Two heads are better than one: Single stranded DNA translocation of UvrD-family dimers vs. monomers.

UvrD-family Superfamily 1A helicases are processive ATP-dependent motor proteins that function during DNA replication, recombination, repair, and transcription. UvrD-family monomers translocate along single stranded (ss) DNA with 3'-to-5' directionality but must be activated by dimerization to become helicases in the absence of force or accessory factors. Mycobacterium tuberculosis ( Mtb ) UvrD1 helicase forms dimers via a disulfide bond between native cysteines in the 2B sub-domains of each monomer. E. coli UvrD forms non-covalent dimers using the same 2B domain interface as in Mtb UvrD1. Usi

Development of a Disposable Chemiresistive Biochip for Tear-Based Retinopathy of Prematurity Diagnosis: Early Screening and Risk Prediction.

Retinopathy of prematurity (ROP) is a dreadful eye condition and is considered as leading cause of blindness in children around the globe. Lack of timely and appropriate treatment for this ocular condition leads to abnormal development of retinal blood vessels and progression to the severe stage ROP, resulting in irreversible blindness. Based on our previous studies that identified matrix metalloproteinases (MMPs) to be potential tear biomarkers, herein we are demonstrating that the increased levels of MMP-9 in tears can help in ROP risk prediction. A biochip integrated with vanadium disulfide

Construction of a near-infrared fluorescent probe for cysteine and its application in oxidative stress research.

Cysteine (Cys) is a key endogenous thiol in the body, and its active sulfhydryl group plays a core role in physiological processes such as glutathione synthesis, antioxidation, protein disulfide bond formation, and metal ion homeostasis. Clinical studies have confirmed that abnormal intracellular Cys levels and oxidative stress damage are mutually causal and can exacerbate free radical reactions and induce neurodegeneration, liver injury, and cardiovascular diseases. Therefore, the development of high-spatiotemporal resolution Cys dynamic monitoring technology is of great significance. Fluores

SARS-CoV-2 Delta variant-specific ORF3a mutations destabilize lysosomal homeostasis to trigger lysosomal damage-mediated cell death.

The variants of concern (VOCs) of SARS-CoV-2 emerged independently and became dominant globally at different times. Despite the emergence of multiple VOCs, the Delta variant of SARS-CoV-2 showed heightened pathogenicity and unprecedented mortality. However, the Delta variant specific mechanisms underlying its increased pathogenicity are unclear. Here, we show that variations in SARS-CoV-2 ORF3a correlate with the pathogenic potential of VOCs and drive Delta variant-specific lysosomal damage that activates inflammatory cell death. ORF3a from the Delta variant shows unique mutational patterns di

Exploiting ER proteostasis in malaria: protein disulphide isomerases as selective antimalarial targets.

INTRODUCTION: The emergence of partial resistance to artemisinin-based therapies has intensified the search for antimalarial targets beyond classical kinases and proteases. Protein disulfide isomerases (PDIs) have emerged as attractive candidates due to their roles in endoplasmic reticulum (ER) oxidative folding, redox homeostasis, and survival under proteotoxic stress. Several Plasmodium falciparum PDI family members are essential during asexual blood stages and contribute to parasite transmission. AREAS COVERED: We summarize PfPDI architecture, catalytic and holdase functions, and their int

The calcium-sensing receptor in chronic kidney disease-mineral and bone disorder: Structural biology, biased signaling, osteoimmune crosstalk, and therapeutic implications.

The calcium-sensing receptor (CaSR) is a Family C G protein-coupled receptor (GPCR) that functions as a principal sensor of extracellular calcium ions ([Ca2+]o) and a broad-spectrum integrator of polyanionic, amino acid, and ionic stimuli. Its structural architecture - encompassing a bilobed Venus flytrap domain (VFT), cysteine-rich domain (CRD), heptahelical transmembrane domain (TMD), and an intrinsically disordered intracellular C-terminal tail - enables a remarkable allosteric landscape that goes far beyond simple on/off switching. CaSR operates as an obligate homodimer stabilized by inter

Preparation and evaluation of Escherichia coli-derived glycoprotein E for development of a subunit vaccine against varicella-zoster virus.

Glycoprotein E (gE) of varicella-zoster virus (VZV) is a validated target for subunit vaccine development. However, gE produced in Chinese hamster ovary (CHO) cells is associated with high manufacturing costs and glycosylation-related heterogeneity. Here, the ectodomain of gE (aa 25-539) was expressed in Escherichia coli, yielding a monomeric, non-glycosylated gE ectodomain (n-gE) that was efficiently prepared and comprehensively characterized. To recover properly folded protein from inclusion bodies, a two-stage refolding strategy was established, in which disulfide bond formation was promote

Riboflavin Salvage Supports Glycolysis in Borrelia burgdorferi Through Flavin-Dependent NAD+ Regeneration.

