Expression of acquired immunity to Heligmosomoides polygyrus in mice concurrently infected with Trypanosoma congolense.

The effects of concurrent Heligmosomoides polygyrus and Trypanosoma congolense infection on the expression of acquired resistance to homologous nematode challenge were studied in female outbred TO mice. Mice were infected with 500 infective larvae (L3) of H. polygyrus and the infection was terminated by anthelminthic treatment on Day 12, when the worms were adults. Eight days later sub-groups of these pre-exposed mice, and of similar mice which had not experienced the previous infection with H. polygyrus, were either simultaneously infected with 500 L3 of H. polygyrus and 10(4) bloodstream for

Successful application of deltamethrin pour on to cattle in a campaign against tsetse flies (Glossina spp.) in the pastoral zone of Samorogouan, Burkina Faso.

1,500-2,000 head of cattle were treated with deltamethrin 1% Spot On in an area of high tsetse densities, notably of Glossina morsitans submorsitans. After four treatments at monthly intervals, the time between two treatments was increased to two months. 11 months after the commencement of the campaign the fly population had decreased from initially 54.2 flies/trap/day to densities varying between 0.06-2.0 flies/trap/day, mostly G. palpalis gambiensis. Blood-meal analysis showed that this species was surviving in limited areas, mainly feeding on monitor lizards; consequently it is unlikely tha

Effects of phenothiazine neuroleptic drugs on the microtubular-membrane complex in bloodstream forms of Trypanosoma brucei.

African trypanosomes are parasitic protozoa causing sleeping sickness in humans and related diseases in domestic animals against which no entirely satisfactory forms of chemotherapy are yet available. It was previously shown that related species of trypanosomes, as well as procyclic (insect) forms of Trypanosoma brucei are extremely sensitive to the action of phenothiazine neuroleptic drugs in vitro. In this work, we have carried out a more detailed investigation of the effects of thioridazine, one of the most potent neuroleptic phenothiazine drugs known, on the morphology of the infective blo

Antitrypanosomal activity of phosphonylmethoxyalkylpurines.

Phosphonylmethoxyalkylpurines and -pyrimidines exhibit potent activity against a broad spectrum of DNA viruses. We evaluated some of these nucleotide analogues for antitrypanosomal activity in vitro and in mice. The most active compounds were (S)-9-(3-hydroxy-2-phosphonylmethoxypropyl) adenine (HPMPA) and (S)-9-(3-hydroxy-2-phosphonylmethoxypropyl)-2,6-diaminopurine (HPMPDAP), which inhibited growth of Trypanosoma brucei brucei by 50% (EC50 value) when incubated in vitro for 24 hr with 0.23-5.69 micrograms drug/ml. Both compounds completely eliminated multidrug-resistant T. b. brucei in cultur

Transgenic mice expressing human apolipoprotein A-I have sera with modest trypanolytic activity in vitro but remain susceptible to infection by Trypanosoma brucei brucei.

Although Trypanosoma brucei brucei fatally infects livestock in much of sub-Saharan Africa, humans are innately resistant to infection, apparently because high-density lipoproteins (HDL) in human serum lyse this unicellular protozoan parasite. Recently, we demonstrated that purified human apolipoprotein (apo) A-I, the major protein (M(r) 28,016) constituent of HDL, had full trypanolytic activity in vitro whereas the apoA-I of cattle and sheep was non-lytic. In the present study, we have sought to confirm the trypanocidal capability of human apoA-I by studying four lines of transgenic mice expr

Phosphorylation differences among proteins of bloodstream developmental stages of Trypanosoma brucei brucei.

Early in an infection the bloodstream forms of the African trypanosome Trypanosoma brucei brucei are long, slender and rapidly dividing. Later, non-dividing, short, stumpy forms may be found. In this report we described biochemical differences between the two parasite populations in the phosphorylation of their proteins in vitro. Compared with the slender populations, the non-dividing stumpy forms of the parasites exhibit decreased phosphorylation of an 80 kDa protein and enhanced phosphorylation of 37 kDa and 42 kDa proteins (pp37 and pp42). These changes occurred regardless of whether the st

In vivo interaction between Trypanosoma gambiense and leucocytes in mice.

Naturally occurring phagocytosis of Trypanosoma gambiense by mouse eosinophils and neutrophils was reported. In vivo and in vitro experiments using monoclonal antibodies confirmed that the phagocytosis is triggered by G1 class antibodies against variable surface antigen. Ultrastructural observation revealed the mode of entry and the intracellular fate of T. gambiense: initial attachment, pseudopodia formation and complete invagination. This phagocytosis resulted in the killing of T. gambiense by mouse eosinophils and neutrophils, suggesting that eosinophils and neutrophils give at least partia

Arsenic and old taxa: subspeciation and drug sensitivity in Trypanosoma brucei.

