Recurrent implantation failure in assisted reproduction: how to counsel and manage. A. General considerations and treatment options that may benefit the couple.

Recurrent implantation failure is a distressing phenomenon, both for the infertile couple and for the physician responsible for their treatment. Aetiology is often not clear and treatment options are vague. Particularly when transferred embryos are of good quality, recurrent implantation failure may be attributed to less than optimal embryo transfer technique, pathological lesions of the uterine cavity, the presence of hydrosalpinges, fibroids and endometriosis. Poor embryo quality, especially when repetitive, is a major impediment to successful implantation and cannot be corrected at the pres

Clinical signs, reproduction of attaching/effacing lesions, and enterocyte invasion after oral inoculation of an O118 enterohaemorrhagic Escherichia coli in neonatal calves.

Attaching and effacing (AE) lesions are produced among others by enteropathogenic Escherichia coli and enterohaemorrhagic E. coli (EHEC), which differs from the former by the production of cytotoxins active on various cell cultures, the verocytotoxins, or shigacytotoxins. EHEC are associated with diarrhoea and dysentery in humans and in ruminants, mainly calves from two to eight weeks of age. Clinical signs and/or lesions have been reproduced experimentally with EHEC strains belonging to serotypes O5:K4/Nm, O26:K-:H11, O111:Nm, and O157:H7 which are isolated from cattle and/or humans. The purp

Exposure to different spectra of light, continuous light and treatment with melatonin affect reproduction in the Indian desert gerbil Meriones hurrianae (Jerdon).

In two separate experiments, the gonadal response to melatonin treatment and exposure to continuous white light (LL) and different spectra o … inealocytes and pinealocyte nuclear diameters. Therefore, the influence of light and its spectra on reproduction may be mediated by the pineal gland, though extrapineal mechanisms cannot be ruled out.

Characteristics of GnRH binding in the gonads and effects of lamprey GnRH-I and -III on reproduction in the adult sea lamprey.

In the present study, both lamprey GnRH-I and -III stimulated steroidogenesis and induced ovulation in adult female sea lampreys during their final reproductive stage. One injection of lamprey GnRH-III at 0.1 or 0.2 microg/g lamprey stimulated plasma estradiol levels in lampreys held at each of three water temperatures, 13 degrees , 17 degrees , and 19 degrees , corresponding to increasing stages of maturation. Four successive injections, 3 to 4 days apart, of lamprey GnRH-III at 0.1 or 0.2 microg/g body weight induced ovulation in 100 or 88% of lampreys, respectively, compared to 21% in contr

Inositol phosphate formation in uterine tissue from two species of reptiles is stimulated by arginine vasotocin and influenced by stage of reproduction.

Phosphoinositide hydrolysis, resulting in inositol trisphosphate (IP3) and diacylglycerol (DG) formation, has been implicated in oxytocin-stimulated pulsatile secretion of prostaglandin F2 alpha (PGF2 alpha) from uterine endometrium of sheep and other mammals. In reptiles, arginine vasotocin (AVT) stimulates uterine secretion of PGF2 alpha. These studies investigated the ability of AVT to stimulate incorporation of [3H]inositol into inositol mono-, bis-, and trisphosphates in two reptilian species. In Experiment 1, AVT stimulated (P less than 0.01) IP formation in uterine from late-gravid (150

Metabolic interfaces between growth and reproduction. I. Nutritional modulation of gonadotropin, prolactin, and growth hormone secretion in the growth-limited female lamb.

The acute and long term effects of dietary restrictions on gonadotropin secretion were studied in ovariectomized female lambs. Nutritionally growth-restricted lambs which were chronically maintained at a body weight comparable to that at weaning (approximately 20 kg) became hypogonadotropic, exhibiting a low frequency of episodic LH discharges. Repeated administration of physiological doses of GnRH to these females at hourly intervals produced corresponding LH pulses, leading to the hypothesis that the dietary-induced hypogonadotropism arises from a deficiency in endogenous GnRH release, rathe

Reproductive Function in Thyroid Cancer Survivors: A Gender-Based Perspective on Outcomes.

BACKGROUND: Thyroid cancer is one of the most common endocrine malignancies, with an excellent long-term survival rate. As survivorship increases, attention has shifted toward chronic endocrine sequelae, including disturbances of gonadal function and fertility. Thyroid cancer therapies, including thyroidectomy, long-term thyroid-stimulating hormone (TSH) suppressive therapy, radioiodine (RAI), and targeted systemic treatments, may interfere with reproductive health through complex endocrine and gonadal mechanisms. OBJECTIVE: This narrative review aims to examine the pathophysiological pathway

Pregnancy from implantation: clinical utility and residual ambiguity in reproductive terminology.