Borrelia burgdorferi (Bb), the etiological agent of Lyme disease, lacks the genes for de novo riboflavin biosynthesis and depends on host-derived riboflavin to generate flavin cofactors FMN and FAD. Riboflavin salvage by Bb has demonstrated importance to infection and metabolic homeostasis, though the critical roles of flavin cofactors to central metabolism are not well defined. Here, we demonstrate that riboflavin availability directly controls glycolytic flux through regulation of intracellular redox balance. Disruption of riboflavin uptake by deletion of gene bb0318 encoding the putative AT

Botulinum neurotoxins: A distinct subclass of SNARE-Cleaving endopeptidases.

Botulinum neurotoxins (BoNTs) are bacterial protein toxins that cause the severe and potentially fatal neuro-paralytic disease botulism in humans and vertebrate animals. Together with tetanus toxin (TeNT), BoNTs comprise the large family of clostridial neurotoxins (CNTs). Structurally, CNTs share conserved features: a 50 kDa zinc-dependent light chain endopeptidase with an HExxH motif linked via a disulfide bond to a 100 kDa heavy chain with distinct domains for translocation and receptor binding that mediate neuronal specificity. During the last decade, genomic analyses have uncovered bont-li

An Inhalable Hybrid Nitric Oxide Nanogenerator for Lung Adenocarcinoma Treatment via Bioorthogonal-Activated Gas-Immunotherapy.

Nitric oxide (NO) holds promise for circumventing resistance to platinum-based chemotherapy by reprogramming the tumor microenvironment (TME) in lung adenocarcinoma (LUAD). However, their further application is greatly hindered by uncontrolled release kinetics and off-target toxicity. Herein, an inhalable NO nanogenerator (10m@FOMs-Cu) that reprograms the TME via specific NO release is developed by encapsulating a biorthogonal-activated NO-cisplatin prodrug into TME-responsive hybrid micelles, allowing for sustained and effective LUAD treatment. The prodrug integrates a Pt-based catalyst with

Structure and immunogenicity of an engineered soluble prefusion-stabilized EBV gB antigen.

Epstein-Barr virus (EBV), the causative agent of mononucleosis, is linked to over 140,000 annual cancer-related deaths globally and increases the risk of multiple sclerosis by up to 32-fold. As a herpesvirus, EBV establishes lifelong infection, and over 90% of U.S. adults are EBV-seropositive. Despite its significant disease burden, no approved EBV vaccines or therapeutics exist. Among EBV envelope glycoproteins, the fusion protein (gB) is strictly required for epithelial and B cell infection. Here, using a combination of AlphaFold-guided modeling, rational design, and ThermoMPNN-informed opti

Camptothecin-loaded Surface-modified Nanostructured Lipid Carriers Consisting of L-Cysteine- and L-Cystine-derived Lipids: Physicochemical Characterization and In Vitro Evaluation.

Camptothecin (CPT) is a potent chemotherapeutic agent used in the treatment of colorectal, breast, ovarian, colon, and stomach cancers. However, its clinical application has been significantly restricted due to several drawbacks, including poor water solubility, nonselective toxicity, drug resistance, and inherent instability. To address some of these challenges and to enhance the stability, solubility, and biological efficacy of CPT, in this work, we have attempted to explore nanoparticulate delivery strategies using nanostructured lipid carriers (NLCs) for the targeted delivery of CPT. We ha

Integrated Genomic Analysis Identifies Clinically Relevant Molecular Subtypes and Disulfidptosis-Related Prognostic Signature of Gastric Cancer.

Disulfidptosis is a newly defined disulfide stress-induced cell death. However, there are gaps in the prognostic role of disulfidptosis-related genes (DRGs) and their correlation with the tumor microenvironment (TME) in gastric cancer (GC). In here, we systematically investigated DRG changes at genomic and transcriptional levels, prognostic value, and their expression patterns in GC. Fifteen DRGs were used to identify the different subtypes, and the differences in prognosis and immune infiltration among the subtypes were examined. We identified 3 distinct molecular subtypes and observed profou

Distribution of epithelial endoplasmic reticulum stress-related proteins in adult and pediatric Crohn's disease: Association with inflammation and fibrosis.