Resistance to the trypanocidal drugs atoxyl and tryparsamide was traditionally considered to be a diagnostic feature of rhodesian sleeping sickness and, consequently, of Trypanosoma rhodesiense. In examining the tryparsamide sensitivity of 13 isoenzymically defined stocks of the subgenus Trypanozoon, typical West African stocks showed no greater drug sensitivity than did those of East and Central Africa. The greatest resistance to tryparsamide was shown by two stocks isolated in the Ivory Coast. There was no evidence of strain differences in drug sensitivity to melarsoprol (Mel B) among 26 tes

Variable antigen type (VAT) composition of Trypanosoma brucei rhodesiense: discrepancy between results obtained using VAT-specific monoclonal antibodies and rabbit antisera.

Monoclonal antibodies have been made against clones of Trypanosoma brucei rhodesiense from the WRATAR 1 serodeme and analyzed by indirect immunofluorescence assay for specificity against homologous and heterologous clones. These antibodies were shown to be variable antigen type (VAT)-specific as they identified the majority of parasites in the homologous clones but few parasites in other clones. The reactivities of these VAT-specific monoclonal antibodies with uncloned human trypanosome isolates from Kenya were compared with the reactivities of polyvalent, VAT-specific rabbit sera on the same

Trypanosoma musculi and Trichinella spiralis: concomitant infections and selection for resistance genotypes in mice.

Trypanosoma musculi infections were given to mice of different strains before, at the same time, and after an infection with 400 Trichinella spiralis. Examined parameters of the host response to T. spiralis were worm rejection, antifecundity responses, development of immunological memory, and muscle larvae burden. After dual infection, each mouse strain showed characteristic effects on resistance to T. spiralis. This was due to a dynamic interaction between the genes controlling rejection of T. spiralis and those influencing T. musculi growth. C3H mice develop high trypanosome parasitemias. Th

Induction of T cell activity in athymic (nu/nu) mice infected with Trypanosoma rhodesiense.

Infection of C57BL/10 (B10)3 nu/nu mice with Trypanosoma rhodesiense results in the development of significant T-cell reactivity in spleen and lymph nodes. The proliferative responses to mitogens, such as concanavalin A (Con A) and phytohemagglutinin (PHA), and in mixed-lymphocyte reactions (MLR) to alloantigens are enhanced compared with control uninfected nu/nu mice. These results serve to emphasize the stimulatory nature of trypanosomes on the immune system.

Interference with anti-trypanosome immune responses in rabbits infected with cyclically-transmitted Trypanosoma congolense.

Rabbits were infected with two clones of antigenically distinct stocks of Trypanosoma congolense transmitted through Glossina morsitans. Local skin reaction development and the appearance of neutralizing antibodies were followed in animals infected with one or other of the trypanosome stocks, with both stocks simultaneously or with both stocks consecutively. There was little difference in local skin reaction development on rabbits infected with a single stock or with both stocks simultaneously but, in rabbits exposed to a heterologous stock 14 or 21 days after a primary infection reactions wer

Pathogenesis of anemia in Trypanosoma brucei-infected mice.

The pathogenesis of anemia was studied in trypanosome-infected mice. A strain of Trypanosoma brucei, TREU 667, was used which first produces an acute phase marked by waves of parasitemia. Erythrocytes from infected animals were coated with immunoglobulin M during or just before the waves of anemia and parasitological crises. Erythrocytes from normal animals could be sensitized with "precrisis" sera presumably containing antigen and antibody. These data suggest that anemia during the acute phase is due to sensitization of erythrocytes with immunoglobulin M-antigen complexes. The anemia is parti

Non-specific immune responses in CBA/N mice infected with Trypanosoma brucei.

CBA/N mice carry a genetic defect which causes a maturational arrest of normal B lymphocyte development, and results in an inability to mount IgM antibody responses against certain T lymphocyte-independent antigens. These mice were found to survive an experimental infection with Trypanosoma brucei two to three times longer than conventional mice. This enhanced survival does not appear to be related to the level of infection of the host since parasitaemias were similar in all strains tested. In addition, the level of non-specific polyclonal B cell activation as assessed by the enumeration of an

Murine T lymphocyte specificity for African trypanosomes. II. Suppression of the T lymphocyte proliferative response to Trypanosoma brucei by systemic trypanosome infection.

Previously, we described a system allowing the study of murine T cell-dependent proliferative responses to Trypanosoma brucei antigens. It was observed that T. brucei-specific T cells could be demonstrated in the regional lymph nodes of primed mice for only 2 to 3 weeks following priming. The results of the present study indicate that this inability to demonstrate a long-lived memory response is due to an immunosuppressive effect of the resulting T. brucei infection. The exact mechanism of the suppression is not known, and appears to function in the absence of demonstrable suppressor cells. Si

Trypanosoma rhodesiense infection in congenitally athymic (nude) mice.

Athymic nude mice (nu/nu), heterozygous litter mates (nu/+), and thymic reconstituted homozygous animals (nu/nu TxR) were infected with Trypanosoma rhodesiense. A reduced parasitemia and prolonged survival were observed in the nu/nu animals after infection and cure or after immunization with irradiated organisms. These studies indicate that resistance of mice to T. rhodesiense infection is relatively independent of thymic lymphocyte function.
