Chronic histiocytic intervillositis: Insights into immunopathogenesis and prospective treatments.

Chronic histiocytic intervillositis (CHI) is a rare placental inflammatory disorder associated with adverse pregnancy outcomes, including fetal growth restriction, miscarriage, and stillbirth. Its diagnosis is made retrospectively via histopathology, limiting timely intervention. This review explores the hypothesis that CHI is an alloimmune condition resulting from aberrant maternal immune responses to paternally derived fetal antigens. Key findings from current literature include perivillous fibrin deposition and maternal mononuclear cell accumulation in the placental intervillous space, both

Impaired embryogenesis following intracytoplasmic sperm injection with an Arrdc5-/- mouse model of oligoasthenoteratospermia†.

Oligoasthenoteratospermia (OAT) describes male infertility due to a combination of low sperm count, reduced sperm motility, and abnormal sperm morphology. A common course of action to overcome infertility for males with this condition is intracytoplasmic sperm injection (ICSI). Here we used the Arrdc5 knockout mouse model of OAT to examine the ramifications of bypassing natural barriers preventing fertilization by defective sperm via ICSI on the viability of embryos and health of offspring. ARRDC5 is expressed specifically in the male germline where it has an essential role in sperm morphogene

Blood Lead Levels and Associated Sociodemographic and Occupational Factors in Children from Two Mexican Communities: A Comparative Study.

Lead is a neurotoxic metal. No safe blood lead level (BLL) has been identified in children; the CDC reference value was lowered to 3.5 µg/dL in 2021, while Mexico's regulatory threshold remains at 10 µg/dL. Lead-glazed earthenware is the main documented source of pediatric exposure in Mexico. We studied children 0-15 years insured by the Mexican Social Security Institute (IMSS) in two Jalisco municipalities: Tonalá (n = 500), historically a ceramic-producing center, and Zapopan (n = 374), an urban area without this tradition. BLL was measured by graphite furnace atomic absorption spectrophotom

Differential Transcriptome Analysis of Intrauterine UPD6pat Fetuses With Distinct Phenotypes.

OBJECTIVE: Uniparental disomy of chromosome 6, typically paternal in origin (UPD6pat), results in significant phenotypic variability across different cases. This study integrates transcriptome-wide profiling of amniotic fluid cell-free RNA (cfRNA) with molecular diagnostics to elucidate the gene expression landscape and pathway dysregulation in two phenotypically distinct UPD6pat cases. METHODS: RNA sequencing (RNA-seq) was performed on amniotic fluid supernatants from two UPD6pat fetuses and two gestational age-matched controls. RESULTS: Differential expression analysis revealed 372 signifi

Gillespie syndrome caused by a novel ITPR1 gene variant: a phenotype-genotype correlation.

BACKGROUND: Gillespie syndrome (GS) is a rare congenital disorder with ocular and neurological defects. Classically, it has been noted to have a triad of cerebellar ataxia, variable absence of iris but always with a scalloped edge and intellectual disability. This scalloped iris edge is essential to make a diagnosis of GS. Both autosomal dominant and recessive inheritance have been reported with de novo pathogenic variants in the inositol 1,4,5-trisphosphate receptor type 1 gene (ITPR1). We report a novel variant of ITPR1 gene causing Gillespie syndrome in multiple members of a family. MATERI

Maternal Overnutrition and Fetal Programming: Long-Term Metabolic, Cognitive, and Epigenetic Consequences.

Maternal nutrition during pregnancy critically influences fetal programming, shaping the offspring's lifelong health and disease susceptibility. Both undernutrition and overnutrition affect fetal metabolism, predisposing offspring to obesity and cardiometabolic disorders in adulthood. This review examines current evidence on how maternal nutrition, particularly overnutrition and its complications, such as gestational diabetes mellitus (GDM) and obesity, affects offspring health. It also explores the biochemical and epigenetic mechanisms underlying aberrant fetal programming induced by an unfav

The Inheritance Puzzle: A Case of Dual Genetic Kidney Disease.

Autosomal Dominant Polycystic Kidney Disease (ADPKD) and X-Linked Alport Syndrome (AS) are the most common monogenic causes of chronic kidney disease (CKD), each capable of independently leading to end-stage kidney disease (ESKD). However, the concurrent presence of both disorders in a single individual is exceedingly rare and poses significant diagnostic complexity. We report a unique case involving dual diagnosis of ADPKD and AS, attributed to a paternal PKD1 variant and a maternally inherited COL4A5 variant. Initial genetic testing through next-generation sequencing (NGS) identified PKD1 an

Sibling Osteosarcoma Without Retinoblastoma Associated With a Low Penetrance RB1 Variant: Whole Genome Findings From a Single Family.

PURPOSE: Osteosarcoma is the most common primary malignant bone tumor in children and adolescents, yet its genetic etiology remains poorly understood. In this study, we described a family from the Shanghai General Hospital Osteosarcoma (SGH-OS) cohort in which two siblings developed osteosarcoma as their first primary malignancy, and we investigated the germline and somatic genetic basis underlying this familial presentation. METHODS: Whole-genome sequencing (WGS) was performed on tumors and matched germline DNA from the affected siblings, as well as germline DNA from their parents and unaffe

Somatic mosaicism in the δ-aminolevulinate dehydratase gene causing late-onset porphyria with erythroid-driven pathogenesis.