BACKGROUND/AIMS: Intestinal strictures in Crohn's disease (CD), driven by fibrosis remain challenging to treat. Current treatments focus on inflammation, but are less effective against fibrosis. Endoplasmic Reticulum Stress-Related Proteins, including Protein disulfide isomerases (PDIs), may contribute to fibrosis; their roles in CD remain unclear. This study investigated the distribution of AGR2, BiP, PDIA6, ERP44 in intestinal epithelium and their association with fibrosis and inflammation in pediatric and adult CD. METHODS: We retrospectively analyzed 224 patients (2009-2023). CD patients

Berberine-Functionalized Bismuth-Doped Carbon Dots in a Pathogen-Responsive Hydrogel System: A Multifaceted Approach to Combating Periodontal Diseases.

Periodontal disease, a global health burden linked to dysbiotic oral polymicrobial communities and disrupted immune-inflammatory responses, is critically mediated byPorphyromonas gingivalis(Pg)─the keystone pathogen that sabotages host immunity, triggers tissue inflammation and destruction, and disrupts microbiota balance. Effective therapies should combine antimicrobial action, immune modulation, virulence suppression, and microbiome restoration. Bismuth ions and berberine, which exhibit antimicrobial and epithelial barrier-protecting effects, show potential effectiveness in treating periodon

Structural insights into the role of reduced cysteine residues in SOD1 amyloid filament formation.

The formation of superoxide dismutase 1 (SOD1) filaments has been implicated in amyotrophic lateral sclerosis (ALS). Although the disulfide bond formed between Cys57 and Cys146 in the active state has been well studied, the role of the reduced cysteine residues, Cys6 and Cys111, in SOD1 filament formation remains unclear. In this study, we investigated the role of reduced cysteine residues by determining and comparing cryoelectron microscopy (cryo-EM) structures of wild-type (WT) and C6A/C111A SOD1 filaments under thiol-based reducing and metal-depriving conditions, starting with protein sampl

Amino acid chemistry and post-translational modifications underlying marine adhesive proteins: Biochemical insights for designing underwater adhesives.

Underwater adhesion remains a complex challenge due to the interference of water with traditional bonding mechanisms. Marine organisms, such as mussels, sandcastle worms, and barnacles, have evolved specialized adhesive proteins that overcome these limitations through functional amino acid motifs. This review highlights how post-translationally derived amino acid motifs, such as 3,4-dihydroxylphenylalanine (DOPA) and phosphoserine, drive interfacial adhesion and the curing process through interfacial binding, metal coordination, and crosslinking. Alongside these, canonical amino acids, such as

Ultrasound-responsive nanobubble-mediated sonodynamic therapy sensitizes disulfidptosis in the treatment of liver hepatocellular carcinoma.

Disulfidptosis, a newly identified regulated cell death, is linked to tumor progression, particularly in cancers with elevated SLC7A11 expression. This study investigates SLC7A11 expression in liver hepatocellular carcinoma (LIHC) and evaluates the therapeutic potential of ICG@C3F8-KL nanobubbles (NBs) combined with sonodynamic therapy (SDT) for inducing disulfidptosis. Bioinformatics analysis of TCGA datasets revealed upregulation of SLC7A11 in LIHC tissues. The synthesized ICG@C3F8-KL NBs exhibited a mean diameter of 156.46 nm and stable properties, with high encapsulation efficiencies of 51

Cysteine variants in PMM2 lead to protein instability and higher sensitivity to oxidative stress in PMM2-CDG.

PMM2-congenital disorder of glycosylation (PMM2-CDG) is caused by genetic defects in PMM2, the gene encoding phosphomannomutase 2. Effective therapies for this disorder remain elusive. Recent studies emphasize cysteine's vulnerability to oxidative modifications that can instigate disease by facilitating inter-protein disulfide bonding, reducing protein mobility, highlighting its potential as a target for therapeutic intervention. Specifically, five cysteine-related pathogenic mutants have been identified in PMM2-CDG, namely Phe11Cys (F11C), Tyr64Cys (Y64C), Tyr76Cys (Y76C), Tyr106Cys (Y106C) a

TIGAR plays neuroprotective roles in MPP+/MPTP-induced Parkinson's disease by alleviating ferroptosis.

Parkinson's disease (PD) is a common neurodegenerative disorder worldwide, characterized by the loss of dopaminergic (DA) neurons in the substantia nigra and is associated with iron dyshomeostasis. Ferroptosis, a form of programmed cell death, involves iron-dependent lipid peroxidation and serves as a significant regulatory mechanism in PD. This study identified Tp53-induced glycolysis and apoptosis regulator (TIGAR) as a potential regulator of ferroptosis resistance in PD development. In this study, we demonstrated that in HT22 cells, 1-methyl-4-phenylpyridinium (MPP+) increased lipid peroxid
