoming gait signals, especially over an extended time scale like the entire gait cycle, is crucial. During this period, devices or gait retraining programs can respond to dynamic changes while considering multiple factors in the neurological and musculoskeletal systems. This enables effective adjustments, ultimately optimising outcomes based on the unique rehabilitation goals. However, current state-of-the-art models, whether driven by physical modelling or data modelling approaches, are constrained by short prediction time scales, limited accuracy, and high computation…

Antiobesity medications in rheumatology. Quo vadis?

Obesity is a well-recognised comorbidity in the context of rheumatic and musculoskeletal diseases (RMDs), adversely affecting disease-related outcomes. Adipose tissue, through immunological mechanisms, induces a low-grade inflammatory state; as a result, there has been growing interest in evaluating the potential role of antiobesity drugs in the management of RMDs. Although they were initially approved for type 2 diabetes mellitus, obesity, and their cardiorenal associations, there is increasing evidence that glucagon-like peptide-1 receptor agonists (GLP-1 RAs) in particular may exert immunom

Non-Hodgkin's lymphoma presenting with lower extremity edema and radiculopathy in a post-kidney transplant patient: A case report.

RATIONALE: Non-Hodgkin lymphoma (NHL) can present with vague and nonspecific symptoms, making early diagnosis challenging, especially in immunocompromised patients. Kidney transplant recipients, due to long-term immunosuppressive therapy, have an increased risk of developing malignancies, including NHL. PATIENT CONCERNS: A post-kidney transplantation patient presented with lower extremity symptoms. Initial lumbar spine magnetic resonance imaging suggested foraminal stenosis and disc herniation. However, persistent symptoms and lack of clinical improvement prompted further investigation. A con

Ultrasound Imaging and Frozen Shoulder: Unlocking the Benefits, Navigating the Risks.

Frozen shoulder (FS), or adhesive capsulitis, remains a challenging clinical entity due to its insidious onset, variable presentation, and diagnostic overlap with other shoulder pathologies. While FS is traditionally diagnosed clinically, musculoskeletal ultrasound (MSKUS) is emerging as a valuable adjunct tool capable of identifying early-stage structural and vascular changes. This review synthesizes the current evidence on MSKUS's diagnostic utility, highlighting its capacity to visualize characteristic features such as capsular thickening, rotator interval (RI) hypervascularity, and effusio

Atrogin-1 promotes muscle homeostasis by regulating levels of endoplasmic reticulum chaperone BiP.

Skeletal muscle wasting results from numerous pathological conditions affecting both the musculoskeletal and nervous systems. A unifying feature of these pathologies is the upregulation of members of the E3 ubiquitin ligase family, resulting in increased proteolytic degradation of target proteins. Despite the critical role of E3 ubiquitin ligases in regulating muscle mass, the specific proteins they target for degradation and the mechanisms by which they regulate skeletal muscle homeostasis remain ill-defined. Here, using zebrafish loss-of-function models combined with in vivo cell biology and
