From lignin to living systems: Carbon dots modulate early cytoskeletal organization and phenotypic marker expression in undifferentiated mesenchymal stem cells.

PURPOSE: Carbon dots (CDs) are emerging bioactive nanomaterials for tissue engineering applications. This study aimed to reproduce softwood lignin-derived carbon dots (LCDs) and nitrogen-doped LCDs (N@LCDs) and evaluate their effects on early human adipose-derived mesenchymal stem cell (hMSC) behavior in 2D monolayer and 3D culture systems. METHODS: LCDs/N@LCDs were reproduced and characterized. CDs were ultrasonically sprayed onto 2D substrates and polyethylenimine (PEI)-treated electrospun poly(lactic-co-glycolic acid) (PLGA, 65:35) 3D scaffolds for in vitro analyses. Distribution after dro…

Arginine-substituted Mastoparan-C derivatives combat dual bacterial pathogens: in vitro mechanistic insights and in vivo efficacy in polymicrobial wounds.

UNLABELLED: The synergistic interactions in multi-pathogen infections compromise wound healing and limit therapeutic efficacy. In this study, we designed and synthesized arginine-substituted derivatives of the antimicrobial peptide Mastoparan-C (MP-C). Among them, Arg²MP-C and Arg4.11.12MP-C exhibited potent, broad-spectrum activity against both Escherichia coli and Staphylococcus aureus. Their enhanced antibacterial activity is associated with increased positive charge and optimized hydrophobicity. Mechanistically, both peptides employ a dual-target strategy, disrupting bacterial membranes an

Nanoparticle-Mediated Drug Delivery for Treatment of Ischemic Heart Disease.

The regenerative capacity of an adult cardiac tissue is insufficient to repair the massive loss of heart tissue, particularly cardiomyocytes (CMs), following ischemia or other catastrophic myocardial injuries. The delivery methods of therapeutics agents, such as small molecules, growth factors, exosomes, cells, and engineered tissues have significantly advanced in medical science. Furthermore, with the controlled release characteristics, nanoparticle (NP) systems carrying drugs are promising in enhancing the cardioprotective potential of drugs in patients with cardiac ischemic events. NPs can

Emulsion-based synthesis of polycaprolactone/bioactive glass 45S5 microparticles for bone regeneration applications.

Polycaprolactone (PCL) and bioactive glass 45S5 (BG) are widely employed within bone tissue engineering due to their mutually complementary properties: PCL contributes structural integrity and biodegradability, whereas BG imparts osteoinductive properties to the composite. While patient-specific bulk scaffolds can restore structural defects, their fabrication requires individualized imaging, design, and manufacturing, increasing both time and cost. Hydrogels offer better conformity to irregular geometries but often lack sufficient mechanical stability and usually degrade rapidly. Microparticle…

Graphene and Ti3C2Tx MXene Nanomaterial-Infused Bioinks for Mechanical and Electrical Improvement of 3D Bioprinted Scaffolds.

The precise control of bioink formulation, process parameters, and scaffold geometries has enabled some critical limitations of bioprinting to be overcome and has transformed the construction of functional 3-dimensional (3D) bioscaffolds for tissue engineering (TE). Current bioscaffolds fall short in producing functional tissue due to a lack of the physiological, mechanical, and electrical environments necessary for optimal cell development. In this study, we report the incorporation of the nanomaterial (NM) graphene/graphene oxide (GGO) and titanium carbide (Ti3C2Tx MXene) into an alginate-ce…

In vivo bioengineered tooth formation using decellularized tooth bud extracellular matrix scaffolds.

The use of dental implants to replace lost or damaged teeth has become increasingly widespread due to their reported high survival and success rates. In reality, the long-term survival of dental implants remains a health concern, based on their short-term predicted survival of ~15 years, significant potential for jawbone resorption, and risk of peri-implantitis. The ability to create functional bioengineered teeth, composed of living tissues with properties similar to those of natural teeth, would be a significant improvement over currently used synthetic titanium implants.
