The emergence of antimicrobial resistance (AMR) poses a significant threat to global health and food security. Antimicrobial peptides (AMPs), particularly those characterised by α-helical structures, represent a promising alternative due to their broad-spectrum activity and unique mechanisms of action. Pectobacterium brasiliense is a destructive bacterial pathogen affecting solanaceous crops but effective control measures remain insufficient. This study aims to optimise pseudothionin AMPs using AI-based protein language models, ESM-3 and ESMFold, to enhance their antibacterial efficacy. Using

Betulin, a lupane-based pentacyclic triterpenoid, is renowned for its sustainability and potent antifungal properties. By leveraging a spontaneous amino-yne reaction in conjunction with betulin, novel biomolecules representing a diverse library of triterpenoid derivatives are designed and synthesized for potential antifungal development. In this work, betulin was initially transformed into 28-propynoylbetulin via a traditional DCC coupling reaction. Subsequently, mono- and di-β-aminoacrylated betulin derivatives were synthesized by reacting betulin with various secondary (di)amines via an amin

During 2022-2023, the Antimicrobial Resistance Laboratory Network tested 8,033 Candida auris clinical isolates in the United States. Overall, 95% of isolates were fluconazole resistant, 15% amphotericin B resistant, and 1% echinocandin resistant. Laboratory capacity for C. auris identification and antifungal susceptibility testing is essential to address this emerging public health threat.

In 2022, the World Health Organization published a list highlighting Candida auris and Cryptococcus neoformans as among the priority pathogens in need of new therapeutic alternatives. The essential oil of Cymbopogon citratus (EOCC) is used in folk medicine for its various properties, including antimicrobial activity. However, there are few reports of its activity against these yeasts and the possible action mechanism. EOCC was characterized by gas chromatography combined with mass spectrometry. Antifungal activity was determined by broth microdilution against C. parapsilosis ATCC 22,019, C. kr

The clinical deployment of antibiotics is undermined by antimicrobial resistance. Without new agents to treat antibiotic-resistant bacterial infections, mortality rates are predicted to reach 10 million people per year by 2050. Most antibiotics are derived from natural products (NPs) produced by bacteria; however, this resource was abandoned by industry because of high rediscovery rates. We are amid a natural product renaissance fuelled by inexpensive access to genome sequencing and sophisticated bioinformatic tools, which have highlighted that most of the biosynthetic pathways for NPs are not

The clinical efficacy of existing antibiotics is threatened by the emergence of multidrug-resistant pathogens. In India, traditional herbal medicine has attracted much of the scientific interest and increasing continuously. This study evaluates the antimicrobial activity of crude plumbagin extract (CRPB) from Plumbago auriculata and standard plumbagin (PB) (which is a 98% pure compound from Sigma Aldrich, USA) against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Klebsiella pneumoniae, Citrobactor, and their MDR strains with disc diffusion assay and minimum inhibitory concen

BACKGROUND: Repeated exposure to sub-minimum inhibitory concentrations (sub-MICs) of antimicrobials can impose selective pressure on bacterial pathogens, potentially promoting adaptive responses beyond classical resistance. This study investigated whether sequential sub-MIC exposure, followed by biofilm derivation, is associated with resistance development and phenotypes consistent with reduced bactericidal susceptibility in Aeromonas hydrophila KCTC 2358.
METHODS AND RESULTS: A parental population (R0) was subjected to ten cycles of sub-MIC exposure to generate an adapted population (R10), f

Fluoroquinolone antibiotics, like danofloxacin, are considered as crucial veterinary drugs due to their high antibacterial potential, a broad spectrum of activity and good pharmacological properties. However, owing to the widespread use of this group of pharmaceuticals, microbial resistance to them is becoming a serious worldwide concern. In the present study, novel silver and copper complexes of danofloxacin were prepared and characterized using 1H NMR, 19F NMR and IR spectroscopy, ESI-MS spectrometry, and elemental analysis. The antimicrobial properties of the obtained complexes were determi

UNLABELLED: Chemical fungicides act by targeting specific proteins or pathways, known as modes of action (MoAs). In this study, we established a systematic screening platform using Fusarium graminearum gene knockout mutants to evaluate the efficacy of fungicides and identify novel antifungal compounds. Target genes corresponding to major fungicide classes were deleted by homologous recombination, and the resulting mutants exhibited altered sensitivity to the corresponding fungicides compared with the wild-type strain. Deletion of genes encoding known fungicide targets revealed distinct sensiti

