Cryo-EM Structure Guided Engineering of Botulinum Neurotoxin A With Advanced Receptor Binding Affinity and Therapeutical Benefits.

Botulinum neurotoxin type A (BoNT/A) has been extensively used in treating a wide range of neurological disorders and aesthetics. However, insufficient binding affinity between BoNT/A and its receptor SV2C could lead to mild to severe side effects. An open active conformation of BoNT/A cryo-EM structure at a resolution of ∼2.85 Å was resolved. Guided by the BoNT/A cryo-EM structure, saturation mutagenesis libraries were constructed and screened by BACTH system to identify mutants that can boost toxin-SV2C affinity. The engineered toxin binding domain achieved up to approximately six-fold impro

Systematic Engineering of Proteases in Saccharopolyspora Spinosa Reveals Synergistic Enhancement of Spinosad Biosynthesis via Substrate Flux Optimization.

Natural product biosynthesis is tightly coupled to nitrogen allocation. However, scalable strategies to amplify pathway outputs remain limited. Here, atmospheric and room temperature plasma (ARTP) mutagenesis of Saccharopolyspora spinosa yielded a high-performing strain, D184, which showed improved utilization of extracellular nitrogen sources together with enhanced spinosad production. To identify novel regulatory mechanisms linking nitrogen utilization with natural product biosynthetic efficiency, we established a combinatorial engineering workflow that integrates systematic protease genetic…

Enhancing secretory expression of CSFV E2 glycoprotein via signal peptide engineering in a recombinant PRRSV vector platform.

Porcine Reproductive and Respiratory Syndrome (PRRS) and Classical Swine Fever (CSF) are major threats to swine production, emphasizing the need for effective bivalent vaccines. Using the attenuated Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) strain vHuN4-F112 as the backbone, we generated two recombinant viruses: rPRRSV-E2 (carrying an endogenous signal peptide and transcription regulatory sequence (TRS) element) and rPRRSV-E2-△SP (signal peptide-deleted but TRS-retained). We investigated how the signal peptide modulates the expression, secretion, structural features, and immu

Engineering synergistic VO-Co3+ pairs in CoOOH nanosheets for highly efficient and selective catalytic ozonation of CVOCs.

The catalytic activation of O3 on solid surfaces generates a spectrum of reactive oxygen species (ROS) with vastly different oxidative power. A central challenge lies in identifying surface sites that selectively produce highly oxidizing radicals over less reactive species, and in harnessing these potent ROS for efficient pollutant destruction. Here, we uncover a surface site dichotomy in cobalt oxides that governs O3 activation selectivity in the photo-enhanced catalytic ozonation of dichloromethane (DCM). Using CoOOH, Co3O4, and CoO as a model system, we demonstrate that O3 preferentially ad

Diels-Alder Click Chemistry as a Dynamic-Covalent Crosslinking Method in Spheroid-Encapsulating Hydrogels for Cartilage Engineering.

In cartilage tissue engineering, there is a growing interest in dynamic hydrogels that promote spheroid fusion and cartilaginous matrix deposition, while maintaining sufficient stability for long-term construct maturation. In this study, Diels-Alder click chemistry is employed as a dynamic-covalent crosslinking method to create hydrogels composed of hyaluronic acid, gelatin, and PEG. By adjusting the pH during crosslinking, the tuneability of hydrogel stiffness and stability at pH values around the physiological pH of native cartilage is demonstrated. This pH modulation does not compromise hyd…

Single-cell and longitudinal transcriptomics-guided engineering of FZD1-targeting precision nanotherapy against osteosarcoma cancer stem cells.

Osteosarcoma is the most prevalent primary malignant bone tumor in children and adolescents. Osteosarcoma cancer stem cells (OCSCs) critically drive tumorigenesis and progression through their self-renewal capacity and therapy resistance. Understanding mechanisms maintaining OCSC stemness and identifying key therapeutic targets are thus critical for effective intervention. Here, we systematically analyzed OCSC transcriptomic features using single-cell and longitudinal RNA-sequencing, identifying transcription factor 7 like 1 (TCF7L1)-associated transcriptional activity in relation to OCSC stem

Missing Landmark Estimation Using Reverse Engineering: Challenges and Potential Solutions for the Study of Hominin Long Bones.

Fragmentary preservation represents a fundamental limitation in palaeoanthropological research, particularly for postcranial elements where symmetry cannot be exploited and large portions of bone are frequently missing. Although a wide range of virtual reconstruction methods exist, many rely on strong anatomical priors, localized interpolation, or reference specimens that risk biasing reconstructions toward predefined morphologies. The present study introduces a new approach for estimating the coordinates of missing landmarks, termed the Reverse Engineering (RE) approach. This method establish…

Engineering cell ring organoids for efficient establishment of patient-derived orthotopic xenotransplantation (r-PDOX) model in sarcoma.

Patient-derived orthotopic xenotransplantation (PDOX) models have recently emerged as a potential approach for experimental therapeutics and precision medicine in malignant disease. However, the clinical application of PDOX models was restricted by the varying success rate of the animal models, long generation time, limited number of patient tumor cells and distinct stromal environments. To address this critical barrier, we developed an innovative patient-derived cell ring organoid platform using scaffold-free tumor cell rings. By co-culturing primary sarcoma cells with human skin fibroblasts

pKa engineering of a near-infrared fluorescent probe for sensitive imaging of oxidative stress in atherosclerotic plaques.

Reactive oxygen species (ROS) are key mediators in the progression of atherosclerosis, making ROS imaging valuable for evaluating oxidative stress in plaques. However, most activatable fluorescent probes show limited fluorescence enhancement under physiological conditions because their fluorophores are insufficiently ionized at neutral pH, resulting in weak signal output and low sensitivity. To address this issue, we developed a pKa-engineering strategy by introducing electron-withdrawing fluorine atoms at the ortho-positions of the phenolic hydroxyl group to construct the fluorophore DCM-2F-O…
