[Yersinia pestis factors, assuring circulation and maintenance of the plague pathogen in natural foci ecosystems. Report 1].

Everlasting reproduction of Yersinia pestis, plague bacillus in natural pestholes needs virulent causative agent to invade into the host entity, be potent to overcome protection powers of the rodent organism and to pullulate to entail bacteriemia for subsequent conveyance the plague bacillus to the new host by fleas. All of legs of life cyclic patterns of Yersinia pestis are maintained by a number of plague bacillus factors acting jointly or separately, participating at the different stages of infectious process or conveyance. However these factors provide the perpetuation of the plague bacill…

ALBAN (GETUG-AFU 37): a phase III, randomized, open-label international trial of intravenous atezolizumab and intravesical Bacillus Calmette-Guérin (BCG) versus BCG alone in BCG-naive high-risk, non-muscle-invasive bladder cancer (NMIBC).

BACKGROUND: Standard treatment of high-risk non-muscle-invasive bladder cancer (NMIBC) after transurethral resection of bladder tumor is intravesical instillation with Bacillus Calmette-Guérin (BCG) (induction and maintenance regimens); however, BCG therapy still fails in 30%-40% of patients. Prior studies suggest that programmed death-ligand 1 (PD-L1) expression and alterations in immune infiltration might be associated with BCG failure. PATIENTS AND METHODS: The ALBAN trial is an international, 1 : 1 randomized, open-label phase III trial comparing the combination of atezolizumab (an anti-P…

Radiotherapy Combined with a Radiosensitizer for Bacillus Calmette-Guérin-unresponsive Non-muscle-invasive Carcinoma In Situ Bladder Cancer: An Open-label, Single-arm, Multicenter, Phase 2 European Organisation for Research and Treatment of Cancer Trial.

Radical cystectomy with pelvic lymph node dissection and urinary diversion is the standard of care for patients with bacillus Calmette-Guérin (BCG)-unresponsive non-muscle-invasive bladder cancer (NMIBC). However, many patients are unwilling or unable to undergo such major surgery associated with high morbidity and a negative impact on quality of life. Chemoradiotherapy is an established treatment option for muscle-invasive bladder cancer. However, it has not been investigated adequately in NMIBC until now. The European Organisation for Research and Treatment of Cancer (EORTC) 2235 study (NCT0…

[Study of the antagonistic action of actinomycetes on anthrax bacilli].

Capacity for the growth inhibition of the highly virulent causative agent of Siberia plague was studied with respect to actinomycetous strains from soil samples of the Ukraine. It was found that on nutrient media 88.4, 88.9, 88.1 and 93.4% of the isolates inhibited the growth of Bac. anthracis, Bac. cereus, Bac. mycoides and Tsenkovsky vaccine strain respectively. Representatives of very different taxonomic groups were found among the antagonistic actinomycetes. Clearance of the soil from Siberia plague bacilli stable to the outer effect with the help of the active strain L-721 of Act. chromof

The use of ultraviolet light generated from light-emitting diodes for the disinfection of transvaginal ultrasound probes.

Transvaginal ultrasound probes (TVUS) are used for several gynecological procedures. These need to be disinfected between patient use. In th … d repeated high-level disinfection was analysed by deliberately contaminating probes with spores of Bacillus subtilis and then performing the UVC disinfection and bacterial culture. 50% of probes were contaminated with bacteria, most commonly Bacillus sp., directly after in vivo use. Whereas 97% were sterile after UVC disinfection for 90 seconds. The UVC t …

Comparison of surface sampling methods for an extended duration outdoor biological contamination study.

Bacillus anthracis, the causative agent for anthrax, is a dangerous pathogen to humans and has a history as a bioterrorism agent. While sampling methods have been developed and evaluated for characterizing and clearing contaminated indoor sites, the performance of these sampling methods is unknown for use in outdoor environments. This paper presents surface sampling data for Bacillus atrophaeus spores, a surrogate for B. anthracis, from a 210-day outdoor study that evaluated the detection and recovery of spores using five different sampling methods as follows: sponge sticks, 37-mm vacuum filte…

The ups and downs of negative (and positive) selection of B cells.

The purpose of this study was to construct a Coxsackie virus A16 (CA16) mucosal vaccine and evaluate its ability to induce immune response. VP1 gene of CA16 was inserted into the genome of Bacillus subtilis via recombination and displayed on the surface of the spores. This Bacillus-based vaccine was used for intranasal immunization of mice and the serum antibody titer was determined by enzyme-linked immunosorbent assay (ELISA). Neutralization activity of the serum from immunized mice was analyzed by an in vitro neutralizing test. VP1 gene was successfully integrated into the genome of Bacillus…

Gene expression of pro-inflammatory (IL-8, IL-18, TNF-α, and IFN-γ) and anti-inflammatory (IL-10) cytokines in the duodenum of broiler chickens exposed to lipopolysaccharides from Escherichia coli and Bacillus subtilis.

