'Catch and Release' Polymeric Fibers: Versatile Interfaces for Engineering Reversible Platforms for Biomolecular Immobilization and Antibacterial Coatings.

Stimuli-responsive polymeric fibers have emerged as an indispensable material for numerous biomedical applications. Strategies to conjugate functional molecules with high specificity onto these nanofibers are vital to tailor these materials for specific applications. When the functionalization is reversible, these materials can serve as a 'catch and release' platform, which widens their applicability. Herein, polymeric fibers with an average diameter of about 237 ± 44 nm, amenable to reversible conjugation, are fabricated using electrospinning. The thiol-disulfide exchange reaction is employed

Enabling malic acid production from corn-stover hydrolysate in Lipomyces starkeyi via metabolic engineering and bioprocess optimization.

BACKGROUND: Lipomyces starkeyi is an oleaginous yeast with a native metabolism well-suited for production of lipids and biofuels from complex lignocellulosic and waste feedstocks. Recent advances in genetic engineering tools have facilitated the development of L. starkeyi into a microbial chassis for biofuel and chemical production. However, the feasibility of redirecting L. starkeyi lipid flux away from lipids and towards other products remains relatively unexplored. Here, we engineer the native metabolism to produce malic acid by introducing the reductive TCA pathway and a C4-dicarboxylic ac…

Single Organic Cation Engineering Cu(I)-Based Ionic and Coordinate Type Halides as High-Efficiency Hydrogel Scintillator.

Cu(I)-based halide scintillators are promising candidates for X-ray detection due to excellent scintillation and low-cost solution processability. However, the rational design of Cu(I)-based halide scintillators remains challenging due to insufficient theoretical frameworks elucidating their structure-property correlations. In this work, two Cu(I)-based hybrid metal halides, Cu2I4-IC (Ionic compound) are designed and Cu4I4-CC (Coordination compound) via bonding mode control engineering. The Cu2I4-IC exhibits blue emission, and the Cu4I4-CC exhibits yellow emission, and the photoluminescence qu…

Planar conjugate donor engineering enhanced photodynamic immunogenic cell death stimulator for efficient Triple-Negative breast cancer immunotherapy.

Breast cancer is the most diagnosed cancer type among women worldwide and quite a few will be diagnosed with triple-negative breast cancer (TNBC), which is still a challenge to be treated. Immunotherapy has demonstrated immense therapeutic potential. However, the characteristics of the immunosuppressive tumor microenvironment (ITME) limit the efficacy of immunotherapy in TNBC treatment. Herein, a molecule-electron engineering strategy is employed to optimize the excited state characteristics of endoplasmic reticulum (ER)-targeted photodynamic immunogenic cell death (ICD) stimulators. Beneficia…

Conformational Engineering of Flexible Protein Fragments on the Surface of Different Nanoparticles: The Surface-Atom Mobility Rules.

As a newly emerging technology, conformational engineering (CE) has been gradually displaying the power of producing protein-like nanoparticles (NPs) by tuning flexible protein fragments into their original native conformation on NPs. But apparently, not all types of NPs can serve as scaffolds for CE. To expedite the CE technology on a broader variety of NPs, the essential characteristic of NPs as scaffolds for CE needs to be identified. Herein, we investigate the potential of two distinct types of NPs as scaffolds for CE: CdSe/ZnS quantum dots (QDs), an ionic compound NP, and palladium NPs (P…

Heterologous Production of Forskolin in Tobacco (Nicotiana tabacum) via Glandular Trichome Specific Engineering and Metabolic Flux Redirection.

Plants are promising bioreactor for the sustainable and scalable production of high-value natural bioactive compounds, because they can synthesise phytochemicals from CO2, light, water and minerals through their innate photosynthetic carbon assimilation machinery. However, metabolic engineering in multicellular plants via stable transgene is challenged by the low accumulation of heterologous compounds. Forskolin is a labdane-type diterpenoid that accumulates in the roots of medicinal plant Coleus forskohlii. It activates the cyclic adenosine 3',5'-monophosphate (cAMP) signalling pathway and is…

Transcriptomics Guided Engineering of Exosome-Encapsulated Bifunctional Nanosheets Targeting the Immune-PI3K/Akt Axis for Osteoporosis Therapy.

Osteoporosis, characterized by imbalanced bone metabolism and chronic inflammation, remains a therapeutic challenge due to the limitations of current single-target therapies. This study integrates transcriptomic insights with nanomaterial engineering to develop a multi-target strategy. Transcriptomic analysis of ovariectomized (OVX) mice reveals immune dysregulation and PI3K/Akt pathway activation, driving osteoclastogenesis. To address this, cobalt-aluminum layered double hydroxide nanosheets (f‑CA(OH)) are synthesized, which scavenge reactive oxygen species (ROS) via peroxidase-like activity…

Retinal organoid chip: engineering a physiomimetic oxygen gradient for optimizing long term culture of human retinal organoids.

