Prediction of pathological complete response to chemotherapy for breast cancer using deep neural network with uncertainty quantification.

BACKGROUND: The I-SPY 2 trial is a national-wide, multi-institutional clinical trial designed to evaluate multiple new therapeutic drugs for high-risk breast cancer. Previous studies suggest that pathological complete response (pCR) is a viable indicator of long-term outcomes of neoadjuvant chemotherapy for high-risk breast cancer. While pCR can be assessed during surgery after the chemotherapy, early prediction of pCR before the completion of the chemotherapy may facilitate personalized treatment management to achieve an improved outcome. Notably, the acquisition of dynamic contrast-enhanced

Limitations of panoramic radiographs in predicting mandibular wisdom tooth extraction and the potential of deep learning models to overcome them.

Surgeons routinely interpret preoperative radiographic images for estimating the shape and position of the tooth prior to performing tooth extraction. In this study, we aimed to predict the difficulty of lower wisdom tooth extraction using only panoramic radiographs. Difficulty was evaluated using the modified Parant score. Two oral surgeons (a specialist and a clinical resident) predicted the difficulty level of the test data. This study also aimed to evaluate the performance of a deep learning model in predicting the necessity for tooth separation or bone removal during wisdom tooth extracti

Artificial intelligence-guided design of lipid nanoparticles for pulmonary gene therapy.

Ionizable lipids are a key component of lipid nanoparticles, the leading nonviral messenger RNA delivery technology. Here, to advance the identification of ionizable lipids beyond current methods, which rely on experimental screening and/or rational design, we introduce lipid optimization using neural networks, a deep-learning strategy for ionizable lipid design. We created a dataset of >9,000 lipid nanoparticle activity measurements and used it to train a directed message-passing neural network for prediction of nucleic acid delivery with diverse lipid structures. Lipid optimization using neu

Deep learning of Parkinson's movement from video, without human-defined measures.

BACKGROUND: The core clinical sign of Parkinson's disease (PD) is bradykinesia, for which a standard test is finger tapping: the clinician observes a person repetitively tap finger and thumb together. That requires an expert eye, a scarce resource, and even experts show variability and inaccuracy. Existing applications of technology to finger tapping reduce the tapping signal to one-dimensional measures, with researcher-defined features derived from those measures. OBJECTIVES: (1) To apply a deep learning neural network directly to video of finger tapping, without human-defined measures/featu

AI-Assisted Identification of Primary and Secondary Metabolomic Markers for Postoperative Delirium.

Despite considerable investigative efforts, the molecular mechanisms of postoperative delirium (POD) remain unresolved. The present investigation employs innovative methodologies for identifying potential primary and secondary metabolic markers of POD by analyzing serum metabolomic profiles utilizing the genetic algorithm and artificial neural networks. The primary metabolomic markers constitute a combination of metabolites that optimally distinguish between POD and non-POD groups of patients. Our analysis revealed L-lactic acid, inositol, and methylcysteine as the most salient primary markers

Precise grading of non-muscle invasive bladder cancer with multi-scale pyramidal CNN.

The grading of non-muscle invasive bladder cancer (NMIBC) continues to face challenges due to subjective interpretations, which affect the assessment of its severity. To address this challenge, we are developing an innovative artificial intelligence (AI) system aimed at objectively grading NMIBC. This system uses a novel convolutional neural network (CNN) architecture called the multi-scale pyramidal pretrained CNN to analyze both local and global pathology markers extracted from digital pathology images. The proposed CNN structure takes as input three levels of patches, ranging from small pat

Bioinspired Iron Porphyrin Covalent Organic Frameworks-Based Nanozymes Sensor Array: Machine Learning-Assisted Identification and Detection of Thiols.

Given the crucial role of thiols in maintaining normal physiological functions, it is essential to establish a high-throughput and sensitive analytical method to identify and quantify various thiols accurately. Inspired by the iron porphyrin active center of natural horseradish peroxidase (HRP), we designed and synthesized two iron porphyrin covalent organic frameworks (Fe-COF-H and Fe-COF-OH) with notable peroxidase-like (POD) activity, capable of catalyzing 3,3',5,5'-tetramethylbenzidine (TMB) into oxidized TMB with three distinct absorption peaks. Based on these, a six-channel nanozyme colo

Enhanced Osteoporosis Detection Using Artificial Intelligence: A Deep Learning Approach to Panoramic Radiographs with an Emphasis on the Mental Foramen.

Osteoporosis, a skeletal disorder, is expected to affect 60% of women aged over 50 years. Dual-energy X-ray absorptiometry (DXA) scans, the current gold standard, are typically used post-fracture, highlighting the need for early detection tools. Panoramic radiographs (PRs), common in annual dental evaluations, have been explored for osteoporosis detection using deep learning, but methodological flaws have cast doubt on otherwise optimistic results. This study aims to develop a robust artificial intelligence (AI) application for accurate osteoporosis identification in PRs, contributing to early

Machine learning-based diagnostic prediction of minimal change disease: model development study.

Minimal change disease (MCD) is a common cause of nephrotic syndrome. Due to its rapid progression, early detection is essential; however, definitive diagnosis requires invasive kidney biopsy. This study aims to develop non-invasive predictive models for diagnosing MCD by machine learning. We retrospectively collected data on demographic characteristics, blood tests, and urine tests from patients with nephrotic syndrome who underwent kidney biopsy. We applied four machine learning algorithms-TabPFN, LightGBM, Random Forest, and Artificial Neural Network-and logistic regression. We compared the

Reproducible and clinically translatable deep neural networks for cervical screening.

Cervical cancer is a leading cause of cancer mortality, with approximately 90% of the 250,000 deaths per year occurring in low- and middle-income countries (LMIC). Secondary prevention with cervical screening involves detecting and treating precursor lesions; however, scaling screening efforts in LMIC has been hampered by infrastructure and cost constraints. Recent work has supported the development of an artificial intelligence (AI) pipeline on digital images of the cervix to achieve an accurate and reliable diagnosis of treatable precancerous lesions. In particular, WHO guidelines emphasize
