Natural killer cell stimulatory factor (NKSF) or interleukin-12 is a key regulator of immune response and inflammation.

Natural Killer cell Stimulatory Factor (NKSF) or interleukin-12 (IL-12) is a heterodimeric cytokine of 70 kDa formed by a heavy chain of 40 kDa (p40) and a light chain of 35 kDa (p35). Although it was originally identified and purified from the supernatant of Epstein-Barr virus-transformed B cell lines, it has been shown that among peripheral blood cells NKSF/IL-12 is predominantly produced by monocytes, with lower production by B cells and other accessory cells. The most powerful inducers of NKSF/IL-12 production are bacteria, bacterial products and parasites. In addition to the biologically

CHIKV-ZIKV antibody Co-positivity and preserved neutralising activity in a Nigerian arboviral study.

BACKGROUND: Arboviral coinfections complicate public health responses in endemic regions. Antibody-dependent enhancement (ADE) is a well-characterised phenomenon among related flaviviruses, yet its relevance to cross-family alphavirus and flavivirus interactions remains unclear. We examined patterns of CHIKV-ZIKV coinfection in a Nigerian cohort and asked whether the serological data were consistent with antibody-mediated or alternative mechanisms. METHODS: We carried out serological surveillance of 871 participants across three ecological zones in Nigeria, measuring IgG and IgM responses to

Caregiver acceptability, and uptake of voluntary medical circumcision among male children under 5 years in Gulu city, Northern Uganda.

Childhood male circumcision is a key intervention for reducing the transmission of HIV infection. Despite its proven efficacy, the uptake of childhood male circumcision across Uganda remains suboptimal. This study assessed the caregiver acceptability, and uptake of voluntary medical circumcision among male children under five years old in Gulu city, Uganda. A cross-sectional quantitative approach was employed, involving multistage sampling strategy to randomly select 16 parishes from Gulu City’s two divisions. Systematic household sampling, and random caregiver selection using even-numbered ra

[A case of progressive multifocal leukoencephalopathy presenting white matter MRI lesions extending over the cerebral cortex and a marked decrease in cerebral blood flow on SPECT, and associated with HTLV-I infection].

We report a 47-year-old woman with progressive multifocal leukoencephalopathy (PML). She was a carrier of HTLV-I virus, and developed subacute right hemiparesis and marked motor aphasia. She had a malignant lymphoma in the left neck and basal cell carcinoma in the right inguinal region. Three months after the onset, she became unable to walk because of the right leg weakness or to speak because of motor aphasia. Magnetic resonance imaging (MRI) revealed multifocal T2-high lesions in the white matter of the left frontal lobe, and a brain biopsy revealed demyelinating pathology. A biopsy of the

Memory Strategy Use, Cognition, and Metacognition Among People Aging with HIV.

Living with the HIV virus is a chronic health condition with life expectancies among treated people similar to seronegative individuals. However, older people living with HIV (PLWH) are at higher risk for comorbidities, including cognitive impairment, than their seronegative counterparts. Use of memory strategies has been shown to enhance memory performance among older PLWH, yet few studies have examined factors that drive memory strategy use as well as how impairments in metacognition may hinder the initiation of memory strategies. To address this gap, associations between cognitive function,

Pathogenic characteristics of central nervous system infections in AIDS individuals: a retrospective cohort study based on immune status.

BACKGROUND: Human immunodeficiency virus (HIV) infection frequently leads to central nervous system (CNS) complications, especially as immunity declines. While antiretroviral therapy (ART) has transformed HIV into a manageable condition, in clinical practice, a significant subset of individuals still present with advanced immunosuppression (CD4 < 200 cells/μL) due to delayed diagnosis, treatment failure, or poor adherence, leading to AIDS-defining conditions. This study investigated how immune status affects CNS infection characteristics and outcomes specifically within this immunocompromised

Hepatitis C Prevalence and Management Among Patients Receiving Opioid Substitution Treatment in General Practice in Ireland: Baseline Data from a Feasibility Study.

BACKGROUND: Hepatitis C virus (HCV) infection is a major cause of chronic liver disease and death. Injection drug use is now one of the main routes of transmission of HCV in Ireland and globally with an estimated 80% new infections occurring among people who inject drugs (PWID). OBJECTIVE: We aimed to examine whether patients receiving opioid substitution therapy in primary care practices in Ireland were receiving guideline-adherent care regarding HCV screening. Ireland has developed a model of care for delivering opioid substitution treatment in the primary care setting. We conducted this st

A Dual-Modality Herpes Simplex Virus 2 Vaccine for Preventing Genital Herpes by Using Glycoprotein C and D Subunit Antigens To Induce Potent Antibody Responses and Adenovirus Vectors Containing Capsid and Tegument Proteins as T Cell Immunogens.

UNLABELLED: We evaluated a genital herpes prophylactic vaccine containing herpes simplex virus 2 (HSV-2) glycoproteins C (gC2) and D (gD2) to stimulate humoral immunity and UL19 (capsid protein VP5) and UL47 (tegument protein VP13/14) as T cell immunogens. The HSV-2 gC2 and gD2 proteins were expressed in baculovirus, while the UL19 and UL47 genes were expressed from replication-defective adenovirus vectors. Adenovirus vectors containing UL19 and UL47 stimulated human and murine CD4(+) and CD8(+) T cell responses. Guinea pigs were either (i) mock immunized; (ii) immunized with gC2/gD2, with CpG

[Viral superantigens].

Superantigens (SAgs) are microbial proteins produced by various microorganisms that elicit excessive and strong stimulation of T cells via an unconventional mechanism. They cause polyclonal activation of T cells in a non-specific manner, by binding to a particular variable-beta (Vβ) chain of T-cell receptor (TCR) and MHC class II molecule, in unprocessed form and outside of peptide-binding cleft, forming a bridge between the antigen presenting cell and the T cell. SAgs are classified into three groups, namely 1) exogenous (soluble proteins and exotoxins secreted by microorganisms), 2) endogeno

Assessment of differentially culturable tubercle bacteria assays for the detection of tuberculosis infection in asymptomatic household contacts and the implications for intra-household transmission: a longitudinal cohort study.

BACKGROUND: Conventional culture methods for tuberculosis diagnosis miss differentially culturable tubercle bacteria (DCTB), which grow only in liquid assays supplemented with growth-enhancing factors. This limitation, combined with inadequate contact tracing and screening, often fails to identify asymptomatic individuals, with live bacilli detectable by enhanced culture methods. This shortfall results in undiagnosed reservoirs of bacteria, potentially fuelling ongoing transmission. In this study, we aimed to investigate whether DCTB assays provide greater sensitivity by detecting more Mycobac

A Systematic Review of Barriers and Facilitators to the Uptake and Adherence of Injectable PrEP Among Adolescent Girls and Young Women in Sub-Saharan Africa.

Adolescent girls and young women (AGYW) in sub-Saharan Africa bear a disproportionate burden of HIV. Long-acting injectable PrEP (LAI-PrEP) may address adherence challenges associated with daily oral PrEP, but social and structural barriers threaten real-world implementation. Drawing on parallels with injectable contraception and multipurpose prevention technologies (MPTs), this systematic review synthesizes evidence on barriers and facilitators to LAI-PrEP use among AGYW in sub-Saharan Africa. Following PRISMA guidelines, peer-reviewed studies published January 2015 to February 2025 were iden