BACKGROUND AND AIM: Intestinal infections are associated with Gram-negative bacteria like Escherichia coli. When eliminated by treatments during replication, E. coli release lipopolysaccharides (LPS) that can activate the intestinal immune system and increase the expression of cytokines, such as interleukin (IL)-8, IL-18, tumor necrosis factor-alpha (TNF-α), and interferon-gamma (IFN-γ), by the intestinal epithelium under pathological conditions. This study aimed to evaluate the addition of Bacillus subtilis to the duodenal gene expression of pro-inflammatory and anti-inflammatory cytokines in…

Microscopic detection of bacillus Calmette-Guérin mycobacteria in bladder biopsy using fluorescence in situ hybridization: Détection microscopique des Bacilles biliés de Calmette et Guérin (BCG) dans une biopsie vésicale par hybridation in situ en fluorescence.

Intravesical instillation of Bacilli Calmette Guérin (BCG) as a superficial bladder cancer treatment is generally well tolerated, but local or systemic complications may occur, some of which may be life-threatening. Following the suspicion of post-BCG cystitis in a 72-year-old man with a history of urothelial carcinoma treated by intravesical BCG instillation, we used fluorescence in situ hybridization (FISH) targeting the rpoB gene of the Mycobacterium tuberculosis complex to detect Mycobacterium bovis BCG in paraffin-embedded bladder biopsy sections. FISH yielded specific detection of BCG my

Robust Single-Cell Raman Identification Enabled by Target-Interference Library.

Foodborne pathogens, especially spore-forming Bacillus species, present a continuous challenge to food safety owing to their resilience against conventional sterilization methods and their ability to rapidly produce toxins. Single-cell Raman spectroscopy enables rapid and label-free detection of bacterial cells. However, its practical application is often limited by a low signal-to-noise ratio (SNR). This issue becomes particularly pronounced in food testing, where complex sample matrices and diverse interfering components can significantly elevate spectral background noise, thereby compromisi…

[The duration of the persistence of the plaque microbe in the body of the flea Citellophilus tesquorum altaicus].

Experiments have established that the plague bacillus persists long in the flea C. tesquorum altaicus in the Tuva natural foci. Experimentally, 64.2 to 77.3% of the fleas inoculated in autumn survived winter. More than one-half preserved the plague bacillus. From 0.7 to 19.4% of the inoculated fleas survived from one summer to another. The plague persisted in 30% of the insects. Individual C. tesquorum altaicus females survived two winters, preserving the bacillus for 21.5-22.0 months. The fact that the fleas has survived both one and two winter periods in coma transmitted the plague bacillus …

[The problem of natural foci of plague: the search for ways for its resolution].

The paper deals with one of the most important problems of plague natural foci, namely the places and types of its bacillus persistence outside the parasitic system, as well as in the interepizootic period. It also discusses different hypotheses forwarded on the basis of field observations and experimental findings. In particular, it gives the materials suggesting that the plague bacillus can persist in the low-virulent form under natural conditions, as well as due to its transition to a L-form. The paper provides strong evidence for the long-term persistence of the plague bacillus in soil and…

Isolation and identification of keratinolytic probiotic Bucillus licheniformis bacteria from the soil below poultry slaughterhouse waste.

Feathers make up 7% of the total weight of adult chickens and keratin protein makes up 85% of the feathers. Today, the keratinase enzymes of some Bacillus strains are used to degrade and process raw keratin waste for animal and poultry feed. According to various studies, the probiotic properties of some spore-shaped Bacillus have also been proven. The study aimed to isolation of the keratinolytic Bacillus bacteria that they have probiotic properties for using in the livestock and poultry feed industry. We were able to isolate 8 strains of Bacillus licheniformis with kreatin degrading propertie…

Optimization and application of probiotics complex for grouper based on dynamic model.

This study applied systematic microecological method to develop multi-strain probiotics for grouper aquaculture, focusing on rational strain selection and interaction optimization. Five strains from marine source were initially screened through functional and safety assessments. Based on single-strain growth kinetics and pairwise interaction data, the generalized Lotka-Volterra model and multiple linear regression were employed to identify an optimal consortium consisting of Bacillus subtilis, Bacillus megaterium, and Bacillus velezensis (1: 1: 1), which exhibited significantly antagonism agai…

Evaluation of long-term supplementation of a Bacillus subtilis direct-fed microbial and enzymatically hydrolyzed yeast cell culture product used alone or in combination on Clostridia, Clostridium perfringens, Escherichia coli, and Salmonella prevalence in beef steers.

The objective was to determine the influence of long-term supplementation (258 d) of a direct-fed microbial (DFM) and/or yeast cell wall (YCW) product on bacterial populations in beef steers. Single-sourced Charolais × Red Angus steers (n = 256; body weight = 246 ± 1.68 kg) were used in a randomized complete block design and blocked by location into one of four treatments: 1) fed no DFM and no YCW (Control); 2) fed only the DFM (DFM; Certillus CP B1801 Dry, 28 g/steer d-1 ); 3) fed only the YCW (YCW; Celmanax; 18 g/steer d-1 ); and 4) fed the DFM and the YCW (DFM+YCW). Steers were vaccinated f
