Dermatologic Adverse Events Associated with T-Cell Engager Therapy.

T-cell engager therapies, including bispecific T-cell engagers and the immune-mobilizing monoclonal T-cell receptor against cancer tebentafusp, are an emerging class of anticancer immunotherapy, with rapid expansion of the class since initial approval of blinatumomab in 2014 and with distinct dermatologic adverse events increasingly recognized across agents. Tebentafusp and talquetamab demonstrate highest rates of notable dermatologic toxicity reflecting on-target off-tumor cutaneous effects. Tebentafusp produces dermatologic adverse events in the majority of treated patients, characterized by

RNA Helicase DDX21 Controls CD4+ T Cell Proliferation and Promotes Inflammatory Bowel Disease via Translational Control.

Inflammatory bowel disease (IBD) is characterized by dysregulated T cell responses. RNA helicases, including DExD-box helicase 21 (DDX21), are pivotal in RNA metabolism, but their role in T cell-mediated pathology during IBD remains unclear. Here, we demonstrate that DDX21 expression in CD4+ T cells correlates with cell cycle and translation pathways in IBD-affected tissues. Conditional deletion of Ddx21 in mouse T cells disrupts T cell homeostasis and impairs cell proliferation. Consequently, Ddx21-deficient CD4+ T cells exhibit resistance to inducing adoptive transfer colitis, with recipient

Tertiary Lymphoid Structures and CXCL13 as Targets in Intracranial Aneurysm.

BACKGROUND: Intracranial aneurysms (IAs) are localized dilations of cerebral arteries that pose a significant threat of rupture, leading to fatal subarachnoid hemorrhage. The inflammation driven by immune cell infiltration is believed to be a key driver for the pathogenesis of IAs; however, the detailed immune cell landscape and spatial composition of immune cells remain unclear. METHODS: In this study, we employed an integrated multi-omics approach using human IA tissues, including single‑cell RNA sequencing, spatial transcriptomics, microarray data, and multiplex immunofluorescence, to cons

ASSESSMENT OF VITEX AGNUS-COSTES PLANT EXTRACT ON THE HEART OF MICE EXPOSED TO OXIDATIVE STRESS.

Myocardial injury is largely the result of oxidative stress, which alters the structure and function of the heart's cardiomyaceous and conduction cells. The purpose of this investigation is to determine the relative effects of Vitex agnus-castus, a plant that is known to contain anti-inflammatory and antioxidant properties, upon the histology of the hearts of mice exposed to oxidative stress. Histological examinations were performed on the cardiac tissues of experimental groups of mice that were subjected to oxidative stress, either with or without treatment with V. agnus-castus extract. Where

PD-1 regulates latent effector differentiation of thymic cytotoxic CD8+ T cells.

Durable T cell immunity against cancer depends on the continual replenishment of effector CD8⁺ T cells. Thymic output has been associated with favorable prognosis in cancer patients across a range of ages, suggesting that the thymus is an important source for replenishing T cells capable of controlling cancer progression. However, whether CD8⁺ T cells acquire effector potential within the thymus, and how thymic output of effector CD8⁺ T cells contribute to peripheral tumor immunity, remain unclear. In this study, we discover that thymic single-positive (SP) CD8⁺ T cells undergo latent effector

Nanoparticle-enabled CD40 silencing programs immune tolerance via a DC-Treg-Breg axis in autoimmune myocarditis.

Autoimmune myocarditis is a severe inflammatory heart disease that can progress to dilated cardiomyopathy and heart failure, while disease-modifying therapies remain limited. Dysregulated activation of antigen-presenting cells, particularly dendritic cells (DCs), disrupts immune tolerance and drives pathogenic cardiac inflammation. Here, we report a nanoparticle-enabled CD40 silencing strategy for immune tolerance programming in experimental autoimmune myocarditis. Using a polymer-lipid hybrid siRNA formulation, we achieved sustained CD40 silencing in myeloid antigen-presenting cells and induc

Avoidance of MAIT cells is an essential determinant of Listeria monocytogenes pathogenesis.

Mucosal-associated invariant T (MAIT) cells are among the most conserved and abundant innate-like T cells in humans that recognize microbial-derived riboflavin precursors and elicit potent antimicrobial responses. The foodborne pathogen Listeria monocytogenes is a broad host-range facultative intracellular pathogen that lacks the riboflavin biosynthetic pathway, leading us to hypothesize that this deficiency is pathoadaptive and allows the pathogen to evade MAIT cells. Here, we show that L. monocytogenes strains engineered to produce riboflavin (L. monocytogenes-ribDEAHT) are attenuated in wil

IL-17A-producing γδ T cells control pulmonary Mycobacterium abscessus infection in mice.

The specific role of γδ T cells in non-tuberculous mycobacteria (NTM) infections remains incompletely understood. Here we characterized the immune landscape of pulmonary tissues from a Mycobacterium abscessus-infected NTM mice model. Interleukin-17 (IL-17)A+ γδ T cells were essential for controlling M. abscessus infection by directly eliminating extracellular bacteria. These cells conferred protective immunity in two mouse models of infection combined with either pulmonary fibrosis or a lack of functional interferon-gamma (IFNγ)-mediated immunity. IL-17A+ γδ T cells exerted bactericidal activi

A new method to measure cell metabolism of rare cells in vivo reveals a high oxidative phosphorylation dependence of lung T cells.