Six zinc(II) carboxylate complexes were synthesized under reflux reactions and thoroughly characterized by single crystal X-ray diffraction, Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and elemental analysis. They adopt to two general formulas: mononuclear [Zn(DPAA)2(H2O)2] (1) and binuclear [Zn₂(DPAA)4(L)2] (2-6), where DPAA = diphenyl acetate anion and L represents an ancillary ligand. L used are pyridine (py, 2), 2-chloropyridine (2-chloro-py, 3), 2-methylpyridine (2-methyl-py, 4), 2-aminopyidine (2-amino-py, 5

Antimicrobial resistance in pathogenic mycobacteria remains a critical challenge due to poor drug penetration through their complex cell wall, which necessitates prolonged multidrug regimens. Mycobacteriophages encode a lytic machinery that can disrupt this barrier. In this research article, we describe a modular mycolysin platform combining phage enzymes Lysin A and Lysin B with outer membrane-permeabilizing peptides and protein transduction domains using VersaTile shuffling technology. Screening the chimeric libraries against Mycobacterium smegmatis and Mycobacterium bovis Bacillus Calmette-

Mycoplasma iowae is an economically significant pathogen that causes reduced hatchability, late embryo mortality and leg deformities, chondrodystrophy and skeletal lesions in poults. While prevention is essential in the control of infection, the appropriate administration of antibiotics may reduce economic losses during outbreaks. As a first step in the exploration of antimicrobial resistance mechanisms in M. iowae, target modification and efflux pump activity were examined in the present study. Point mutations were analyzed in previously described antibiotic binding sites in the whole genome

Chitosan, a chitin-derived biopolymer, is widely used in agriculture and food preservation. Fungal-derived chitosan offers a sustainable alternative to crustacean sources, and Ganoderma lucidum represents a promising source of this functional material. This study characterized fungal chitosan (F.Cs) extracted from G. lucidum cell-wall chitin by FT-IR, molecular weight, viscosity, and degree of deacetylation. Its antimicrobial activity was evaluated in vitro, and its preservative and disease control efficacy against Monilinia fructicola on postharvest apples was compared to commercial chitosan

Hierarchically restructured platinum-iridium electrodes offer high electrochemical performance for neurostimulation and cardiac rhythm management devices but require added antibacterial functionality to reduce postsurgical infection risks. In this work, electrochemically active antibacterial platinum-iridium electrodes were developed using a two-step process. First, the electrodes were restructured using a femtosecond laser hierarchical surface restructuring. In the second step, reactive magnetron sputtering from a pure zinc target in an Ar/O2 gas mixture was employed to deposit antibacterial

Four new limonoids, designated xylomoluccenoids A-D (1-4), together with six known compounds (5-10), were isolated from the leaves of the mangrove plant Xylocarpus moluccensis. The structures and absolute configurations of the new metabolites were elucidated through extensive HRESIMS and NMR spectroscopic analyses, supported by quantum chemical calculations of their NMR chemical shifts and UV/ECD spectra. Biological evaluation revealed that compounds 1, 2, and 5 possess weak cytotoxic activity, while compounds 5-10 displayed weak antimicrobial activity against the tested bacterial and fungal s

The genus Plectranthus sensu lato is known to produce abietanes, which exhibit significant potency against Gram-positive bacteria. It is hypothesized that abietanes exert their antibacterial effects by disrupting microbial cell walls. Recent studies have classified Plectranthus s.l. abietanes into six distinct groups. However, the impact of this chemical diversity on the response of abietanes against Gram-positive bacteria remains unexamined. Therefore, a structure-activity relationship study on abietanes from species of Plectranthus sensu stricto and Coleus was conducted to identify active fr

Five undescribed cyclopiazonic acid indole alkaloids (CPAs), penibrasdines A - E (1-5), and eleven known analogues were isolated from the fungus Penicillium brasilianum. Their structures and configurations were elucidated based on HRESIMS, 1D and 2D NMR spectroscopic data, ECD calculations, and single-crystal X-ray diffraction. Penibrasdine A (1) bearing a rigid and sterically congested hexacyclic indole-tetrahydrofuran scaffold, is the epimer of penicillamine A. Bioactivity evaluation indicated that all isolated compounds showed no effective antibacterial activity against the tested bacterial