An oxygen gradient across the retina plays a crucial role in its development and function. The inner retina resides in a hypoxic environment (2% O2) adjacent to the vitreous cavity. Oxygenation levels rapidly increase towards the outer retina (18% O2) at the choroid. In addition to retinal stratification, oxygen levels are critical for the health of retinal ganglion cells (RGCs), which relay visual information from the retina to the brain. Human stem cell derived retinal organoids are being engineered to mimic the structure and function of human retina for applications such as disease modeling

Computational Fluid Dynamics Modeling of Material Transport Through Triply Periodic Minimal Surface Scaffolds for Bone Tissue Engineering.

Cell-laden, scaffold-based tissue engineering methods have been successfully utilized for the treatment of bone fractures and diseases, caused by factors such as trauma, tumors, congenital anomalies, and aging. In such methods, the rate of scaffold biodegradation, transport of nutrients and growth factors, as well as removal of cell metabolic wastes at the site of injury are critical fluid-dynamics factors, affecting cell proliferation and ultimately tissue regeneration. Therefore, there is a critical need to identify the underlying material transport mechanisms and factors associated with cel…

Glycosaminoglycans, poly(aspartic acid) and gelatin-methacryloyl combination to obtain resorbable 3D-printed scaffolds for cartilage engineering.

Diseases affecting joints are among the major causes of disability worldwide, since repairing articular cartilage is challenging due to its avascular nature. Tissue engineering, using 3D scaffolds combined with stem cells/chondrocytes, offers promising solutions for cartilage regeneration. The scaffolds not only can support cellular growth but also may mimic the extracellular matrix (ECM) structure. By integrating stem cells that differentiate into chondrocytes, personalized tissues can be created, minimizing rejection risks and enhancing biocompatibility. This study aims to develop and charac…

mRNA engineering of allogeneic mesenchymal stem cells enables coordinated delivery of T cell engagers and immunotherapeutic cues.

Allogeneic cell therapies can enable off-the-shelf products that address limitations of autologous therapies. Mesenchymal stem cells are a robust allogeneic source, but no bioengineered mesenchymal stem cell-based therapies exist. Here we use mRNA engineering to create an off-the-shelf immunotherapy that we term DC-25. DC-25 consists of a mesenchymal stem cell armed with three designed mRNA constructs encoding CXCR4 to direct migration, a T cell engager specific for B cell maturation antigen to target B cell maturation antigen-expressing plasma cells involved in cancer and autoimmunity, and in…

Generation and ex vivo characterization of a full-thickness substitute of the human urethra by tissue engineering.

Tissue engineering may offer efficient alternatives for the surgical repair of severe conditions affecting the human urethra. However, development of tubular full-thickness substitutes is challenging. In this work, we have generated and evaluated ex vivo a novel full-thickness human urethra substitute (FHUS) containing its three main layers: the urethral mucosa (UM), the spongy layer (SP), and the tunica albuginea (AL). Results first showed that the generation of a FHUS significantly improved the biomechanical properties of this artificial tissue as compared to the individual layers, although …

3D-printed polycaprolactone/tricalcium silicate scaffolds modified with decellularized bone ECM-oxidized alginate for bone tissue engineering.

The treatment of large craniofacial bone defects requires more advanced and effective strategies than bone grafts since such defects are challenging and cannot heal without intervention. In this regard, 3D printing offers promising solutions through the fabrication of scaffolds with the required shape, porosity, and various biomaterials suitable for specific tissues. In this study, 3D-printed polycaprolactone (PCL)-based scaffolds containing up to 30 % tricalcium silicate (TCS) were fabricated and then modified by incorporation of decellularized bone matrix- oxidized sodium alginate (DBM-OA).

Sulphur-atom positional engineering in perylenimide: structure-property relationships and H-aggregation directed type-I photodynamic therapy.

An innovative design strategy of placing sulfur (S)-atoms within the pendant functional groups and at carbonyl positions in conventional perylenimide (PNI-O) has been demonstrated to investigate the condensed state structure-property relationship and potential photodynamic therapy (PDT) application. Incorporation of simply S-atoms at the peri-functionalized perylenimide (RPNI-O) leads to an aggregation-induced enhanced emission luminogen (AIEEgen), 2-hexyl-8-(thianthren-1-yl)-1H-benzo[5,10]anthra[2,1,9-def]isoquinoline-1,3(2H)-dione (API), which achieves a remarkable photoluminescence quantum