The 2025 International Glossary on Infertility and Fertility Care defines pregnancy as 'a state of reproduction beginning with implantation of an embryo and ending with the complete expulsion and/or extraction of the products of implantation'. This implantation-based definition is clinically useful, particularly for assisted reproductive technology ART, registries and outcome reporting. However, it also leaves a biologically relevant interval - between fertilization and implantation - outside the term 'pregnancy'. This interval is not a semantic void: it includes fertilization, cleavage, blast…

Semaglutide Reduces Odorant-Driven Chemotaxis and Reproductive Output in Caenorhabditis elegans.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) reduce food intake and body weight in humans, partly via neural circuits governing reward and energy homeostasis. Using Caenorhabditis elegans as a tractable model, we examined neurobehavioral and physiological effects of semaglutide, a long-acting GLP-1RA. Wild-type N2 animals exposed to semaglutide (1 μM and 10 μM) showed significantly reduced chemotaxis toward the volatile odorant diacetyl. Semaglutide also reduced egg-laying and delayed larval development at both concentrations tested. These phenotypes demonstrate that semaglutide alters

Impact of Bariatric Surgery on Reproductive Hormones and Menstrual Regularity in Women with Obesity.

BACKGROUND: Obesity affects 2.5 billion adults, disrupting the hypothalamic-pituitary-ovarian (HPO) axis through adipokine imbalance and aromatase activity. This causes hormonal dysregulation, infertility, and pregnancy risks. Bariatric surgery can restore hormonal balance and improve menstrual regularity. This study evaluates post-bariatric surgery changes in reproductive hormones and cycle regularity over six months in Egyptian women with obesity. METHODS: This prospective cohort study included women aged 18-45 (BMI ≥ 35 kg/m²) undergoing laparoscopic bariatric surgery. Serum FSH, LH, estra

Jasmonate biosynthesis regulates key events in barley vegetative and reproductive development.

Cereal grain is a global food staple. Filial tissues in grain, such as the embryo and endosperm, grow embedded in the maternal ovule and surrounding ovary wall. Maternal tissues provide nutrition and protection but are progressively reduced and degraded as filial tissues expand, driving grain expansion between floret hulls. While we know some enzymatic players, regulation of this process remains less understood. However, our previous work in barley, Hordeum vulgare spp. vulgare suggested a role for the phytohormone jasmonate (JA). We edited the barley ALLENE OXIDASE CYCLASE (HvAOC) gene encodi

Lysine succinylation: a review of its impacts on the reproductive function and related disease.

Lysine succinylation is a recently identified post-translational modification of proteins that has been demonstrated to play critical roles in various biological activities. In recent years, research on succinylation has progressed considerably in the context of reproductive biology and associated diseases, underscoring the importance of a comprehensive review on this topic. A key mechanism underlying succinylation involves changes in the activity of succinyltransferases and desuccinylases, which in turn regulate alterations in the succinylation level of target proteins, and thereby influence

CSF3R signalling beyond granulopoiesis: new paradigms in female reproductive biology.

The Colony-Stimulating Factor 3 Receptor (CSF3R), the receptor for granulocyte colony-stimulating factor (G-CSF), has expanded from a hematopoietic regulator to a multifunctional signalling molecule with significant relevance in gynaecological biology. High-resolution transcriptomic and proteomic studies now identify CSF3R expression in ovarian granulosa cells, endometrial epithelium, cervical tissue, placental trophoblasts, and gynaecologic tumour microenvironments, highlighting its integration into immune, endocrine, and tissue-remodeling networks. Structurally anchored by Box1/Box2 motifs,

Ceramide: a pivotal lipid mediator shaping female reproductive homeostasis and pathophysiology.

Ceramide, a central hub in sphingolipid metabolism, serves as a critical bioactive signalling mediator that integrates lipid metabolism with multiple signalling pathways, primarily promoting autophagy and apoptosis. It influences female reproductive homeostasis and pathophysiology through multiple dimensions: altered ceramide levels have been observed in various reproductive disorders-such as polycystic ovary syndrome (PCOS), premature ovarian insufficiency (POI) and endometriosis (EM)-as well as across the female lifespan. These changes impact key processes including gonadotropin release, fol

Phytochemicals as eco-friendly modulators of ovarian vitellogenesis and reproductive efficiency in aquaculture.

The search for natural alternatives to chemical additives and artificial hormones to enhance reproductive function in aquatic animals is driven by demand for eco-friendly aquaculture. This review explores how bioactive plant compounds can enhance ovarian vitellogenesis and reproductive efficiency in crustaceans and finfish. Vitellogenesis is regulated by the hypothalamic-pituitary-gonadal axis, controlling yolk protein deposition and embryogenesis. While synthetic hormones have significant effects, their environmental and health impacts have sparked interest in phytochemicals such as carotenoi
