Elucidating the role of medicinal plants in melanoma treatment: a review on metastatic progression and phytochemical intervention.

Metastatic melanoma remains one of the most highly aggressive and treatment-resistant cancers, highlighting the need for enhanced understanding of its progression and novel therapeutic strategies. The metastatic cascade is an intricate, dynamic, and coordinated series of events which initiates with melanoma cell proliferation and the release of regulatory signalling molecules, followed by angiogenesis in response to hypoxia and nutrient deprivation, and extracellular matrix (ECM) degradation and invasion via matrix metalloproteinase (MMP) activity. Intravasation generates circulating tumour ce

Establishment of a mouse allergy model for Culicoides (Diptera: Ceratopogonidae).

BACKGROUND: Culicoides is a genus of ubiquitous biting midges (Ceratopogonidae). Female midges have blood-sucking habit. They not only bite and harass humans and animals but also may be an important vector of disease transmission. Therefore, building an animal allergy model caused by Culicoides biting is very beneficial for studying its pathogenesis and exploring the therapeutic methods. MATERIAL AND METHOD: Kunming mice were used in this study to build the model and sensitised by two-step injection of midge extracts. Scratching behaviour and histological examination were used to check the im

Hydroxyanthracene derivates citotoxicity: A differential evaluation between single molecule and whole plant extract.

Hydroxyanthracene derivates (HADs) are a group of natural or synthetic compounds with a wide range of biological activities (for instance, anti-inflammatory, antibacterial, and antiarthritic). In addition, because of their properties for helping the normal bowel function, HADs are widely used in constipation as pharmacological drugs and nutritional supplements. Nevertheless, during the past years, a safety usage of HAD products has been under consideration because some studies reported that HADs are not lacking toxicity (i.e., genotoxic and carcinogenic activity). Thus, the first objective of

Intrinsic physiological properties underlie auditory response diversity in the avian cochlear nucleus.

Sensory systems exploit parallel processing of stimulus features to enable rapid, simultaneous extraction of information. Mechanisms that facilitate this differential extraction of stimulus features can be intrinsic or synaptic in origin. A subdivision of the avian cochlear nucleus, nucleus angularis (NA), extracts sound intensity information from the auditory nerve and contains neurons that exhibit diverse responses to sound and current injection. NA neurons project to multiple regions ascending the auditory brain stem including the superior olivary nucleus, lateral lemniscus, and avian infer

Whole-grain oats (Avena sativa L.) as a carrier of lactic acid bacteria and a supplement rich in angiotensin I-converting enzyme inhibitory peptides through solid-state fermentation.

This study explored a novel strategy to develop solid-state whole-grain oats as a novel carrier of lactic acid bacteria and a nutraceutical supplement rich in ACE inhibitory peptides. Oats were fermented by Lactobacillus plantarum B1-6, Rhizopus oryzae, or a combination of L. plantarum B1-6 and R. oryzae. L. plantarum showed a much better growth performance in oats when it was combined with R. oryzae than when it was cultured alone, as evidenced by an increase in viable cell count to 9.70 log cfu g-1 after 72 h of fermentation. The coinoculated fermented oats (CFO) and the R. oryzae-fermented

Dopamine in the ink defence system of Sepia officinalis: biosynthesis, vesicular compartmentation in mature ink gland cells, nitric oxide (NO)/cGMP-induced depletion and fate in secreted ink.

The biosynthesis, localization and fate of catecholamines in the ink gland of the cuttlefish Sepia officinalis were investigated by combined biochemical and immunohistocytochemical methodologies. HPLC analysis of crude ink gland extracts indicated the presence of dopa (2.18+/-0.82 nmol/mg of protein) and DA (dopamine, 0.06+/-0.02 nmol/mg of protein), but no detectable noradrenaline or adrenaline. DA was shown to derive from L-tyrosine, according to experiments performed by incubating intact ink glands with [L-14C]tyrosine. The biosynthetic process involves a tyrosine hydroxylase and a dopa dec

Hedgehog signaling regulation of homeodomain protein islet duodenum homeobox-1 expression in pancreatic beta-cells.

Insulin gene expression in pancreatic beta-cells is regulated by signals from developmental morphogen proteins known as hedgehogs (Hhs). By analyzing 5'-deletion insulin promoter-reporter constructs in transient transfections of clonal INS-1 beta-cells, we located activating Hh-responsive regions within the rat insulin I promoter that include the glucose-response elements Far (E2) and Flat (A2/A3). Activation of Hh signaling in INS-1 cells by ectopic Hh expression increased (and inhibition of Hh signaling with the Hh-specific inhibitor cyclopamine decreased) transcriptional activation of a mul