Regulation of cellular metabolism is a central element governing the fate and function of T cells. However, the in vivo metabolic characteristics of rare cells, such as nonlymphoid tissue T cells, are poorly understood because of experimental limitations. Most techniques measuring cell metabolism require large cell numbers. The recent SCENITH method allows for studying the metabolism of rare cells by flow cytometry. However, this technique requires cells to be isolated and cultured ex vivo, which may alter their metabolism. Here, we propose a new experimental approach, called in vivo SCENITH,

Comparative Genomics Analysis Provides Insights into Pregnancy Maintenance Mechanisms in the Ovoviviparous Teleost Black Rockfish (Sebastes schlegelii).

The black rockfish (Sebastes schlegelii), an economically important ovoviviparous teleost in Chinese aquaculture, exhibits a distinctive reproductive strategy characterized by internal fertilization and prolonged embryonic development within females until live birth. However, this reproductive mode poses substantial challenges in artificial breeding practices due to a poor understanding of pregnancy (hereafter referring to intraovarian gestation in S. schlegelii) maintenance mechanisms. Here, we conducted a comparative genomic analysis between S. schlegelii and representative oviparous and ovo

Single-cell multiomic analysis unveils the immune landscape dynamics of graves' ophthalmopathy.

Graves' ophthalmopathy (GO) is an autoimmune disease that occurs concurrently with Graves' disease, potentially leading to facial disfigurement and irreversible vision loss. However, the molecular characteristics of cellular changes in the peripheral immune environment of GO patients remain unclear. This study presents a multi-omics single-cell analysis of peripheral blood from GO patients and healthy controls, revealing key molecular alterations in immune cell subpopulations. We identify increased chromatin accessibility and regulatory activity of pro-inflammatory factors, such as FOS and NF-

A novel iron bioresorbable scaffold: a potential strategy for pulmonary artery stenosis.

A big diameter bioresorbable scaffold is expected to be used for treatment of vessel stenosis of children with congenital heart disease to adapt the growth characteristics of vessel of children and avoid the late adverse events of permanent stent implanted in children. However, it is challenging to fabricate a big diameter bioresorbable scaffold that is appropriate for percutaneous implantation with enough mechanical performance and can be smoothly delivered in children's small vessel. In this study, a novel iron big and bioresorbable Scaffold (BBS) for pulmonary artery stenosis of children wi

Ameloblastoma with adenoid features with multiple local recurrences: report of a case with clinicopathologic and immunohistochemical studies.

Ameloblastoma with adenoid features is characterized by the formation of cribriform or glandular tubular structures in addition to the conventional ameloblastoma; however, many aspects of the clinical and histologic characteristics are unclear. We report a case of ameloblastoma with adenoid features that occurred in the mandible and had multiple recurrences. The patient was a 54-year-old woman who presented with a chief complaint of painless swelling in the left mandibular molar region. On the first visit, a bone expansion was noted in the lingual alveolar region of the left mandibular molar.

Gosha-Jinki-Gan Reduces Inflammation in Chronic Ischemic Stroke Mouse Models by Suppressing the Infiltration of Macrophages.

Ischemic stroke is a primary cause of cerebrovascular diseases and continues to be one of the leading causes of death and disability among patients worldwide. Pathological processes caused by vascular damage due to stroke occur in a time-dependent manner and are classified into three categories: acute, subacute, and chronic. Current treatments for ischemic stroke are limited to effectiveness in the early stages. In this study, we investigated the therapeutic effect of an oriental medicine, Gosha-jinki-gan (TJ107), on improving chronic ischemic stroke using the mouse model with middle cerebral

Inhibition of Abdominal Aortic Aneurysm Progression Through the CXCL12/CXCR4 Axis via MiR206-3p Sponge.

Notably, the C-X-C Motif Chemokine Ligand 12/C-X-C Chemokine Receptor Type 4 (CXCL12/CXCR4) signalling pathway's activation is markedly increased in a mouse model of abdominal aortic aneurysms (AAA). Nonetheless, the precise contribution of this pathway to AAA development remains to be elucidated. The AAA mouse model was induced by local incubation with elastase and oral administration of β-aminopropionitrile. The activity level of the CXCL12/CXCR4 axis was evaluated in both human AAA patients and the mouse model. Smooth muscle cell lineage tracing determined the expression and localisation of

Oral and Skin Sensitisation to Peanut Show Different Immunological Features in Brown Norway Rats.

Food allergy may develop after both oral and skin exposure to food allergens. Identifying immunological features associated with the sensitisation route is important for understanding the extent to which patients are sensitised through the oral or skin route and may be required to improve food allergy immunotherapy. Thus, the present study aimed at comparing the immunological features of peanut sensitisation induced through the oral and skin route in Brown Norway (BN) rat models. Sensitisation to peanut protein extract (PPE) was induced via oral or skin administration in BN rats. Allergy devel

Dietary cysteine enhances intestinal stemness via CD8+ T cell-derived IL-22.

A fundamental question in physiology is understanding how tissues adapt and alter their cellular composition in response to dietary cues1-8. The mammalian small intestine is maintained by rapidly renewing LGR5+ intestinal stem cells (ISCs) that respond to macronutrient changes such as fasting regimens and obesogenic diets, yet how specific amino acids control ISC function during homeostasis and injury remains unclear. Here we demonstrate that dietary cysteine, a semi-essential amino acid, enhances ISC-mediated intestinal regeneration following injury. Cysteine contributes to coenzyme A (CoA) b