δ-Aminolevulinate dehydratase deficiency porphyria (ADP) is an extremely rare autosomal recessive disorder, with only 12 previously reported cases. It is characterized by the accumulation of delta-aminolevulinic acid (ALA), which is associated with neurovisceral symptoms. We report the first case of ADP in Russia with late-onset symptoms and unique genetic findings. A 47-year-old Russian woman experienced recurrent abdominal pain, tachycardia, hypertension, and neuropathy onset at age 38. Biochemical tests revealed a marked and persistent elevation of urinary ALA, whereas PBG levels showed onl

Prenatal diagnosis of apert syndrome at 23 weeks: a case report with isolated syndactyly and sacrococcygeal appendage preceding cranial malformation.

BACKGROUND: Apert syndrome is a rare congenital malformation caused by a mutation in the fibroblast growth factor receptor 2 (FGFR2) gene. Characteristic imaging findings include bilateral premature closure of the coronal suture, midfacial hypoplasia, and symmetrical syndactyly of the fingers and toes. These features may be accompanied by lateral ventricle dilation, absence of the corpus callosum, cervical vertebral fusion, and other anomalies. However, in the second trimester, syndactyly and extracranial anomalies may be the only detectable signs, which complicate early diagnosis. The purpose

Immunology of Normal Pregnancy and Preeclampsia and the Role of Placental Non-Classical HLA and Decidual NK Cells.

Pregnancy is considered a unique immunological process in which contact between the maternal innate immune system and the placental allograft results in maternal tolerance to paternally derived antigens. A growing body of research indicates that interactions between non-classical placental Human Leukocyte Antigen (NCHLA) and receptors on decidual Natural Killer (dNK) cells from early implantation are crucial to this process. Preeclampsia (PE), one of the leading causes of maternal and fetal morbidity and mortality, is also considered an autoimmune process. Currently, there is no treatment for

Expansion of the allelic and phenotypic spectrum of MED25-related developmental disorder: novel compound heterozygous variants with structural domain implications.

MED25-related developmental disorder (Basel-Vanagaite-Smirin-Yosef syndrome) is a rare autosomal recessive disorder, defined by severe neurodevelopmental delay, corpus callosum abnormalities, ocular involvement, epilepsy, and marked facial appearance. MED25 pathogenic variants interfere with the functioning of the Mediator complex, which is responsible for RNA polymerase II transcription. We report a 9-year-old girl who presents with significant global developmental delay, agenesis of the corpus callosum, congenital cataracts, epilepsy, hypotonia, musculoskeletal abnormalities, and typical cra

Case report: novel DNAH11 compound heterozygous variants including an exon 30-54 duplication in a child with a highly suggestive primary ciliary dyskinesia phenotype.

Primary ciliary dyskinesia (PCD) is a rare disorder characterized by dysfunction of motile cilia and chronic progressive respiratory disease, mianly inherited in an autosomal recessive manner. Biallelic variants in dynein axonemal heavy chain 11 (DNAH11) have been reported in association with PCD and are typically associated with normal ciliary ultrastructure. In addition, exon-level copy number variants (CNVs) in DNAH11, particularly duplications, remain poorly characterized and are prone to being overlooked by conventional sequencing workflows. We report a 9-year-old Chinese girl with recurr

40+ years of the Huntington disease predictive genetic testing protocol: Recommendations for an update.

Guidance for predictive genetic testing for Huntington disease (HD) has been available for more than four decades and has been applied to other adult-onset hereditary conditions, especially neurogenetic conditions. Yet this protocol has not been updated since 2016 even though there have been significant changes in healthcare delivery and policy/legal issues, including widespread use of telemedicine and new legislation impacting patients' ready access to medical records. There also have been technological advances where HD status can be revealed with genomic sequencing and societal paradigm shi

Unraveling the Roles of UBE3A in Neurodevelopment and Neurodegeneration.

The ubiquitin-protein ligase E3A (UBE3A, aka E6-AP), an E3 ligase belonging to the HECT family, plays crucial roles in the stability of various proteins through the proteasomal degradation system. Abnormal UBE3A activity is essential for the initiation and progression of several cancers. A gain of function and an overdosage of maternal UBE3A is associated with an increased risk of autism spectrum disorders. Conversely, a loss of function due to mutations, deletions, paternal duplications, or imprinting defects in neurons leads to Angelman syndrome. Emerging evidence suggests that abnormal UBE3

Small non-coding RNA profiles in sperms from depressive-like mice induced by chronic unpredictable mild stimulations.

Major depressive disorder (MDD) is a complex, common, and moderately heritable illness, and accumulating evidence suggests that psychological stress may affect male fertility. Mounting evidence indicates sncRNAs in sperm are sensitive to environmental changes and mediate the inheritance of paternally acquired metabolic and mental traits. In order to know the impact of psychological stress on reproduction and alterations in sperm sncRNAs, in this study, depressive-like mice induced by chronic unpredictable mild stimulation (CUMS) were used to investigate the impact of psychological stress on re
